Project description:The tumor microenvironment (TME) is programmed by cancer cells and critically influences antitumor immune responses. Within the TME, CD8+ T cells undergo full effector differentiation and acquire cytotoxic antitumor functions in specialized niches. While interactions with conventional type-1 dendritic cells (cDC1s) have been implicated in this proces, underlying cellular players and molecular mechanisms remain incompletely understood. Here, we show that inflammatory monocytes can adopt a critical role in intratumoral T cell stimulation. They express Cxcl9, Cxcl10 and Il15, but as opposed to cDC1s that cross-present antigens, inflammatory monocytes obtain and present peptide-major histocompatibility complex class I (pMHCI) complexes from tumor cells through “cross-dressing”. Hyperactivation of MAPK signaling in cancer cells hampers this process by coordinately blunting the production of type I interferon (IFN-I) and inducing the secretion of prostaglandin E2 (PGE2), impairing the inflammatory monocyte state and intratumoral T cell stimulation. Enhancing IFN-I production and blocking PGE2 secretion restores this process and thereby re-sensitizes tumors to T cell-mediated immunity. Together, our work uncovers a central role of inflammatory monocytes in intratumoral T cell stimulation, elucidates how oncogenic signaling disrupts T cell responses via counter-regulation of PGE2 and IFN-I, and proposes rational combination therapies to enhance immunotherapies.

Acquired resistance to targeted therapy confers cross-resistance to immunotherapy

Project description:We identify a profoundly remodelled TME in targeted therapy resistant (RTT) tumours. We provide bulk transcriptomic data of targeted therapy naive (NTT) and RTT tumours of 3 murine models. Additionally, we provide low-input sequencing of CD103+ dendritic cells and T cells from NTT and RTT Braf/Pten tumours. We identified that in RTT cell lines the MAPK pathway is reactivated and provide SLAM-seq data to determine transcriptional targets of MAPK pathway inhibition in NTT and RTT cell lines. We also provide ATAC-Seq data of NTT and RTT cell lines sorted from tumors of the Braf/Pten melanoma model.

CITE-Seq analysis of intratumoral immune cells from YUMM1.7 melanoma treated with anti-PD-1 therapy.

Project description:Metastatic melanoma remains a disease difficult to treat, as 50% of the patients are resistant to immunotherapies or experience high toxicities. Investigations to identify underlying mechanisms focus on T cells or the immunosuppressive tumormicroenvironment (TME). Tumor-associated macrophages (TAMs) make up a large fraction of the TME and high levels of TAM have been associated with poor survival of cancer patients. In this study, the subcutaneous YUMM1.7 melanoma model was used to investigate the responsiveness to anti-PD-1 therapy. The tumor cell line was injected into GRf/f and GRf/f-LysMCre mice to enable insights into glucocorticoid-dependent effects within the myeloid compartment and how they contribute to the responsiveness to anti-PD-1 immunotherapy. Therefore, we profiled sorted CD45+ immune cells from tumors of GRf/f and GRf/f-LysMCre treated with anti-PD-1 therapy or an isotype control antibody via the cellular indexing of transcriptomes and epitomes-method (CITE-Seq).
