Expression profiles of satellite cells and alpha7Sca1 cells in mdxITGAM-DTR mice

Project description:We took advantage of a dystrophic mouse model of transient macrophage-depletion, mdxITGAM-DTR mice, in order to analyze the role of macrophage in skeletal muscle regeneration. We generated the transcriptome of satellite cells (SCs) and alpha7Sca1 cells purified by cell sorting from mdxITGAM-DTR mice. The mice were treated, by intramuscular injection, with PBS, as vehicle, or with Diphtheria toxin (DT) in order to achieve the macrophage depletion form hind-limb muscle We described a shift in identity of muscle stem cells dependent on the crosstalk between macrophages and satellite cells. Indeed macrophage depletion determines an exacerbated dystrophic phenotype associated with adipogenic conversion of SCs and reduction of the SC pool.

Expression data from FITC challenged skin of CLEC4C-DTR mice vs. littermate controls treated with diptheria toxin

Project description:CLEC4C-DTR exhibit exacerbated contact hypersensitivty to the FITC hapten. Here we assess the transcriptional differences in whole skin of FITC challenged CLEC4C-DTR+ mice and DTR- mice treated with diptheria toxin. Overall, CLEC4C-DTR upregualated 439 genes and downregulated 152 genes compared to transgene negative controls, filtering output on 2 fold expression level difference and FDR<0.05.

Transcriptional profile of injured hearts treated with the cholinesterase inhibitor pyridostigmine (PYR) at Day 5

Project description:It has been shown that acetylcholine has anti-inflammatory properties. The aim of this study is to investigate the role of acetylcholine in cardiac inflammation. We used the diphtheria toxin (DT)-cardiomyocyte ablation model (DTR model) (Lavine et al., 2014 - PNAS) to induce cardiac injury (DT, 2 days, 2.5µg/kg, IP). Beginning one week prior to DT injury, mice were treated with vehicle (PBS, phosphate buffered saline) or the cholinesterase inhibitor pyridostigmine (PYR, 3mg/kg, SQ). Hearts were harvested at Day 5 post-DT-injury. Here, we provide the transcriptional profile of wild-type/PBS, wild-type/PYR, DTR/PBS and DTR/PYR hearts.

Project description:Comparison of transcript levels after diphtheria-toxin deletion of the Nav1.8-expressing cells using the cre/loxP system.<br> Pain requires input from nociceptors which are specialised peripheral sensory neurons in dorsal root ganglia (DRG). To explore the nociceptor-specific mRNA profiles, we used microarray analysis to examine the altered repertoire of genes expressed in DRG from mice depleted of Nav1.8-expressing neurons which are mainly nociceptors. To generate the animal, a Nav1.8 knock-in Cre-expressing mouse (Nav1.8-Cre) was used to excise a floxed stop upstream of globally expressed diphtheria toxin A-subunit gene (ROSA26-eGFP-DTA). By crossing heterozygous Cre mice with homozygous toxin-expressing floxed mice, experimental toxin-expressing (DTA) mice, in which DTA expressing nociceptors had been deleted, were generated. 6 DTA mice and 6 litter mate controls were used for microarray analysis. 3 Affymetrix Mouse Genome 430 2.0 Array chips were employed for analysis of the gene transcripts of each conditions.

Effect of Amphiregulin on global gene expression in injured skeletal muscle in the presence or absence of CD4+Foxp3+ regulatory T cells (Tregs).

Project description:Global gene expression analysis of injured skeletal muscle showed that amphiregulin (Areg), a growth factor over-expressed by muscle Treg cells, enhances muscle regeneration both in the presence and in the absence of Tregs. Foxp3-DTR+ and Foxp3-DTR- mice were injured with cardiotoxin in TA muscle at day 0 and treated with diphtheria toxin every other day from day 0 until dissection. Amphiregulin or PBS were administered im on day 0 and ip every other day until dissection. TA muscle was flash-frozen and RNA was extracted using Trizol. RNA from whole tissue samples was amplified, labeled, and hybridized to Affymetrix Mouse Gene 1.0 ST Arrays.
