Separation of circulating microRNAs using apheresis in patients with systemic lupus erythematosus

Project description:Examined the removal or reduction of circulating miRNAs with apheresis. The levels of expression of a large number of circulating miRNAs were measured in the plasma samples separated by the primary membranes from all 3 patients with systemic lupus erythematosus. This is the first report that circulating miRNAs in peripheral blood can be separated and possibly directly removed using membrane separation apheresis.

Gene expression changes in multiple sclerosis relapse suggest activation of T and non-T cells

Project description:paired comparison of RNA expression in peripheral blood mononuclear cells in the same group of 14 multiple sclerosis patients while stable and while in relapse. A defining feature of multiple sclerosis is the occurrence of clinical relapses separated by periods of clinical stability. Better understanding of the events underlying clinical relapse might suggest new approaches to treatment. We used microarrays to measure mRNA expression in the peripheral blood of 14 MS patients during clinical relapse and while stable. Seventy-one transcripts changed expression at the p<0.001 significance level. The most notable finding was decreased expression of transcripts with regulatory function, expressed primarily in non-T cells. Transcripts with increased expression were primarily expressed in T cells. RBPome of primary mouse T helper cells

Project description:Post-transcriptional gene regulation by mRNA-binding proteins (RBPs) ensures an important layer to control protein outcome. It includes many fundamental processes, ranging from splicing to transport, decay and translation. Although recent technical developments allow the identification of mRNA-binding proteins, the total RBPome of different primary immune cells remains unknown. Here, we performed an RBP capture of the two major primary T helper cell types in mouse: T helper effector-like and regulatory-like (Foxp3+) cells. The two datasets show a high overlap of ~80% to reveal a high confidence common T helper cell RBPome of 240 proteins. 60% of those proteins have not been previously associated with any immune function. Most importantly, 11 RBPs were not previously identified through any mammalian mRNA-capture approach. These novel RBPs include some well-known T cell proteins associated with non-immune functions such as Stat1 and Vav1. Overall our data suggest an underestimated complexity of gene regulatory pathways.

Lung Natural Helper cells

Project description:Natural Helper cells constitute a unique lineage of Th2-cytokine producting innate lymphocytes, here we characterize the gene expression profile of non-stimulated or PMA/ionomycin-stimulated Natural Helper cells from naive C57Bl/6 mouse lungs. We used microarrays to detail the gene expression profile of stimulated and unstimulated lung Natural Helper cells in mice. Lineage(-)Sca-1(+)c-Kit(-/low)CD127(+)CD25(+) Lung Natural Helper cells were purified from naive 6-8 week old B6 mice by FACS. RNA was extracted immediately from un-stimulated Natural Helper cells after FACS. Stimulated Natural Helper cells were cultured with PMA/ionomycin for 3 days followed by RNA extraction.

Effector T Helper Cells Are Selectively Controlled During Pregnancy and Related to a Postpartum Relapse in Multiple Sclerosis.

Project description:Background: Multiple sclerosis (MS) patients are protected from relapses during pregnancy and have an increased relapse risk after delivery. It is unknown how pregnancy controls disease-contributing CD4+ T helper (Th) cells and whether this differs in MS patients who experience a postpartum relapse. Here, we studied the effector phenotype of Th cells in relation to pregnancy and postpartum relapse occurrence in MS. Methods: Memory skewing and activation of effector Th subsets were analyzed in paired third trimester and postpartum blood of 19 MS patients with and without a postpartum relapse and 12 healthy controls. Ex vivo results were associated with circulating levels of pregnancy-induced hormones and mirrored in vitro by exposing proliferating Th cells to corresponding serum samples. Results: Based on HSNE-guided analyses, we found that effector memory proportions of Th cells were increased in postpartum vs. third trimester samples from MS patients without a postpartum relapse. This was not seen for relapsing patients or healthy controls. CXCR3 was upregulated on postpartum memory Th cells, except for relapsing patients. These changes were verified by adding sera from the same individuals to proliferating Th cells, but did not associate with third trimester cortisol, estradiol or progesterone levels. For relapsing patients, activated memory Th cells of both third trimester and postpartum samples produced higher levels of pro-inflammatory cytokines. Conclusion: Effector Th cells are differentially regulated during pregnancy in MS patients, likely via serum-related factors beyond the studied hormones. The pro-inflammatory state of memory Th cells during pregnancy may predict a postpartum relapse.
