Project description:The widespread presence of microplastics (MPs) in freshwater systems has raised concerns about their potential ecotoxicity on aquatic organisms. In this study, we evaluated the effects of four MPs with different compositions, namely polyethylene (PE), polystyrene (PS), polyvinyl chloride (PVC) and polypropylene (PP), on freshwater microalgae Chlamydomonas reinhardtii. PS and PVC MPs caused greater growth inhibition and stronger oxidative stress response than PP and PE MPs. Proteomics analysis was employed to explore the mechanical understanding of the composition-dependent toxicity of MPs on microalgae. Proteins involved in photosynthesis processes were identified as contributors to the diverse responses of microalgae to differently composed MPs. Photosynthesis activity of algae, including pigment content and photoprotective response, was determined to reflect the distinct effect of the four MPs. The indicated down-regulated expression of photosynthetic proteins by proteomics analysis was further confirmed using western blot, with PVC and PS showing greater impacts on their expression reduction. Our findings not only show the composition-dependent effect of MPs on microalgae but also provide important insights into the molecular mechanism of MPs’ toxicity on natural phytoplankton species.

Project description:Microplastics represent a growing environmental concern for the oceans due to their potential capability to adsorb different classes of pollutants, thus representing a still unexplored source of exposure for aquatic organisms. In this study polystyrene (PS) microplastics were characterized for their capability to adsorb pyrene (PYR) as model compound for polycyclic aromatic hydrocarbons, and transfer this chemical to filter feeding mussels Mytilus galloprovincialis. Gene expression analyses of Mytilus galloprovincialis exposed to polystyrene (PS) microplastics and to polystyrene contaminated with pyrene (PS-PYR) have been performed trough a DNA microarray platform.

Project description:<p>The phycosphere is an important ecological niche for bacteria and antibiotic resistance genes (ARGs). However, whether and how the interaction between microalgae and bacteria changed, and its further effect on the transmission of ARGs under pollutant stress remains enigmatic. Here, Auxenochlorella pyrenoidosa was co-cultured with bacteria screened from lake water to explore the algal-bacteria interaction and ARGs transmission in the presence of florfenicol (FF) and polylactic acid microplastics (PLA MPs). Our study demonstrated that the growth and metabolism of A. pyrenoidosa were promoted under FF treatment or co-treatment with PLA MPs, validated by phenotypic, transcriptome, and metabolome analyses. In contrast, the abundance of phycospheric bacteria was decreased as a result of niche competition. Nonetheless, the transmission of ARGs in the phycosphere was promoted due to the enrichment of antibiotic-resistant bacteria, especially Pseudomonas, rather than horizontal gene transfer. Algal-bacteria co-culture experiment further suggested that vitamin B6 secreted by Pseudomonas sp. likely contributes to underpinning A. pyrenoidosa survival under FF and PLA MPs stress. These findings underscore the dynamic interplay and co-evolution between algae and bacteria under pollutant exposure, and reveal a potential mechanism of vitamin B6-mediated mutualism. This study provides new insights into the assembly of phycospheric bacterial communities and the adaptive strategies of microalgae in contaminated aquatic environments.</p>

Project description:Biodegradable plastics, such as polylactic acid (PLA), are widely considered as sustainable alternatives to fossil fuel-derived polymers; however, their degradation generates polylactic acid microplastics (PLA-MPs), which raises ecological concerns in freshwater ecosystems. In this study, we investigated the responses of Microcystis aeruginosa to PLA-MPs (1, 10, and 50 mg/L) across a 45-day co-culture experiment. We found the PLA-MPs initially inhibited algal growth (days 18, 27) through oxidative stress, as indicated by elevated MDA content (1.94 nmol/mg protein at 50 mg/L, 1.57 times higher than the control) and SOD activity (1.30 U/mg Protein, 2.45 times higher than the control). However, during stabilization (days 36 and 45), PLA-MPs promoted growth in a dose-dependent manner, with Chl-a content in the 1, 10, and 50 mg/L groups reaching 1.12, 1.36, and 1.44 times that of the control, respectively. Concurrent shifts in inorganic carbon speciation, EPS secretion, and cellular aggregation highlighted dynamic adaptation strategies. Finally, based on the proteomic analysis, it was found that PLA-MPs promoted the growth of M. aeruginosa by improving photosynthetic efficiency, enhancing the expression of the TCA cycle and oxidative phosphorylation, and promoting cell division and proliferation. Our results confirm that PLA-MPs have a long-term effect on the growth of M. aeruginosa, providing fundamental theoretical guidance for investigating the ecological and environmental impacts of PLA-MPs in aquatic ecosystems.
