Project description:This study set out to evaluate the hypothesis that PFAS exposure alters EV regulation via upstream molecular mediators of EV-based alterations through cellular transcriptomics assessment in a concentration-response manner. To evaluate this hypothesis, HepG2 liver cells were treated in concentration-response with individual PFOS, PFOA, or PFHxA, in addition to an equimolar PFAS mixture. Global gene expression was measured. These analyses set out to evaluate concentration-dependent changes in mRNA signatures within HepG2 immortalized liver cells exposed to individual PFOS, PFOA, PFHxA or an equimolar mixture containing all three PFAS.

Project description:Per- and polyfluoroalkyl substances (PFAS) are pervasive environmental pollutants with diverse toxic effects, including endocrine disruption and hepatotoxicity. As a key class of immune cells, macrophages are critical target for endocrine disrupting chemicals (EDCs); however, their role in PFAS-induced toxicity and the underlying mechanisms remain poorly understood. In this study, we developed a high-content cell model by utilizing the activation and differentiation of human THP-1 monocytes into macrophages, enabling rapid quantitative screening of various PFAS chemicals. Our results identified that PFOA, PFUnDA, and HFPODA suppressed macrophage alternative activation to varying extents by disrupting the PPAR signaling pathway. In vivo, oral exposure to PFOA and PFUnDA in wild-type C57BL/6J mice significantly impaired macrophage alternative activation in the liver, resulting in hepatocyte hypertrophy, liver dysfunction, and systemic lipid metabolism disorders. Furthermore, macrophage-specific PPARγ knockout exacerbated PFAS-induced hepatotoxicity. These findings highlight the immunometabolic regulatory role of macrophage activation in PFAS-induced liver toxicity and provide novel insights into the health risks posed by PFAS exposure in humans.

Ex Vivo Human Model for Assessing Immunotoxicity of Per- and Polyfluoroalkyl Substances (PFAS)

Project description:Immunotoxicity has emerged as a key health concern for per- and polyfluoroalkyl substances (PFAS), with numerous animal studies showing reduced T-dependent antibody responses (TDAR) and some human epidemiological studies reporting decreased vaccine antibody titers. In recent years, many PFAS have been heavily regulated in Canada; however, with more than 14,000 PFAS and their well-known environmental persistence, evaluating immunotoxicity for each chemical is critical but impractical. This highlights the need for efficient, human-relevant test systems. Here, we assessed immunomodulatory effects of six PFAS analogues using an ex vivo human peripheral blood mononuclear cell (PBMC) model. PBMCs were pre-exposed to PFAS prior to stimulation to mimic environmentally relevant exposure scenarios, in which PFAS exposure precedes an immune challenge. Two immune stimuli were applied, including lipopolysaccharide (LPS) to trigger innate responses and phytohemagglutinin (PHA) to activate T-cell-mediated adaptive pathways. Multiple endpoints were assessed, including cytokine secretion, T-cell proliferation, and transcriptomic alterations. This study found that several PFAS analogues suppressed cytokine secretion and disrupted T-cell proliferation to varying degrees following PHA activation. Transcriptomics results suggest decreased immunoglobulin gene expression, consistent with an immunosuppressive phenotype, although additional validation is required. Overall, the human PBMC model proved to be a cost-effective, ethical, and sensitive new approach method (NAM) that aligned well with prior in vivo data.

Project description:Per- and polyfluoroalkyl substances (PFAS) are a large group of persistent synthetic chemicals that are ubiquitous environmental contaminants. Mounting evidence demonstrates that PFAS can bioaccumulate and induce a suite of adverse health outcomes, including compromise of male reproduction. Despite this knowledge, there remains no clear consensus as to how PFAS elicit these responses. Accordingly, here we aimed to determine how an environmentally relevant cocktail of PFAS impact the fertility of male mice. After twelve weeks of continuous PFAS exposure, blood plasma was isolated for hormone and PFAS profiling and reproductive tissues and spermatozoa were subjected to a comprehensive battery of histological and functional analyses. These analyses revealed that PFAS exposure significantly reduced the rate of daily sperm production linked with reduced levels of circulating testosterone and dihydrotestosterone. Functional profiling of mature spermatozoa from PFAS exposed males failed to identify any overt changes in sperm viability, motility, DNA integrity, or ability to undergo capacitation and support in vitro fertilization and early embryonic development. PFAS exposed spermatozoa did, however, present with pronounced changes in their small non-coding RNA profile and were linked with significant dysregulation of early embryonic gene expression. These observations afford new mechanistic insight into how PFAS exposure impacts male reproductive health.
