Project description:The heterogeneity in symptomatology and phenotypic profile attributable to COVID-19 is widely unknown. For the first time, our study provides the unique advantage of obtaining samples from the Middle Eastern population, an underrepresented region in genetic studies, and explore new genotypes in this population that will yield to novel genetic association. Specifically, we studied 646 patients in the United Arab Emirates. We describe strong association signals from genes on chromosomes 2, 3, 5, 11 and 13, which carry genes that are expressed in the lung, have been associated with tumour progression, emphysema, airway obstruction, and surface tension within the lung. Identifying genetic variants associated to COVID-19 susceptibility and severity may uncover novel biological insights into disease pathogenesis and identify mechanistic targets for therapeutic and vaccine development.

Project description:understanding the biology of methicillin resistant staphylococcus aureus (MRSA) is crucialto unlocking insights for new targets in the fight against this pathogen, there is howeverlimited reports of methadological approaches for carrying proteomic and metabolomic profiling in S.aureus. Therefore, we describe the use of a dual-functionality methanol extraction method for the concurrent extraction of protein and metabolites from S.aureus and reporton the comparative analysis of the proteomic and metabolomic profiles of MRSA versus methicillin sensitive S. aureus (MSSA). Using a reference strain from MRSA and MSSA, we first compared the MRSA proteome extracted using the methanol method to the one from the traditionally used urea method. Then using the methanol extraction method, we compared the proteome and metabolome of MRSA versus MSSA. Through this study, we demonstrated the effectivnessof the methanol-based-dual-extraction method, providing simultaneous insights into the proteomic and metabolomic landscapes of S.aureus strains, demonstrating the utility of proteomic and metabolomic profiling for elucidating the biological basis of antimicrobial resistance

Project description:Staphylococcus aureus is a leading cause of hospital-associated infections. In addition, highly virulent strains of methicillin-resistant S. aureus (MRSA) are currently spreading outside health care settings. Survival in the human host is largely defined by the ability of S. aureus to resist mechanisms of innate host defense, of which antimicrobial peptides form a key part especially on epithelia and in neutrophil phagosomes. Here we demonstrate that the antimicrobial-peptide sensing system aps of the standard community-associated MRSA strain MW2 controls resistance to cationic antimicrobial peptides. The core of aps-controlled resistance mechanisms comprised the D-alanylation of teichoic acids (dlt operon), the incorporation of cationic lysyl-phosphatidylglycerol (L-PG) in the bacterial membrane (mprF), and the vraF/vraG putative antimicrobial peptide transporter. Further, the observed increased production of L-PG under the influence of cationic antimicrobial peptides was accompanied by the up-regulation of lysine biosynthesis. In noticeable difference to the aps system of S. epidermidis, only selected antimicrobial peptides strongly induced the aps response. Heterologous complementation with the S. epidermidis apsS gene indicated that this is likely caused by differences in the short extracellular loop of ApsS that interacts with the inducing antimicrobial peptide. Our study shows that the antimicrobial peptide sensor system aps is functional in the important human pathogen S. aureus, significant interspecies differences exist in the induction of the aps gene regulatory response, and aps inducibility is clearly distinguishable from effectiveness towards a given antimicrobial peptide. Project description:Methicillin-resistant Staphylococcus aureus (MRSA) is a major human pathogen in both community and health care settings, which causes a wide range of infections. Its resistance to β-lactam antibiotics and methicillin in particular, greatly complicates treatment options and success rate due to the limited number of antibiotics with activity against MRSA. To further the development of alternative therapeutics, the mechanisms that mediate antibiotic resistance in MRSA need to be fully understood. Cannabinoid compounds including cannabidiol (CBD), tetrahydrocannabinol (THC) and cannabinol (CBN) have shown promise as potential antibiotic adjuvants. In the present study, MRSA cells were subjected to antibiotic stress from methicillin in combination with three cannabinoid compounds, and subsequently analysed using metaproteomics to assess the metabolic response. Subjecting MRSA to methicillin made the cells more viable and increased their energy production, as well as upregulation of penicillin-binding protein 2 (PBP2). The cannabinoids all showed antimicrobial activity against MRSA, and inhibited the energy production of the cells as well as PBP2 when used in combination with methicillin. Furthermore, all three cannabinoid compounds inhibited resistance mechanisms in MRSA, resulting in a decrease in the minimum inhibitory concentration (MIC) of methicillin when used in combination.

Project description:Background: Bacterial small regulatory RNAs (sRNAs) have been implicated in important processes including antimicrobial stress response. However, the full extent of sRNA involvement in antimicrobial response in Staphylococcus aureus, an important pathogen, is incompletely understood. We investigated the transcriptional profiles of a linezolid-resistant, livestock-associated methicillin-resistant S. aureus (LA-MRSA) strain ST398 under conditions of linezolid stress. Methods: Cells in mid-exponential growth were subjected to low (8 µg/ml) and high (16 µg/ml) dose linezolid treatments followed by high throughput RNA sequencing. Read mapping and differential expression analysis were performed followed by detection and interrogation of various sRNA and mRNA transcripts. Results: Twenty-three putative regulatory RNA transcripts were expressed under low- and high-dose exposure conditions. Cis-acting regulatory elements, mainly targeting ribosomal biogenesis constituted the majority of transcriptional response with limited antisense and small RNA expression. Conclusions: This is the first study to investigate linezolid-responsive small RNA transcription in LA-MRSA strain ST398 and the first to query regulatory RNAs on a background of linezolid resistance. It provides preliminary insights and a basis for interrogating small RNAs in other strains in the quest to understand drug-responsive regulatory RNAs and identify potential anti-staphylococcal therapeutic candidates.
