Spatial Transcriptomics identifies Prosaposin (PSAP) as a Novel Therapeutic Vulnerability in Pulmonary Sarcoidosis

Project description:Sarcoidosis is a chronic inflammatory disorder characterized by the presence of tiny collections of immune cells known as granulomas. An incomplete understanding of etiology and immune-pathologic pathways have limited the development of quality biomarkers and novel targeted therapies. We found a Fsp1-Cre mediated TSC1/2 deletion mice unexpectedly led to development of a disease state, which was identified and characterized by spatial transcriptomics and single-cell RNA sequencing, similar to sarcoidosis. Notably, we identified CCL24 as a novel key chemotactic regulator in this model of sarcoidosis and found decreased CCL24 in the sarcoid group, thus translating our model and hypothesis to the human form of the disease. Intriguingly, we found that CCL24 and azithromycin significantly blocked the progression of granuloma formation in this model. Our findings not only improve an integrated understanding of the progression of sarcoidosis but also provide the basis for developing novel potential clinical interventions in sarcoidosis therapeutics.

IFITM3 restricts virus-induced inflammatory cytokine production by limiting Nogo-B mediated TLR responses

Project description:Interferon-induced transmembrane protein 3 (IFITM3) is a restriction factor that limits viral pathogenesis and exerts poorly understood immunoregulatory functions. Here, using human and mouse models, we demonstrate that IFITM3 promotes MyD88-dependent, TLR-mediated IL-6 production following exposure to cytomegalovirus (CMV). IFITM3 also restricts IL-6 production in response to influenza and SARS-CoV-2. In dendritic cells, IFITM3 binds to the reticulon 4 isoform Nogo-B and promotes its proteasomal degradation. We reveal that Nogo-B mediates TLR-dependent pro-inflammatory cytokine production and promotes viral pathogenesis in vivo, and in the case of TLR2 responses, this process involves alteration of TLR2 cellular localization. Nogo-B deletion abrogates inflammatory cytokine responses and associated disease in virus-infected IFITM3-deficient mice. Thus, we uncover Nogo-B as a driver of viral pathogenesis and highlight an immunoregulatory pathway in which IFITM3 fine-tunes the responsiveness of myeloid cells to viral stimulation.

Differential Transcriptome Features of Peripheral Blood Mononuclear Cells in Pulmonary Sarcoidosis with and without Extrapulmonary Lesions

Project description:Sarcoidosis is a systemic granulomatous disease of unknown etiology. Pulmonary sarcoidosis with extrapulmonary lesions (EPL), especially in cardiac sarcoidosis, is associated with poor prognosis. The transcriptome features of peripheral blood mononuclear cells (PBMCs) could be crucial in sarcoidosis pathogenesis. However, the gene expression characteristics associated with EPL development in pulmonary sarcoidosis remain unknown. Therefore, we investigated gene expression patterns associated with the development of EPL in pulmonary sarcoidosis. First, we conducted trascriptome analysis between patients with pulmonary sarcoidosis and healthy controls. Principal component analysis (PCA) revealed a clear distinction between them, with 227 differentially expressed genes detected. Enrichment analysis revealed upregulated immunological pathways related to granuloma formation in pulmonary sarcoidosis PBMCs, including T helper 17 and tumor necrosis factor-alpha signaling pathways, inflammatory cytokine production, and response to external stimuli. Furtheremore, PCA differentiated patients with pulmonary sarcoidosis with and without EPL, and 206 differentially expressed genes identified, including interferon-gamma (IFNG) and interferon lambda receptor 1 (IFNLR1). Gene ontology (GO) analysis revealed that interleukin 6 (IL-6) and IL-23 production were upregulated in patients with pulmonary sarcoidosis and EPL. In cardiac sarcoidosis, nitric oxide synthases (NOS), including NOS1 and NOS2, were upregulated in PBMCs. The current study strengthens our understanding of the mechanisms underlying granuloma formation in sarcoidosis and demonstrates the differential transcriptome features of PBMCs in patients with pulmonary sarcoidosis with and without EPL, highlighting IFNG and IFNLR1 upregulation. The upregulation of these genes may be related to EPL development and could serve as potential therapeutic targets for sarcoidosis.
