Project description:Human pathogens use cell-cell communication systems, quorum sensing, to regulate virulence factors. Our recent work aiming at deciphering the quorum sensing regulon of Streptococcus mutans, a caries pathogen, revealed a locus encoding an atypical type II restriction-modification (R-M) system, DpnII, comprising two adenine methyltransferases. A recent survey of bacterial genomes demonstrated the widespread occurrence of methylated DNA, and the importance of DNA methylation was highlighted by the diverse processes in which they function. The present study aimed at deciphering the role of DNA methylation during S. mutans biofilm formation. A mutant strain, deficient in the DpnII R-M locus composed of the SMU.504 and SMU.505 genes encoding the two adenine methyltransferases and the SMU.506 gene encoding the DpnII restriction enzyme, was constructed in S. mutans wild-type (WT) strain. Transcriptome analysis was then performed to evaluate the differential biofilm gene expression between the WT cells and the DpnII R-M deficient cells.

Lysine lactylation regulates biofilm formation of streptococcus mutans

Project description:Lactate produced in glycolysis has recently been discovered to modify lysine residues on eukaryotic proteins, a post-translational modification (PTM) called lysine lactylation (Kla). This modification not only participates in regulating gene expression through lactylation of histones, but also impacts the function of non-histone proteins. However, lactylation in prokaryotes has not been reported. Here, we report the identification of 1869 lysine lactylation sites in 465 proteins in the cariogenic organism Streptococcus mutans using a newly developed 4-dimensional label-free quantitative proteomic approach, representing the first Kla dataset in prokaryotes. Combining quantitative analysis, we report that the modification levels of 282 sites from 124 proteins increased by over 1.5-fold, and 80 sites from 52 proteins decreased by over 0.67-fold in the biofilm growth state. These results provide a systematic perspective of the distribution and function of Kla and improve our understanding of the role of lactate in the metabolic regulation of prokaryotes.

Project description:In this experiment we collected small molecule data that represent excreted molecules by Streptococcus mutans growing as a biofilm. The S. mutans biofilms were established and incubated in anaerobic conditions. Samples were collected before and after a drastic pH drop due to glucose amendments. Control samples are included in this folder that represent molecules that were extracted from sterilized growth media only. These peaks should be subtracted from the biofilm samples prior to analyses.

Replicate samples for MS/MS-small molecule and transcription analyses were collected at pH 7 and at pH 3.5 (20 hrs after glucose spiking). Only secreted molecules were analyzed with MS/MS (no cell lysis protocol was employed).

Project description:Streptococcus mutans was grown for 48 h in a biofilm in the absence (single species) and in the presence (dual species) of Veillonella parvula. In addition V. parvula single species 48 h biofilms were grown, to be used as a control. RNA was harvested from all types of biofilms and the transcript levels of the two types of biofilms containing S. mutans were compared with the use of S. mutans microarrays. V. parvula RNA was hybridized to S. mutans microarrays as a control for possible cross-hybridisation.
