Project description:FTO, an N6-methyladenosine demethylase, has emerged as a promising target for the treatment of specific acute myeloid leukemia (AML) subtypes. Here, we investigate the antiproliferative effects of the FTO inhibitor FB23-2 in leukemia. We demonstrate that FB23-2 potently inhibits proliferation across both AML and CML cell lines, irrespective of their responsiveness to FTO depletion. Interestingly, FB23-2 induces cell cycle arrest without a concurrent increase in m6A levels, suggesting an alternative mechanism of action.

FB23-2 exerts therapeutic effects against T-ALL through the FTO/m6A/MYC axis.

Project description:T-cell acute lymphoblastic leukemia (T-ALL) is a pediatric hematological malignancy that presents significant challenges. RNA N6-methyladenosine (m6A) methylation is crucial in tumorigenesis, and the m6A demethylase FTO has emerging importance in cancers; however, its specific role in T-ALL pathogenesis remains incompletely understood. In vitro studies employed T-ALL cell lines treated with the FTO inhibitor FB23-2. Our findings indicate that FB23-2 may exert therapeutic effects against T-ALL primarily through the FTO/m6A/MYC axis.

Targeting FTO suppresses hepatocellular carcinoma by inhibiting ERBB3 and TUBB4A expression [MeRIP]

Project description:Fat mass and obesity-associated protein (FTO) is an N6-methyladenosine (m6A) demethylase and plays a critical oncogenic role in various cancers. Here we show that FTO is an effective target in hepatocellular carcinoma (HCC). FTO is highly expressed in clinical patients of HCC and presents as a poor prognostic factor. Genetic depletion of FTO dramatically attenuated HCC progression in vivo. Pharmacological inhibition of FTO by FB23/FB23-2 remarkably suppressed the proliferation and migration of HCC cell lines in vitro, as well as inhibiting the HCC tumorigenicity in xeno-transplanted mice. Mechanistically, FB23-2 suppressed the expression of Erb-b2 receptor tyrosine kinase 3 (ERBB3) and human tubulin beta class Iva (TUBB4A) by increasing the m6A level in these mRNA transcripts. The decreased ERBB3 resulted in inhibited Akt-mTOR signaling that subsequently impaired the proliferation and survival of HCC cells. Moreover, FB23-2 abolished the organization of the tubulin cytoskeleton, while enforced expression of TUBB4A rescued the migration of HCC cells. Collectively, our study demonstrated that FTO plays a critical role in HCC by maintaining the proliferation and migration of cells, and highlights the broad potential of FTO inhibitors for targeting HCC.

The effects of FTO inhibitors on MeRIP-seq profiles in small cell lung cancer cell line

Project description:We reveal that pharmacological inhibition of fat mass and obesity-associated protein (FTO), a key m6A demethylase, exerts anti-tumor effects on the human small cell lung cancer (SCLC) cell line H69, with a close correlation to the remodeling of m6A modification landscape. We observed that FTO inhibitor treatment significantly elevates global m6A levels in H69 cells, which is attributed to the suppressed demethylase activity of FTO. Meanwhile, the FTO inhibitor FB23-2 can inhibit the proliferation of SCLC.

Impact of gut microbiota on m6A/m epitranscriptomic mRNA modification in different host tissues

Project description:In this study we performed MeRIP-Seq to study N6-methyl adenosine (m6A) and and N6,2′ -O-dimethyladenosine (m6Am) modification of mRNA. We investigated the effect of the microbiota on the transcriptome and epitranscriptomic modifications in murine liver and cecum. We compared m6A/m modification profiles in cecum of conventionally raised (CONV) and germ-free (GF) mice. We additionally included GF mice colonised with the flora of CONV mice for four weeks (ex-GF), for which show that they exhibit similar patterns of the most abundant genera of gut bacteria as CONV mice. We added mice treated with several antibiotics to deplete the gut flora (abx)and vancomycin treated mice in which the genera Akkermansia, Escherichia/Shigella and Lactobacillus were enriched. Furthermore, we included GF mice colonised with the commensal bacterium Akkermansia muciniphila (Am), Lactobacillus plantarum (Lp) and Escherichia coli Nissle (Ec) and analysed their m6A/m modification profiles. In addition, we analysed changes in m6A/m- modified liver RNA for CONV, GF, and Am, Lp and Ec mice.
