Project description:In plants, sperm cell formation involves two rounds of pollen mitoses, in which the microspore initiates the first pollen mitosis (PMI) to produce a vegetative cell and a generative cell, then the generative cell continues the second mitosis (PMII) to produce two sperm cells. DUO1, a R2R3 Myb transcription factor, is activated in the generative cell to promote S-G2/M transition during PMII. Loss-of-function of DUO1 caused a complete arrest of PMII. Despite the importance of DUO1, how DUO1 is regulated is largely unexplored. We previously demonstrated that ARID1, an ARID transcription factor, stimulates DUO1 transcription. Here, we show that cell proliferation suppressor RBR1 interacts with ARID1 to stabilize DUO1. While the C-terminus of RBR1 is dispensable for vegetative growth, it plays a crucial role in reproductive development and facilitates interaction with ARID1. Moreover, DUO1 is a short-lived protein, ARID1 promotes the RBR1-DUO1 interaction, and RBR1 stabilizes DUO1 in a proteasome-dependent manner. Thus, RBR1 promotes DUO1-dependent PMII progression via antagonizing its repressive role in the cell cycle factors CDKA;1 and CYCB1;1. Collectively, we uncover that ARID1 and RBR1 act in concert to regulate DUO1 at both the transcriptional and post-transcriptional levels, balancing cell specification and cell division.

Project description:Heterochromatin de-condensation in companion gametic cells is conserved in both plants and animals. In plants, microspore undergoes asymmetric pollen mitosis (PMI) to produce a vegetative cell (VC) and a generative cell (GC). Subsequently, the GC undergoes pollen mitosis (PMII) to produce two sperm cells (SC). Consistent with heterochromatin de-condensation in the VC, H3K9me2, a heterochromatin mark, is barely detected in VC. However, how H3K9me2 is differentially regulated during pollen mitosis remains unclear. Here, we show that H3K9me2 is gradually evicted from the VC since PMI but remain unchanged in the GC and SC. ARID1, a pollen-specific transcription factor that facilitates PMII, promotes H3K9me2 maintenance in the GC/SC but slows down its eviction in the VC. The genomic targets of ARID1 mostly overlaps with H3K9me2 loci, and ARID1 recruits H3K9 methyltransferase SUVH6. Our results uncover that differential pattern of H3K9me2 between two cell types is regulated by ARID1 during pollen mitosis.

Project description:Crop reproductive success is significantly challenged by heatwaves, which are increasing in frequency globally. The main reason is reduced male fertility due to abnormal pollen development, but the mechanism behind the developmental deviation is not well understood. Here, long-term mild heat (LTMH) treatment, mimicking a heatwave, was applied to flowers or whole plants and followed up by cytological, transcriptomic and biochemical analyses. LTMH was shown to act directly on the flowers and not via a systemic effect on other plant tissue. The meiosis to early microspore stage was the most to LTMH and three to four days of LTMH exposure around this period was sufficient to significantly reduce pollen viability. Extensive cytological analysis showed that abnormalities in pollen development could first be observed after pollen mitosis I and tapetum development appeared unaffected. Transcriptomic and biochemical analyses suggested that pollen development suffers from tapetal ER stress, with a limited role for oxidative stress. These characteristics differentiate the response of developing anthers and pollen to LTMH from that to severe heat stress.

Project description:Purpose:The molecular mechanism of pollen abortion in interspecific hybrids of B.rapa and B.nigra was revealed by cytological and transcriptional analysis Methods:floral buds (contain two developmental progress,1.1-1.5 mm and 1.5-6.0 long floral buds) were collected, then Separated male organs were kept in liquid nitrogen immediately until use. For the quantitative real-time polymerase chain reaction (qRT-PCR) analysis Results:In this study, there were extensive variations in gene expression patterns of this hybrid, and down-regulated expression of key genes for pollen development, such as meiosis of pollen mother cells, DNA damage repair and pollen sperm cell differentiation, may lead to pollen abortion. Styrene-propyl synthesis and wax biosynthesis pathway were blocked, which affected pollen wall formation. Conclusions:The wide variation of genes showed that the expression of A and B genomes were affected differently. B genome tended to downregulate the expression of key genes during meiosis and mitosis, while A genome tended to downregulate the expression of genes related to stress and pollen wall formation. The results showed that the development of male gametophyte was seriously affected after synthesis.

Project description:Globally, the three categories of intact SWI/SNF complexes in control cells have shared and unique binding peaks, with cBAF showing the most abundant binding sites followed by ncBAF and pBAF. In control cells, within cBAF, ARID1B bound to only a subset of ARID1A peaks. In control cells, the binding pattern of BAF45d corresponded to ARID1A patterns, but binding was virtually eliminated in ARID1-less cells. In ARID1-less cells, ARID2 binding was essentially eliminated whereas Brd9 binding was unaltered.

Project description:Oryza longistaminata is an African wild rice species that possesses special traits for breeding applications. Self-incompatibility is the main cause of sterility in O. longistaminata, but here we demonstrated that its pollen fertility and vitality are normal. Lipid and carbohydrate metabolism were active throughout pollen development. In this study, transcriptomics quantitative analysis was used to investigate the profiles of genes related to lipid and carbohydrate metabolism in 4-, 6- and 8.5-mm O. longistaminata spikelets before flowering. We documented cytological changes throughout important stages of anther development, including changes in reproductive cells as they formed mature pollen grains through meiosis and mitosis. RNA-seq and proteome association analysis indicated that fatty acids were converted to sucrose after the 6-mm spikelet stage, based on the abundance of most key enzymes of the glyoxylate cycle and gluconeogenesis. In conclusion, our study provides novel insights into the pollen viability of O. longistaminata at the transcriptome level, which can be used to improve the efficiency of male parent pollination in hybrid rice breeding applications.

In mitosis, it serves as an assembly platform that interacts with chromatin and then with nucleoporin subcomplexes to initiate the formation of novel nuclear pore complexes. Here we describe the interaction of ELYS with the membrane protein VAPB. In mitosis, ELYS becomes phosphorylated at many sites, including a predicted FFAT (two phenylalanines in an acidic tract) motif, which is shown to mediate interaction with the MSP (major sperm protein)-domain of VAPB. Phosphorylation-dependent binding of VAPB to ELYS is demonstrated by peptide binding assays and co-immunoprecipitation experiments. In anaphase, the two proteins co-localize to the non-core region of the newly forming nuclear envelope. Depletion of VAPB resulted in prolonged mitosis and slow progression from meta- to anaphase and also to chromosome segregation defects. Together, our results suggest an active role of VAPB in recruiting membrane fragments to chromatin and in the biogenesis of a novel nuclear envelope during mitosis.
