Project description:Cognitive decline is the most common and detrimental non-motor symptom of Parkinson’s disease (PD), and is an understudied disease entity. Pathologically, cognitive decline in PD is associated with alpha-synuclein (aSyn) misfolding and synapse loss in hippocampus and prefrontal cortex, leading to cognitive impairment and, ultimately, dementia. The mechanisms of cognitive decline and the factors driving them in PD are unknown. In the present study, we have used longitudinal gene expression profiling to characterise hippocampal molecular events in a transgenic mouse overexpressing E46K mutated aSyn, a model of early PD. Uncovering early events leading to disease is an essential step toward prognostic biomarker identification and early interventions. Transgenic aSyn induced synaptophysin loss in hippocampus and cortex of these mice. Comparing 4 different ages of mice from both sexes uncovered that hippocampal gene expression changes are sexually dimorphic and strongly modulated by the age, and by aSyn overexpression. Pathways that emerged across different comparisons were connected to a variety of neuronal functions, collagen synthesis/remodeling, cellular stress, and inflammatory responses. The findings indicate that sex and age are essential factors to consider when studying PD-associated cognitive decline. This may have important implications for prognostic biomarker identification and monitoring, and for timing of therapeutic approaches.

Project description:Parkinson's disease (PD) is a common human neurodegenerative movement disorder. Studies of the genetic forms of PD have helped to reveal disease mechanisms. Functional interactions between some Parkinson's disease (PD) genes, like PINK1 and parkin, have been identified, but whether other ones interact remains elusive. Here we report an unexpected genetic interaction between two PD genes, VPS35 and EIF4G1. We provide evidence that EIF4G1 upregulation causes defects associated with protein misfolding. Expression of a sortilin protein rescues these defects, downstream of VPS35, suggesting a potential role for sortilins in PD. We also show interactions between VPS35, EIF4G1 and alpha-synuclein, a protein with a key role in the pathogenesis of both sporadic and familial PD. We extend our findings from yeast to an animal model and show these interactions are conserved in neurons. We also connect VPS35 impairments to neurodegeneration in alpha-synuclein transgenic mice. Our studies reveal unexpected genetic and functional interactions between two seemingly unrelated PD genes and functionally connect them to alpha-synuclein pathobiology in yeast, worms, and mouse. Finally, we provide a resource of candidate PD genes for future genetic and functional interrogation. Project description:Aging is an inevitable process for everyone. Aging is mostly characterized by decline of cognitive function. Themechanisms are still not clear. Hippocampus has been shown to participate in cognition process,the basic mechanism of which are involved in molecularalterations in genes expression. Objective of the present research was to find the transcriptomic changes influenced by aging in hippocampus using RNA-sequencing. The 22-monthrats with cognition decline were used as a model system to analyze the changes in transcriptional networks. There were121 differentially expressed genes identified in the hippocampus of aged rats, including 54 genes upregulated and67 genes downregulated. After GO analysis,differentially expressed genes were found to be enriched inprotein hetorotrimerization, positive regulation of kinase activity, multivesicular body,mitochondrial membrane, oxygen binding and protease binding. KEGG analysesshowed thatDEGs were mainly observed at biochemical signaling pathways, containing Neuroactive ligand-receptor interaction (likeAplnr,Htr2a,Nmur2,Rxfp3), Apelin signaling pathway(likeAplnr,Cdh1,Serpine1),Serotonergic synapse（like Htr2a,Tph2）,Tryptophan metabolism（Tph2）,and so on. These DEGsin the hippocampusmay be involved in the formation of learning and memory. At last, quantitative reverse transcription-polymerase chain reaction was applied to verify the RNAsequencing data.Altogether, the above analysis help to identify specific genes in the aging hippocampus, which might be used as potential molecular targets for the prevention and treatment of aging-related cognition disorders.

Project description:The aim of this study was to identify alterations in hippocampal synaptic mRNA expression with aging and cognitive decline. Transcriptional profiling and subsequent bioinformatic analysis was performed to identify the most highly regulated pathways of genes. Interestingly, the antigen processing and presentation pathway was identified as the most highly regulated pathway with aging. Adult (12 month) and aged (28 month) Fischer 344 x Brown Norway (F1) hybrid rats were assessed for cognitive performance using the Morris water maze task and were divided into Adult (n=5), Aged Cognitively Intact (n=8), and Aged Cognitively Impaired (n=7) groups. One week following testing, all animals were sacrificed, the hippocampi were dissected, and synaptosomes were isolated for subsequent transcriptomic profiling. Only 5 cognitively intact animals were processed on the arrays.

Project description:Type 2 diabetes (T2D) is consistently associated with an elevated risk of cognitive decline and dementia. The underlying molecular mechanisms relating T2D to cognitive decline remain unclear, impeding the ability to detect the earliest stages of cognitive decline in older adults with T2D, consequently hindering advancement of preventive interventions. This study compared quantitative alterations in glycoproteoforms, represented by tryptic glycopeptiforms—specific glycan compositions on amino acid sites of proteins—in older T2D adults. We utilized label free, mass spectrometry-based glycoproteomics to analyze protein N-glycosylation and glycation patterns in serum samples, collected at two time points (T1 and T2), an average 52 months apart, from two initially cognitively healthy T2D groups. One group was comprised of individuals who experienced cognitive decline (“decliners”; N=8), and the other, of individuals who maintained normal cognition (“non-decliners”; N=14) over time. Comparison of these two groups revealed distinct patterns for glycosylation and glycation. Glycosylation primarily affected platelet function pathways. Glycation was also involved in immune system and platelet function along with various metabolic pathways. These findings suggest that aberrant glycoproteomic modifications may contribute to the development of cognitive impairment in T2D and may serve as potential early biomarkers. Further research is warranted to elucidate the functional implications of glycopeptiform modifications in T2D-related cognitive decline.

Project description:<p>Parkinson's disease (PD) is a common neurodegenerative disorder. It is marked by motor dysfunction and cognitive decline. In recent years, scientific studies have found that PD's pathogenesis may be tied to an imbalance in the gut microbiota. This offers new perspectives for PD treatment. Modulating the gut microbiota is recognized as a potential way to enhance PD symptoms. While aerobic exercise can positively influence the gut microbiota, research on how the gut microbiota mediates aerobic exercise's effects on PD cognitive impairment is still limited. Thus, this study aimed to explore the potential mechanisms by which aerobic exercise improves cognitive impairment in PD patients. It does so by modulating the gut microbiota's structure and, in turn, improving cognitive function. Through this study, we hope to offer new strategies and a theoretical basis for treating PD cognitive impairment.</p>

Project description:GBA mutations are major risk factors for Parkinson’s disease (PD) and Dementia with Lewy Bodies (DLB), two common α-synucleinopathies associated with cognitive impairment. Here, we investigated the role of GBA mutations in cognitive decline by utilizing Gba L444P mutant mice, SNCA transgenic (tg), and Gba-SNCA double mutant mice. Notably, Gba mutant mice showed early cognitive deficits but no PD-like motor deficits up to 12 months old. Conversely, SNCA tg mice displayed age-related motor deficits but no cognitive abnormalities. Gba-SNCA mice exhibited exacerbated motor deficits and cognitive decline. Immunohistological analysis revealed cortical phospho-α-synuclein pathology in SNCA tg mice, which was exacerbated in Gba-SNCA mice, especially in layer 5 cortical neurons. Significantly, Gba mutant mice did not show α-synuclein pathology. Single-nucleus RNA sequencing of cortices instead uncovered selective synaptic vesicle cycle defects in excitatory neurons of Gba mutant and Gba-SNCA mice, via robust downregulation in gene networks regulating synapse vesicle cycle and synapse assembly. Meanwhile SNCA tg mice displayed broader synaptic changes. Immunohistochemical and electron microscopic analyses validated these findings. Together, our results indicate that Gba mutations, while exacerbating pre-existing α-synuclein aggregation and PD-like motor deficits, contribute to cognitive deficits through α-synuclein-independent mechanisms, likely involving dysfunction in synaptic vesicle endocytosis.
