Project description:Background and aimsThe ubiquitous feather mosses Pleurozium schreberi and Hylocomium splendens form a thick, continuous boundary layer between the soil and the atmosphere, and play important roles in hydrology and nutrient cycling in tundra and boreal ecosystems. The water fluxes among these mosses and environmental factors controlling them are poorly understood. The aim of this study was to investigate whether feather mosses are capable of internal transport and to provide a better understanding of species-specific morphological traits underlying this function. The impacts of environmental conditions on their internal transport rates were also investigated.MethodsCells involved in water and food conduction in P. schreberi and H. splendens were identified by transmission electron microscopy. Symplasmic and apoplasmic fluorescent tracers were applied to the moss stems to determine the routes of internal short- and long-distance transport and the impact of air humidity on the transport rates.Key resultsSymplasmic transport over short distances occurs via food-conducting cells in both mosses. Pleurozium schreberi is also capable of apoplasmic internal long-distance transport via a central strand of hydroids. These are absent in H. splendens. Reduced air humidity significantly increased the internal transport of both species, and the increase was significantly faster for P. schreberi than for H. splendens.ConclusionsPleurozium schreberi and Hylocomium splendens are capable of internal transport but the pathway and conductivity differ due to differences in stem anatomy. These results help explain their varying desiccation tolerance and possibly their differing physiology and autecology and, ultimately, their impact on ecosystem functioning.

Project description:The global significance of marine non-cyanobacterial diazotrophs, notably heterotrophic bacterial diazotrophs (HBDs), has become increasingly clear. Understanding N2 fixation rates for these largely uncultured organisms poses a challenge due to uncertain growth requirements and complex nitrogenase regulation. We identified Candidatus Thalassolituus haligoni as an Oceanospirillales member, closely related to other significant γ-proteobacterial HBDs. Pangenome analysis reinforces this classification, indicating the isolate belongs to the same species as the uncultured metagenome-assembled genome Arc-Gamma-03. Analysis of the nifH gene in amplicon sequencing libraries reveals the extensive distribution of Cand. T. haligoni across the Pacific, Atlantic and Arctic Oceans. Through combined proteomic analysis and N2 fixation rate measurements, we confirmed the isolate’s capacity for nitrate independent N2 fixation, although a clear understanding of nitrogenase regulation remains unclear. Overall, our study highlights the significance of Cand. T. haligoni as the first globally distributed, cultured model species within the understudied group of Oceanospirillales, and γ-HBDs in general.

Project description:Legumes can utilize atmospheric nitrogen via symbiotic nitrogen fixation, but this process is inhibited by high soil inorganic nitrogen. So far, how high nitrogen inhibits N2 fixation in mature nodules is still poorly understood. Here we construct a co-expression network in soybean nodule and find that a dynamic and reversible transcriptional network underlies the high N inhibition of N2 fixation. Intriguingly, several NAC transcription factors (TFs), designated as Soybean Nitrogen Associated NAPs (SNAPs), are amongst the most connected hub TFs. The nodules of snap1/2/3/4 quadruple mutants show less sensitivity to the high N inhibition of nitrogenase activity and acceleration of senescence. Integrative analysis shows that these SNAP TFs largely influence the high N transcriptional response through direct regulation of a subnetwork of senescence-associated genes and transcriptional regulators. We propose that the SNAP-mediated transcriptional network may trigger nodule senescence in response to high N.

Project description:Although N2 fixation can occur in free-living cyanobacteria, the unicellular endosymbiotic cyanobacterium Candidatus Atelocyanobacterium thalassa (UCYN-A) is considered to be a dominant N2-fixing species in marine ecosystems. Four UCYN-A sublineages are known from partial nitrogenase (nifH) gene sequences. However, few studies have investigated their habitat preferences and regulation by their respective hosts in open-ocean versus coastal environments. Here, we compared UCYN-A transcriptomes from oligotrophic open-ocean versus nutrient-rich coastal waters. UCYN-A1 metabolism was more impacted by habitat changes than UCYN-A2. However, across habitats and sublineages genes for nitrogen fixation and energy production were highly transcribed. Curiously these genes, critical to the symbiosis for the exchange of fixed nitrogen for fixed carbon, maintained the same schedule of diel expression across habitats and UCYN-A sublineages, including UCYN-A3 in the open-ocean transcriptomes. Our results undersore the importance of nitrogen fixation in UCYN-A symbioses across habitats, with consequences for community interaction and global biogeochemical cycles.
