Project description:Our study demonstrates that the short-chain fatty acid, butyrate, greatly enhances the in utero development of ENCSCs and adult enteric neurogenesis, which was partially mediated through the receptors MCT1 and Ras signaling pathway. In conclusion, butyrate treatment may represent a potential therapy for various gastrointestinal motility disorders.

Project description:Enteric glia cells (EGCs) are highly plastic and has been shown to give rise to neurons in adult gut upon injury to the enteric nervous system (ENS). To better characterise the dynamic molecular changes associated with EGC activation and its subsequent neurogenic differentation trajectory, we performed bulk RNAseq experiments from FACS sorted lineage-labelled cells (Sox10::CreERT2;R26-tdTomato) from adult small intestines (DIV0) and compared their transcriptomic profiles to cultured FACS sorted tdTomato+ cells from days in vitro (DIV) 4, 11 and 20, which were subjected to a protocol to support neurogenesis in culture.

Inflammation Rewires the Enteric Nervous System Through Neurogenic Monocyte Recruitment

Project description:The proper organization of the enteric nervous system (ENS) is critical for normal gastrointestinal (GI) physiology. Inflammatory bowel disease (IBD) dysregulates GI physiology including bowel movements (motility), but in many IBD patients, GI motility disorders persist in remission through a poorly understood pathological process. Here we uncover that post-inflammatory GI dysmotility (PI-GID) stems from structural ENS remodeling driven by a combination of neuronal loss and neurogenesis. Enteric neurons respond to mucosal inflammation by upregulating CCL2 expression and facilitating the recruitment of CCR2+ monocytes into the neural myenteric plexus of the intestinal muscle followed by the expansion of monocyte-derived macrophages, their migration into the myenteric ganglia and phagocytosis of neurons. However, excessive recruitment of monocytes promotes disproportionate ENS remodeling and PI-GID. Expansion of immune cells in the tissue also promotes tissue hypoxia. We find that enteric neurons are hypoxic upon colitis; hypoxia-induced signaling via HIF1alpha induces an adaptation program in enteric neurons to suppress CCL2 expression and limit monocyte recruitment. We demonstrate that reinforcing HIF1alpha signaling in enteric neurons prevents PI-GID by reducing colitis-associated monocyte recruitment in the myenteric plexus and mitigating ENS remodeling. In summary, our findings unveil PI-GID pathogenesis and identify a regulatory axis for its prevention.

A branching model of lineage differentiation underpinning the neurogenic potential of enteric glia (Single-cell expression analysis of small intestine enteric nervous system developmental data)

Project description:Glial cells have been proposed as an endogenous source of progenitors for the treatment of neural deficits. However, the cellular and molecular mechanisms underpinning the neurogenic potential of certain populations of adult glial cells, are not known. Using single cell transcriptomic profiling, we show here that enteric glial cells represent a cell state attained by autonomic neural crest cells as they transition during development along a linear default differentiation trajectory that allows them to retain neurogenic potential while acquiring a gene expression profile associated with their role in neuronal support and immunomodulation. Key neurogenic loci in early enteric nervous system progenitors remain in open chromatin configuration in mature enteric glia, thus facilitating neuronal differentiation under appropriate conditions. Molecular profiling and gene targeting of enteric glial cells in a novel cell culture system of enteric neurogenesis and a gut injury model, demonstrated that neuronal differentiation of glia is driven by transcriptional programs employed in vivo by early progenitors. Our work provides mechanistic insight into the dynamic regulatory landscape underpinning the development of intestinal neural circuits and generates a platform for advancing glial cells as therapeutic agents for the treatment of neural deficits.
