Project description:RGG motif-containing proteins are the second largest group of RNA-binding proteins. Here, we identify a unique and conserved role of yeast Scd6 and its human ortholog LSM14A in hydroxyurea (HU)-mediated genotoxic stress response. Scd6/LSM14A, but not all tested RGG-containing proteins, localize to cytoplasmic puncta in an RGG-dependent manner upon HU treatment. The absence of Scd6 increases cellular HU tolerance under normal conditions but sensitizes the cells to HU upon overexpression of SRS2, which is known to dampen the response to DNA damage. Scd6 binds the SRS2 mRNA to repress its translation upon HU stress. This interaction occurs in cytoplasmic granules and is modulated by the arginine methylation status of Scd6 and the Scd6 LSm domain, which acts as a cis-regulator of Scd6 arginine methylation. Polysome profiling experiments indicate that LSM14A regulate the translation of mRNAs encoding non-homologous end joining (NHEJ) DNA damage repair factors such as DNL4 (DNA ligase 4) and RTEL1 (the Srs2 ortholog), as well as the NHEJ activity in response to HU. Overall, the work shows the contribution of arginine methylation and the RGG-containing proteins Scd6/LSM14A in the genotoxic stress response by determining the translation status of a subset of mRNAs.

RGG-motif self-association regulates eIF4G-binding translation repressor protein Scd6.

Project description:Regulation of mRNA translation plays a key role in the control of gene expression. Scd6, a conserved RGG-motif containing protein represses translation by binding to translation initiation factor eIF4G1. Here we report that Scd6 binds itself in RGG-motif dependent manner and self-association regulates its repression activity. Scd6 self-interaction competes with eIF4G1 binding and methylation of Scd6 RGG-motif by Hmt1 negatively affects self-association. Results pertaining to Sbp1 indicate that self-association could be a general feature of RGG-motif containing translation repressor proteins. Taken together, our study reveals a mechanism of regulation of eIF4G-binding RGG-motif translation repressors.

Genome-wilde expression profiling analysis of yeast cells lacking the decapping activator Scd6

Project description:To assess the role of the decapping activator Scd6 in mRNA decay, we used RNA-Seq to analyze the expression profile of yeast cells harboring a deletion of the SCD6 gene. Consistent with our recent model for decapping regulation, we found that Scd6 targets a small number of specific mRNAs in yeast cells. Interestingly, degradation of Scd6-targeted transcripts also requires the functions of the decapping activators Pat1, Lsm1, and Dhh1, suggesting that Scd6 functions together with Pat1, Lsm1, and Dhh1 in promoting mRNA decapping.

Comparison of the specificities of the RGG/RG-domains of FMR1, FXR1 and FXR2 for small RNA

Project description:In previous studies, we identified a sequence motif (GDCGG) in almost all microRNAs downregulated in the serum of patients with amyotrophic lateral sclerosis. We found that FMR1, FXR1 and FXR2 directly and specifically interact with microRNAs containing the GDCGG motif via their RGG/RG-domains. Here, we compare the specificities for microRNAs of the RGG/RG-domains of FMR1, FXR1 and FXR2 on a transcriptome-wide scale using the Affymetrix GeneChip™ miRNA 3.0 Arrays.

Scd6 targets eIF4G to repress translation: RGG motif proteins as a class of eIF4G-binding proteins.

Project description:The formation of mRNPs controls the interaction of the translation and degradation machinery with individual mRNAs. The yeast Scd6 protein and its orthologs regulate translation and mRNA degradation in yeast, C. elegans, D. melanogaster, and humans by an unknown mechanism. We demonstrate that Scd6 represses translation by binding the eIF4G subunit of eIF4F in a manner dependent on its RGG domain, thereby forming an mRNP repressed for translation initiation. Strikingly, several other RGG domain-containing proteins in yeast copurify with eIF4E/G and we demonstrate that two such proteins, Npl3 and Sbp1, also directly bind eIF4G and repress translation in a manner dependent on their RGG motifs. These observations identify the mechanism of Scd6 function through its RGG motif and indicate that eIF4G plays an important role as a scaffolding protein for the recruitment of translation repressors.
