Project description:The mammalian testis is a highly heterogeneous tissue with a vast array of functional and regulatory RNA molecules that participate in the timely control of the spermatogenesis. This process remains largely unexplored in stallions. We utilized transcriptomic analysis to explore gene expression across various stages of testicular development in horses, from infancy to adulthood, including sperm. The analysis aimed to explore the regulatory networks involving messenger RNAs (mRNAs), long non-coding RNAs (lncRNAs), circular RNAs (circRNAs), and microRNAs (miRNAs). High-throughout RNA sequencing revealed dynamic changes in gene expression. The study identified key genes complex interactions between mRNAs, lncRNAs, circRNAs and miRNAs, and biological pathways associated with each developmental stage that drive the spermatogenesis in stallions and provide new insights into the regulatory mechanisms of horse testis spermatogenesis. This could aid in the improvement of the reproductive health and fertility management in horses.

Transcriptome profiling of developing testes and spermatogenesis in the Mongolian horse.

Project description:BackgroundHorse testis development and spermatogenesis are complex physiological processes.MethodsTo study these processes, three immature and three mature testes were collected from the Mongolian horse, and six libraries were established using high-throughput RNA sequencing technology (RNA-Seq) to screen for genes related to testis development and spermatogenesis.ResultsA total of 16,237 upregulated genes and 8,641 downregulated genes were detected in the testis of the Mongolian horse. These genes play important roles in different developmental stages of spermatogenesis and testicular development. Five genes with alternative splicing events that may influence spermatogenesis and development of the testis were detected. GO (Gene ontology) and KEGG (Kyoto Encyclopedia of Genes and Genomes) pathway analyses were performed for functional annotation of the differentially expressed genes. Pathways related to "spermatogenesis," male gamete generation," "spermatid development" and "oocyte meiosis" were significantly involved in different stages of testis development and spermatogenesis.ConclusionGenes, pathways and alternative splicing events were identified with inferred functions in the process of spermatogenesis in the Mongolian horse. The identification of these differentially expressed genetic signatures improves our understanding of horse testis development and spermatogenesis.

This dataset, consisting of the transcriptome of the developing testes from late larvae and early pupae of the African malaria mosquito, Anopheles gambiae, closes the existing gap.

Transcriptome Profiling of Developing Testes and First Wave of Spermatogenesis in the Rat.

Project description:Spermatogenesis is a complicated course of several rigorous restrained steps that spermatogonial stem cells undergo to develop into highly specialized spermatozoa; however, specific genes and signal pathways, which regulate the amplification, differentiation and maturation of these cells, remain unclear. We performed bioinformatics analyses to investigate the dynamic changes of the gene expression patterns at three time points in the course of the first wave of rat spermatogenesis. Differently expressed genes (DEGs) were identified, and the features of DEGs were further analyzed with GO (Gene Ontology), KEGG (Kyoto Encyclopedia of Genes and Genomes) and Short Time-series Expression Miner (STEM). A total of 2954 differentially expressed genes were identified. By using STEM, the top 10 key genes were selected in the profile according to the enrichment results, and the distinguishable biological functions encoded by these DEGs were automatically divided into three parts. Genes from 6, 8 and 10 days were related to biosynthesis, immune response and cell junction, and genes from 14, 15 and 16 days were related to energy metabolic pathways. The results also suggest that genes from 29, 31 and 35 days may shift metabolic to sperm motility, sperm flagellum and cilium movement.

Drosophila melanogaster late third instar larval testes and gonadal fatbody RNA-seq profiling

Project description:In order to study the effect of protease treatment on dissected testes, we performed different strategies of cleaning and dissociation on Drosophila melanogaster w1118 larval testes. We collected testes without protease treatment with fatbody just attached around the gonad, fatbody alone dissected from around the testes, testes without fatbody and testes dissociated by either by Papain or Trypsin and Collagenase cocktail. We prepared poly A+ RNA-Seq libraries and performed transcriptional profiling to generate 50 bp stranded single end reads.
