Chronic intermittent hypoxia inhibits the revascularization of the ischemic retina and aggravates neurodegeneration and neovascularization

Project description:Neovascular retinopathies and edema are sign threatening complications of ischemic retinopathies, such as retinopathy of prematurity (ROP) and diabetic retinopathy (DR). Sleep apnea that leads to chronic intermittent hypoxia (CIH) is an independent risk factor for severe disease, but the underlying mechanisms are unknown. Here we show that experimental CIH during the ischemic phase of oxygen induced retinopathy in mice severely reduces beneficial revascularitzation of the ischemic retina, and increases neuronal loss and pathological neovascularization. Mechanistically we demonstrate that CIH reduces both colony stimulation factor 1 (CSF-1) expression and the ischemia-induced increase of retinal microglial cells that promotes the revascularization of the ischemic retina in the absence of CIH. Local CSF1R inhibition during ischemic retinopathy reduced the number of microglial cells, inhibited revascularization, and exacerbated pathological neovascularization, recapitulating several effects of CIH. Our findings provide a novel mechanism by which sleep apnea and CIH aggravate ischemic retinopathies, underscoring the importance of treating apnea in ROP and DR to help prevent sight threatening severe disease.

Gene Expression Profiling In Murine Retina Neovascularization Models

Project description:In order to find out the vital genes during retinal neovascularization (RNV), we set up OIR (oxygen-induced retinopathy; induced with 75%Â±2% oxygen) and wild-type C57BL/6J murine models. We observed the retinal vascular growth process daily both in OIR and wild-type mice through retinal flat-mount, and isolated total retinal RNA at different time points (P8, P9, P12, P13, P30) both in OIR and wild-type mice for gene expression analysis. At least three different retinae were accessed at each time point for observing the retinal vascular growth process. Ten neural retinae from five mice were harvested and pooled into one sample for gene expression analysis. Three biological replicates were used for each time point. Dye-swaps were performed.

RNA-seq of choroid and retina in tree shrews with choroidal neovascularization

Project description:In order elucidate the key signaling pathways in choroidal neovascularization, we induced choroidal angiogenesis by laser photocoagulation in 12 tree shrews and obtained mRNA profiles of their choroids and retinas by high-throughput transcriptome sequencing. Gene ontology(GO) and kyoto encyclopedia of genes and genomes(KEGG) enrichment analysis, hierarchical cluster analysis, weighted gene co-expression network analysis, protein-protein interaction (PPI) network analysis, and reverse transcription quantitative PCR (RT-qPCR) were performed.

Novel Regulators of Retina Neovascularization

Project description:Mechanisms of vessel remodeling are not yet fully understood. The purpose of this study is to identify proteins that regulate vascular remodeling in an ROP mouse model. Mice were treated with fluctuating oxygen levels and retinas sampled at the peak of vessel regression (PN12) or neovascularization (PN17) for comparative SWATH-MS proteomics using liquid chromatography-tandem mass spectrometry (LC-MS/MS). We further developed an endothelial cell culture ROP correlate to validate expression of target neovascular markers. 5191 proteins were identified in the retinas of which 498 showed significant changes in expression in elevated oxygen and 345 after a return to normoxia. 122 proteins were uniquely associated with vessel regression and 69 with neovascularization (FC>1.5; P<0.05). Of the latter set, 56 were detected in human retinal endothelial cells (HRECs) of which 42 showed altered expression in the HREC-ROP correlate and 23 regulated in the same direction as in the neovascular retinas. In functional analysis, HREC-specific proteins control angiogenesis-related processes including extracellular matrix remodeling, cell migration and adhesion, junction proteins and proliferation. RNA interference and transfection overexpression studies confirmed that VASP and ECH1, showing highest expression levels in hypoxic HRECs, promoted endothelial cell proliferation while SNX1 and CD109, showing lowest expression, inhibited their proliferation.

Expression data from mouse aorta with chronic intermittent hypoxia exposure

Project description:Atherosclerosis is the pathological basis of cardiovascular disease. Obstructive sleep apnea aggravates atherosclerosis, and chronic intermittent hypoxia (CIH) as a prominent feature of obstructive sleep apnea plays an important role during the process of atherosclerosis. The mechanisms of CIH in the development of atherosclerosis remain unclear. The microarray was used to investigate differentially expressed mRNAs and long noncoding RNAs (lncRNAs) in aorta from five groups of ApoE-/- mice fed with a high-fat diet and exposed to various conditions: normoxia for 8 weeks, CIH for 8 weeks, normoxia for 12 weeks, CIH for 12 weeks, or CIH for 8 weeks followed by normoxia for 4 weeks.
