Project description:To understand the roles and mechanisms of neutrophil extracellular traps (NETs) in unilateral ureteral obstruction (UUO)-induced renal fibrosis, we performed RNA sequencing of UUO kidneys from wild-type and PAD4-deficient mice. PAD4 deletion led to reduced expression of fibrosis- and injury-associated genes, as well as inflammatory cytokines in UUO kidneys. Enrichment analysis of the downregulated genes in PAD4-deficient mice revealed a drastic decrease in inflammation pathways, such as T cell-associated pathways and cytokine-cytokine receptor interaction.

Project description:To understand the roles and mechanisms of DMM in unilateral ureteral obstruction (UUO)-induced renal fibrosis, we performed RNA sequencing of UUO kidneys from mice treated with or without dimethyl malonate (DMM). DMM administration led to metabolic reprogramming in obstructed kidneys, the genes associated with the metabolism of fatty acid, organic acid, cellular amino acid, and mitochondrial function were significantly upregulated. Conversely, the downregulated genes in DMM-treated mice were involved in inflammation, including T cell immunity. Moreover, the upregulated genes in DMM-treated mice were associated with PPAR signaling. The RNA-seq analysis suggests that DMM activated PPAR and metabolic pathways while inhibited inflammatory signalings.

Selective EGFR deletion in the fibroblast/pericyte population inhibited interstitial fibrosis in response to unilateral ureteral obstruction, ischemia or nephrotoxins. We used single-nucleus RNA sequencing analysis to demonstrate the role of EGFR activation in UUO model.

Stromal vascular fraction inhibits inhibits kidney fibrosis and inflammation in UUO mice

Project description:To understand the roles and mechanisms of stromal vascular fraction (SVF) in renal fibrosis induced by unilateral ureteral obstruction (UUO), we performed RNA sequencing of UUO kidneys from PBS- or SVF-treated mice. The transcriptome analysis revealed that SVF triggered significant metabolic reprogramming and improved mitochondrial function. Moreover, SVF treatment inhibited kidney inflammation as revealed by the downregulation of immune cell migration pathway. Furthermore, SVF contributed to the activation of PPAR signaling while inhibited TGF-beta signaling pathway.

Expressions of MicroRNA in Mouse Kidney with Unilateral Ureteral Obstruction

Project description:Renal fibrosis is the final common pathway for chronic kidney disease. However, the detailed mechanisms and therapeutic strategies for renal fibrosis have not been established. MicroRNAs (miRNAs) are small, non coding RNAs that regulate various pathological conditions and diseases. Previous studies reported that miRNAs play a pivotal role in renal fibrosis. However, the role of miRNAs in renal fibrosis has not been completely elucidated. Therefore, in this study, miRNAs on renal fibrosis was investigated in a renal fibrosis mouse model generated by unilateral ureteral obstruction. MiRNA microarray analysis revealed 109 miRNAs that were upregulated more than 2 fold and 113 miRNAs that were downregulated less than 0.5 fold in the kidney of UUO mice compared with control mice.

Project description:To find miRNAs that involve in renal epithelial transition and renal fibrosis, we performed unilateral ureteral obstruction of mice for 7 days. After that, we harvested kidneys, and performed microarray of miRNA. Contralateral kidneys without ureteral obstruction were used as controls. miRNAs were purified from kidneys with ureteral obstruction and contralateral kidneys without ureteral obstruction. Then microarray of miRNA was performed (n=4). miRNAs up-regulated in kidneys with ureteral obsctruction compared with contralateral kidneys were sorted. We performed unilateral ureteral obstruction of mice for 7 days, and harvested kidneys.

Kidney proteomics in the unilateral ureter obstruction (UUO) mouse model

Project description:Treatments for kidney fibrosis represent an urgent yet unmet clinical need. Effective therapies are limited due to not well understood molecular pathogenesis. We aimed at generating a comprehensive and integrated multi-omics data set (RNA/ microRNA transcriptomics and proteomics) of fibrotic kidneys which will be searchable through a user-friendly web application. Therefore, two commonly used mouse models were utilized: a reversible chemical-induced injury model (folic acid (FA) induced nephropathy) and an irreversible surgical-induced fibrosis model (unilateral ureteral obstruction (UUO)). RNA and small RNA sequencing as well as MS/MS with 10-plex tandem mass tags proteomics were performed with kidney samples from different time points over the course of fibrosis development. In summary, we present temporal and integrated multi-omics data from fibrotic mouse kidneys which are accessible through an interrogation tool to provide a searchable transcriptome and proteome for kidney fibrosis researcher.
