Project description:Lipid nanoparticles (LNPs) for mRNA delivery have advanced significantly, but LNP-mediated DNA delivery still faces clinical challenges. This study compared various LNP formulations for delivering DNA-encoded biologics, assessing their expression efficacy and the protective immunity generated by LNP-encapsulated DNA in different models. The LNP formulation used in Moderna’s Spikevax mRNA vaccine (LNP-M) demonstrated a stable nanoparticle structure, high expression efficiency, and low toxicity. Notably, a DNA vaccine encoding the spike protein, delivered via LNP-M, induced stronger antigen-specific antibody and T cell immune responses compared to electroporation. Single-cell RNA sequencing (scRNA-seq) analysis revealed that the LNP-M/pSpike vaccine enhanced CD80 activation signaling in CD8+ T cells, NK cells, macrophages, and DCs, while reducing the immunosuppressive signals. The enrichment of TCR and BCR by LNP-M/pSpike suggested an increase in immune response specificity and diversity. Additionally, LNP-M effectively delivered DNA-encoded antigens, such as mouse PD-L1 and p53R172H, or monoclonal antibodies targeting mouse PD-1 and human p53R282W. This approach inhibited tumor growth or metastasis in several mouse models. The long-term anti-tumor effects of LNP-M-delivered anti-p53R282W antibody relied on memory CD8+ T cell responses and enhanced MHC-I signaling from APCs to CD8+ T cells. These results highlight LNP-M as a promising and effective platform for delivering DNA-based vaccines and cancer immunotherapies.

Project description:Gene expression patterns provide insight into complex biological networks in which viruses and host cells interact. Dendritic cells are the most potent antigen presenting cells of the immune system and are crucial for the initiation of T-cell responses to viral pathogens. To better understand the biological effects of the EBV-encoded dUTPase on gene modulation in cells that play a central role in innate and adaptive immune responses, microarray gene expression profiling studies were performed in untreated or EBV-encoded dUTPase treated hDCs using the human genome U133 Plus 2.0 GeneChip. The results of this study identified over 800 differentially expressed genes between control and EBV-encoded dUTPase treated samples for each normalized data set using the criteria fold change >2 and p<0.05.

Advanced Assesement Through Intact Glycopeptide Analysis of Infliximab Biologics and Biosimilar

Project description:We demonstrated high-resolution tandem mass spectrometry with an ultra-high-performance liquid chromatography for characterization and comparison between biologics and biosimilar of infliximab at an advanced level. Comparing the N- and O-glycopeptides profiles, a total of 49 and 54 glycopeptides was identified for each product of the biologics and biosimilar, respectively. We also discovered one novel N-glycosylation site at the light chain from both biopharmaceuticals and one novel O-glycopeptide at the heavy chain from only biosimilar. Site-specific glycopeptide analysis process will be a robust and useful technique for evaluating therapeutic mAbs and complex glycoprotein products.

A new strategy to deliver synthetic protein drugs: self-reproducible biologics using minicircles.

Project description:Biologics are the most successful drugs used in anticytokine therapy. However, they remain partially unsuccessful because of the elevated cost of their synthesis and purification. Development of novel biologics has also been hampered by the high cost. Biologics are made of protein components; thus, theoretically, they can be produced in vivo. Here we tried to invent a novel strategy to allow the production of synthetic drugs in vivo by the host itself. The recombinant minicircles encoding etanercept or tocilizumab, which are synthesized currently by pharmaceutical companies, were injected intravenously into animal models. Self-reproduced etanercept and tocilizumab were detected in the serum of mice. Moreover, arthritis subsided in mice that were injected with minicircle vectors carrying biologics. Self-reproducible biologics need neither factory facilities for drug production nor clinical processes, such as frequent drug injection. Although this novel strategy is in its very early conceptual stage, it seems to represent a potential alternative method for the delivery of biologics.

Extensive modulation of inflammatory and axonal genes led to robust axon regeneration in severe benzalkonium chloride-injured corneas treated with cord blood-derived biologics

Project description:Modern ophthalmic solutions contain benzalkonium chloride (BAK), a potent antimicrobial preservative, that chronically damages the ocular apparatus. Umbilical cord blood serum (UCS) and platelet-rich plasma (UCPRP) are promising treatments for BAK-injured corneas. Here, for the first time, the feasibility of using both of these biologics for corneal treatment was demonstrated. Using animal studies and RNA-seq analysis, it was validated that severe BAK injury led to the upregulation of innate and adaptive immune responses, which corresponded to morphological and pathological changes in the cornea. When UCS and UCPRP were administered, besides significant corneal recovery, widespread immune pathways downregulation was observed. Subsequent verification of some of the genes involved in two of the most notable KEGG pathways suggested that the amelioration of inflammation was likely due to the reduction of immune cells, the decrease in chemoattractants, as well as the lessening of pro-inflammatory MMPs. Besides that, regulation of several key axonal genes led to robust axon recovery that correlated to nearly restored corneal sensitivity. Thus, these findings demonstrated the viability of cord blood-derived therapeutics for treating sustained, BAK-induced injuries in the cornea and also provided evidence that an optimal level of inflammatory response is crucial for stimulating axon regeneration.
