Project description:Fibrosis, neurodegeneration, and cerebral angiomatosis (FINCA) (OMIM 618278) is a rare infantile-onset disease caused by variants in NHL repeat containing 2 (NHLRC2) gene. NHLRC2 is essential for mouse gastrulation (Hiltunen et al., 2022). Here, RNA sequencing was performed on four wild type (WT), four homozygous C57BL/6NCrl-Nhlrc2em1Rthl (FINCA disease causing variant) (Hiltunen et al., 2020), and four homozygous C57BL/6NCrl-Nhlrc2tm1a(KOMP)Wtsi (Nhlrc2 knock out) mouse embryonic stem cell lines each established from individual mouse embryos by the 2i method (Nichols et al., 2009). FINCA variant mESCs express the full length endogenous Nhlrc2 mRNA modified to containing a c.442G > T variant leading to p.Asp148Tyr as well as a silent c.408C>A for genotyping purposes (Hiltunen et al., 2020). Project description:Vibrio species are recognized for their role in food- and water-borne diseases in humans, fish, and aquatic invertebrates. We screened bacterial strains isolated from raw food shrimp for those that are bactericidal to Vibrio strains. Here we identify and characterize Aeromonas dhakensis strain A603 which shows robust bactericidal activity specifically towards Vibrio and related taxa but less potency toward other Gram-negative species. Using the A603 genome and genetic analysis, we show that two antibacterial mechanisms account for its vibriocidal activity -- a highly potent Type Six Secretion System (T6SS) and biosynthesis of a vibriocidal phenazine-like small molecule, named here as Ad-Phen. Further analysis indicates coregulation between Ad-Phen and a pore-forming T6SS effector TseC, which potentiates V. cholerae to killing by Ad-Phen.

Project description:We profile single cells from patients with colorectum cancer using Chromium 3’ and 5’ single-cell RNA-sequencing. Patients EXT001, EXT009, and EXT012 from the KUL dataset were first analyzed by Lee et al., 2020, and the raw data are available in ArrayExpress under the accession codes E-MTAB-8410 and E-MTAB-8107. Patients EXT018, EXT048, EXT113, and EXT121 from KUL dataset were previously analyzed by Joanito et al., 2022. The raw data of those patients are available in EGA under the accession codes EGAD00001008584 and EGAD00001008585.

Project description:The FIDO (Febrile Infant Diagnostic assessment and Outcome) study aimed to look at different diagnostic approaches to identify infants with a fever at low and high-risk of serious infection. FIDO was a multicentre prospective observational cohort study that enrolled infants ≤90 days of age presenting with fever to 35 Paediatric Emergency Research in the UK and Ireland (PERUKI) sites. The FIDO study was prospectively registered at ClinicalTrials.gov (NCT05259683; 28 February 2022). Where possible, residual plasma volume was collected and stored from infants for proteomic and biomarker analysis.
