Project description:Cortical atrophy and dysfunction are hallmarks of many neuropsychiatric diseases including depression, post-traumatic stress disorder (PTSD), and addiction. Psychoplastogens are small molecules with the potential to rapidly rescue these structural and functional deficits, leading to sustained therapeutic effects after a single administration. Classic psychedelics are among the most potent and efficacious psychoplastogens known, with recent data indicating that their clinical and preclinical effects might be more robust and durable than those of ketamine. Moreover, the advent of non-hallucinogenic psychoplastogens structurally related to psychedelics, such as tabernanthalog (TBG), has opened new opportunities for developing scalable alternatives capable of promoting structural and functional neuroplasticity in the prefrontal cortex (PFC). Currently, it is unclear what roles 5-HT2A receptors (5-HT2ARs) and immediate early gene activation play in the neuroplasticity-promoting effects of non-hallucinogenic psychoplastogens like TBG—information necessary to engineer optimized neurotherapeutics related to psychedelics. Here, we use a combination of pharmacological and genetic tools to demonstrate that several non-hallucinogenic psychoplastogens promote cortical neuron growth through the same biochemical pathway as classic psychedelics. Furthermore, we demonstrate that there is a causal relationship between TBG-induced cortical spinogenesis and its sustained antidepressant-like effects. In contrast to psychedelics, TBG promotes cortical neuron growth in the absence of a glutamate burst or immediate early gene (IEG) activation, effects assumed to be necessary for psychedelic-induced neuroplasticity. Our results elucidate how the hallucinogenic effects of psychedelics can be decoupled from their beneficial effects on cortical neuron growth.

Project description:Immediate early genes have been known and studied for decades. Their fast and transient induction, which depends on phosphorylation cascades and is independent of de novo protein synthesis, orchestrates the cellular response to various stimuli. In this study we show that the same phosphorylation cascades also target the splicing machinery to induce an analogous splicing switch that we call immediate early splicing (IES). We characterize hnRNPC2-controlled IES, which depends on the MEK-ERK pathway and the T cell specific PKCθ. This splicing switch mainly targets components of the translation machinery and contributes to reduced de novo protein synthesis early upon T cell activation, to coordinate extensive transcriptional and posttranscriptional changes in gene expression. Together, our data set a paradigm for fast and transient alternative splicing, the immediate cellular response to activation, and provide evidence for its functional relevance during T cell activation.

Project description:Asthma is a chronic inflammatory disease of the airways characterized by airway hyperresponsiveness. Recent studies using mouse models, bronchial biopsies of asthma patients, and lung tissue of patients with fatal asthma have implicated neuroplasticity as a pathological feature of severe asthma, possibly contributing to airway hyperresponsiveness. Currently, we lack the models to study the mechanisms behind sensory neuroplasticity in vitro. Here, we describe the differentiation of sensory neurons from human pluripotent stem cells. Generation of sensory neurons was validated using immunofluorescence, flow cytometry and RNA-sequencing, as well as functional responsiveness to capsaicin using calcium imaging. We exposed these hPSC-derived sensory neurons to TGF-β or Th2-cytokines IL-4 together with IL-13, both of which have important roles in asthmatic airway remodelling. Both treatments induced neuroplasticity-related changes such as increased network density and neuronal sensitivity in the sensory neurons, albeit TGF-β more strongly than IL-4+IL-13. Our results show that hPSC-derived sensory neurons can be generated robustly and reproducibly and provide a good model to investigate the mechanisms behind neuroplasticity. Furthermore, our findings support a role for the asthmatic airway microenvironment, most notably TGF-β, in the development of neuroplasticity.

Project description:Chromatin-organizing factors, like CTCF and cohesins, have been implicated in the control of complex viral regulatory programs. We investigated the role of CTCF and cohesin in the control of the latent to lytic switch for Kaposi's Sarcoma-Associated Herpesvirus (KSHV). We found that cohesin subunits, but not CTCF, were required for the repression of KSHV immediate early gene transcription. Depletion of cohesin subunits Rad21, SMC1, or SMC3 resulted in lytic cycle gene transcription and viral DNA replication. In contrast, depletion of CTCF failed to induce lytic transcription or DNA replication. ChiP-Seq analysis revealed that cohesins and CTCF bound to several sites within the immediate early control regions for ORF50 and more distal 5' sites that also regulate the divergently transcribed ORF45-46-47 gene cluster. Rad21 depletion led to a robust increase in ORF45 and ORF47 transcripts, with similar kinetics to that observed with chemical induction by sodium butyrate. During latency, the chromatin between the ORF45 and ORF50 transcription start sites was enriched in histone H3K4me3 with elevated H3K9ac at the ORF45 promoter and elevated H3K27me3 at the ORF50 promoter. A paused form of RNA pol II was loosely associated with the ORF45 promoter region during latency, but was converted to an active elongating form upon reactivation induced by Rad21 depletion. Butyrate-induced transcription of ORF45 and ORF47 was resistant to cyclohexamide, suggesting that these genes have immediate early features similar to ORF50. Butyrate-treatment caused the rapid dissociation of cohesins and loss of CTCF binding at the immediate early gene locus, suggesting that cohesins may be a direct target of butyrate-mediated lytic induction. Our findings implicate cohesins as a major repressor of KSHV lytic gene activation, and function coordinately with CTCF to regulate the switch between latent and lytic gene activity. Project description:Analysis of Immediate Early Response 2 (Ier2)-inducible NIH 3T3 cells after Ier2 induction with RheoSwitch ligand RSL-1. Results provide insight into the function of Ier2 in NIH 3T3 mouse embryonal fibroblasts. Immediate early genes, including Ier2, are rapidly induced in quiescent cells by proliferation and migration-inducing stimuli.
