Project description:In order to investigate the underlying molecular mechanisms of the integrin-PINCH signaling pathways in angiogenesis, arterial samples from E14.5 mice were obtained for RNA extraction and RNA-seq sequencing. RNA-seq analysis showed that the enriched regulated genes were associated with various biological processes, including the ERK1/2 cascade, collagen biosynthetic process, and multiple cell behaviors , cell adhesion, migration, extracellular matrix remodeling, muscle contraction, etc. The results suggest that the PINCHs play an essential role in angiogenesis by modulating gene expression and signaling pathways as well as regulating cell behaviors of vascular wall, and deletion of PINCHs lead to altered biological processes, influencing the development and functionality of blood vessels, which are associated with cell adhesion, proliferation, apoptosis, and migration.

Pinch loss in early mesenchymal progenitors alters bone marrow environment

Project description:To investigate the alteration of bone marrow environment caused by Pinch loss, we performed single-cell RNA-sequencing on bone marrow cells, including hematopoietic cells and non-hematopietic stromal cells. As results indicated, we found both cell type proportion and transcriptomic signatures were dramatically altered by Pinch loss. We identified genes related to ossification, extracellular matrix organization, skeletal system development, and vasculature structure development were significantly differentially expressed between control group and dKO group in many mesenchymal stromal cells. Our results revealed Pinch loss disrupted homeostasis of bone marrow niche, monocytes in bone marrow were actively involved in inflammatory response, and functions of mesenchymal stromal cells was severely disrupted.

Project description:During peripheral nervous system (PNS) development, Schwann cells (SC) depend on interactions between extracellular matrix proteins, integrins and growth factor signal transduction pathways for survival, sorting and wrapping of axon segments to form myelin. Together with integrin-linked-kinase and parvins, PINCH (Particularly-interesting-new-cystidin-hystidin-rich protein) forms a heterotrimeric complex referred to as the IPP complex, which functions as a signaling hub connecting integrins with various signaling pathways. Using SC-specific conditional gene targeting in mice, gene expression profiling by RNAseq, and other methodologies, we demonstrate distinct functional roles for PINCH1 and PINCH2 in SC.

To identify pathogenic processes underlying these disease subtypes, using single endoscopic pinch biopsies to estabolish 36 expression profiles, we elucidated gene expression patterns of active and inactive areas of UC and CD, and compared these to infectious colitis and healthy controls.
