Project description:Alzheimer's disease (AD) is a neurodegenerative disorder characterized by cognitive decline and memory loss. It has been suggested that alterations in energy metabolism, specifically an increase in resting energy expenditure (REE), may be a key factor in the development and progression of AD. Regular physical activity may improve cognitive function in patients with AD, and although the mechanisms are unclear, organ-to-organ communication may play a role. We hypothesize that changes to extracellular vesicles (EVs) in response to regular physical activity might be involved in organ-to-organ communication mediating the improvement to cognitive function caused by regular physical activity in patients with AD. In this experiment, we profiled the microRNA composition of extracellular vesicles (EVs) purified from the blood of sedentary (sedEV) and/or acutely exercised (exEV) mice as part of understanding this hypothesis.

Project description:Inguinal white adipose tissue (iWAT) is essential for the beneficial effects of exercise training on metabolic health. The underlying mechanisms for these effects are not fully understood and here, we test the hypothesis that exercise training results in a more favorable iWAT structural phenotype. Using biochemical, imaging, and multi-omics analyses we find that 11-days of wheel running in male mice causes profound iWAT remodeling including decreased extracellular matrix (ECM) deposition, increased vascularization and innervation. We identify adipose stem cells as one of the main contributors to training-induced ECM remodeling, show that the PRDM16 transcriptional complex is necessary for iWAT remodeling and beiging, and discover neuronal growth regulator 1 (NEGR1) as a link between PRDM16 and neuritogenesis. Moreover, we find that training causes a shift from hypertrophic to insulin-sensitive adipocyte subpopulations. Exercise training leads to remarkable adaptations to iWAT structure and cell-type composition that can confer beneficial changes in tissue metabolism.

Project description:Timed physical activity might potentiate the health benefits of training. The underlying signaling events triggered by exercise at different times of the day are, however, poorly understood. Here, we found that time-dependent variations in maximal treadmill exercise capacity of naïve mice were associated with energy stores, mostly hepatic glycogen levels. Importantly, running at different times of the day resulted in a vastly different activation of signaling pathways, e.g., related to stress response, vesicular trafficking, repair, and regeneration. Second, voluntary wheel running at the opposite phase of the dark, feeding period surprisingly revealed minimal Zeitgeber (i.e., synchronizing) activity of training. This integrated study provides important insights into the circadian regulation of endurance performance and the control of the circadian clock by exercise. In future studies, these results could contribute to better understand circadian aspects of training design in athletes and the application of chrono-exercise-based interventions in patients.

Project description:Changes and plasticity in both gene expression and protein signaling in skeletal muscle is considered to be a major cause of metabolic syndrome, while it has been shown that mild exercise training at lactate threshold (LT) intensity is a safe and effective for prevention of metabolic syndrome. To elucidate the molecular mechanisms related to the beneficial effects of LT training for 60 min/day for 5 days/wk for 12 wk, we performed serial analysis of gene expression (SAGE) to examine global mRNA expression in human skeletal muscle. Approximately 57000 SAGE tags were analyzed for before training, as well as 5 days, 6 and 12 wk after the training. The LT training has coordinately induced many genes involved in mitochondrial energy metabolism, fat oxidation, glycolysis and creatine metabolism. Another molecular feature associated with this mild exercise regimen has been an induction of many genes encoding for potent antioxidant enzymes and molecular chaperons. Furthermore, the training modulated the expression levels of 233 novel transcripts. Thus, the current study reveals that LT exercise has favorably altered gene expression in human skeletal muscle to the prevention of metabolic syndrome. Project description:Colon cancer is the second most cancer type in Europe. Its development is highly influenced by life style factors such as nutrition and physical inactivity. Detailed biological mechanisms are so far unclear. The purpose of this study was to investigate the effects of chronic wheel running on gene expression in rat colon mucosa. Therefore six-week-old male Wistar rats (exercise (EX) group, n=20) completed a stress free voluntary running exercise period of 12 weeks. Sedentary rats served as a control (CO, n=9) group. In the colon mucosa, steady-state mRNA expression levels of approximately 10,000 genes were compared between both groups by micro-array analysis (MWG rat 10k array). 8,846 mRNAs were detected above background level. Chronic exercise led to a decreased expression of 47 genes at a threshold-factor of 2.0. 3 genes were found to be up-regulated in the EX group. The identified genes encode proteins involved in signal transduction (n=11), transport (n=8), immune system (n=7), cytoskeleton (n=6), protein targeting (n=6) and metabolism (n=5). Among the genes regulated by chronic exercise, the betaine-homocysteine methyltransferase 2 (BHMT2) seems to be of particular interest. Physical activity may protect against aberrant methylation by repressing the BHMT2 gene and thus contribute to a decreased risk of developing colon cancer. In summary, our experiment presents the first gene expression pattern in rat colon mucosa following chronic wheel running and therefore represents an important step in understanding the molecular mechanisms responsible for the preventive effect of physical activity on the development of colon cancer. Project description:Voluntary exercise reduces the risk of cancer and lowers the risk of disease recurrence. Yet the mechanisms for this protection remain to be elucidated. Here we demonstrate that exercise halves tumor growth through an exercise-dependent mobilization and intratumoral infiltration of NK cells in malignant melanoma. Using voluntary wheel running, we show that exercise prior to and during B16 tumor challenge reduced tumor growth by 67%, and this reduction was associated with increased inflammation and immune cell infiltrates, especially NK cells, in the tumors from exercising mice. Depletion of NK cells blunted the exercise-dependent reduction in tumor growth. Moreover, during exercise, NK cells were engaged through an epinephrine-dependent mobilization to the circulation and redistributed to peripheral tissues through an IL-6 dependent mechanism. Project description:BackgroundIncreased physical exercise improves cognitive function and reduces pathology associated with Alzheimer's disease (AD). However, the mechanisms underlying the beneficial effects of exercise in AD on the level of specific brain cell types remain poorly investigated. The involvement of astrocytes in AD pathology is widely described, but their exact role in exercise-mediated neuroprotection warrant further investigation. Here, we investigated the effect of long-term voluntary physical exercise on the modulation of the astrocyte state.MethodsMale 5xFAD mice and their wild-type littermates had free access to a running wheel from 1.5 to 7 months of age. A battery of behavioral tests was used to assess the effects of voluntary exercise on cognition and learning. Neuronal loss, impairment in neurogenesis, beta-amyloid (Aβ) deposition, and inflammation were evaluated using a variety of histological and biochemical measurements. Sophisticated morphological analyses were performed to delineate the specific involvement of astrocytes in exercise-induced neuroprotection in the 5xFAD mice.ResultsLong-term voluntary physical exercise reversed cognitive impairment in 7-month-old 5xFAD mice without affecting neurogenesis, neuronal loss, Aβ plaque deposition, or microglia activation. Exercise increased glial fibrillary acid protein (GFAP) immunoreactivity and the number of GFAP-positive astrocytes in 5xFAD hippocampi. GFAP-positive astrocytes in hippocampi of the exercised 5xFAD mice displayed increases in the numbers of primary branches and in the soma area. In general, astrocytes distant from Aβ plaques were smaller in size and possessed simplified processes in comparison to plaque-associated GFAP-positive astrocytes. Morphological alterations of GFAP-positive astrocytes occurred concomitantly with increased astrocytic brain-derived neurotrophic factor (BDNF) and restoration of postsynaptic protein PSD-95.ConclusionsVoluntary physical exercise modulates the reactive astrocyte state, which could be linked via astrocytic BDNF and PSD-95 to improved cognition in 5xFAD hippocampi. The molecular pathways involved in this modulation could potentially be targeted for benefit against AD.
