Project description:In the model angiosperm Arabidopsis thaliana, SOMATIC EMBRYOGENESIS RECEPTOR-LIKE KINASES (SERKs), which are subfamily II of leucine-rich repeat receptor-like kinases (LRR-RLKs), play diverse roles in development and immunity as co-receptors for a multitude of LRR-RLKs including BRASSINOSTEROID INSENSITIVE 1 (BRI1) and FLAGELLIN SENSITIVE 2 (FLS2) . The conserved tyrosine (Y) residue in AtSERK3 plays a crucial role in signaling specificity for differentiating AtBRI1- and AtFLS2-mediated pathways. BRI ASSOCIATED RECEPTOR KINASE 1 (BAK1)-INTERACTING RECEPTOR-LIKE KINASES (BIRs) interact with SERKs under resting conditions, negatively regulating SERK-mediated pathways. SERK and BIR are highly conserved in land plants, whereas BRI1 and FLS2 homologs are absent or poorly conserved in bryophyte lineages. Very little is known about biological functions of SERK homologs in non-flowering plants. The genome of the model liverwort Marchantia polymorpha encodes the single SERK and BIR homolog, namely MpSERK and MpBIR. We here show that Mpserk disruptants display growth and developmental defects with no observable sexual and vegetative reproduction. Complementation analysis revealed a contribution of the conserved Y residue of MpSERK in growth. Proximity labelling-based interactomics of MpSERK identified MpBIR as an interactor. Mpbir disruptants displayed defects in reproductive organ development. Transcriptomes of Mpserk and Mpbir showed antagonistic patterns, suggesting MpBIR functions as a MpSERK repressor. We revealed that the pathogenic bacterium Pseudomonas syringae pv. tomato DC3000 could hardly grow on Mpbir, indicating a significant role of the MpSERK‒MpBIR module in immunity. Taken together, we propose that the SERK-BIR functional module was already established in the last common ancestor of land plants for regulating both development and immunity.

Project description:The lipid-derived phytohormone jasmonoyl-isoleucine (JA-Ile) regulates plant immunity, growth and development in vascular plants by activating genome-wide transcriptional reprogramming. In Arabidopsis, this is largely orchestrated by the master regulator MYC2 and related transcription factors (TFs). However, the TFs activating this pathway in basal plant lineages are currently unknown. We report the functional conservation of MYC-related TFs between the eudicot Arabidopsis thaliana and the liverwort Marchantia polymorpha, a plant belonging to one of the most basal land-plants lineages. Phylogenetic analysis suggests that MYC function first appeared in charophycean algae, and therefore predates the evolutionary appearance of any other jasmonate pathway component. Marchantia possesses two functionally interchangeable MYC genes, one in females and one in males. Similar to AtMYC2, MpMYCs showed nuclear localization, interaction with JAZ-repressors, and regulation by light. Phenotypic and molecular characterization of loss- or gain-of-function mutants demonstrated that MpMYCs are necessary and sufficient for the activation of the pathway in Marchantia, but unlike their Arabidopsis orthologs, do not regulate fertility. Our results show that despite 450 million years of independent evolution, MYCs are functionally conserved between bryophytes and eudicots. Genetic conservation in one of the most basal lineages suggests that MYC function existed in the common ancestor of land plants and evolved from a pre-existing MYC function in charophycean algae.

Project description:Land plants have co-evolved with microorganisms since its transition to a terrestrial habitat around 500 million years ago. In angiosperms, salicylic acid (SA) activates plant immunity against hemibiotrophic pathogens thought TGA transcription factors, which bind to the promoter of SA-responsive loci, such as pathogenesis-related (PR) genes, to enforce plants immunity. While those mechanisms are well-known in flowering plants, our understanding in bryophytes remains limited, as genetic evidences for the role of SA in plant immunity are still largely missing. Here, we explore the interaction between Marchantia polymorpha and the bacterium Pseudomonas syringae to gain insights into the evolutionary immune function of SA during bryophyte-pathogen interactions. We combined transcriptomic profiling of P. syringae-infected M. polymorpha with the generation of SA-deficient plants in this liverwort by overexpressing the bacterial NahG gene, a SA-degrading enzyme. Our results indicate that P. syringae induced transcriptional footprint is enriched in SA-responsive genes and that Marchantia NahG plants are compromised in immune responses against P. syringae. We found that MpTGA is modulated by SA and essential for controlling resistance against Pseudomonas. Further transcriptional analysis to identify the coregulatory network controlled by SA and MpTGA, indicate that a SA/MpTGA module might execute the boost of plants' defence responses thought a variety of MpPRs, highlighting a notable control of class III secretory peroxidases belonging to the MpPR9 subfamily. Altogether, our data demonstrate the functional conservation of SA as an immune hormone and underpin the existence of a SA-controlled MpTGA transcriptional cluster driving resistance against Pseudomonas in M. polymorpha

Project description:JAZ genes are negative regulators of jasmonate responses with a dual function as repressors of transcription factors and co-receptors, together with COI1, of the hormone jasmonoyl-isoleucine (JA-Ile). This family has been mainly studied in angiosperms, where high gene redundancy hinders the characterization of a complete depletion of JAZ function. Moreover, the recent discovery that JA-Ile is not the sole COI1/JAZ ligand in land plants, as dn-OPDA is the bioactive ligand in Marchantia polymorpha, underscores the importance of studying JAZ co-receptors in bryophytes. Here we exploited the low gene redundancy of the liverwort Marchantia polymorpha to characterize the function of the single MpJAZ in this early-divergent plant lineage. We demonstrate that MpJAZ is the ortholog of AtJAZ and acts as a repressor of dinor-OPDA responses in Marchantia. Mpjaz mutants show a dwarf phenotype and severe developmental defects related to growth inhibition, consistent with a constitutive activation of the dinor-OPDA pathway and the overaccumulation of both dinor-OPDA and its precursor OPDA. The expression of AtJAZ3 in Mpjaz complements MpJAZ repressor function, indicating that JAZ function is conserved across land plants. However, AtJAZ3 is unable to form co-receptor complexes with MpCOI1 and dn-OPDA, which evidences that the Jas domain, and not only COI1, determines ligand specificity.
