Project description:Comparative transcriptional profiling of N. crassa grown on five major crop straws of China (barley, corn, rice, soybean and wheat straws) revealed a highly overlapping group of 430 genes, the Biomass commonly Induced Core Set (BICS). A large proportion of induced carbohydrate-active-enzyme (CAZy) genes (82 out of 113) were also conserved across the five plant straws. Excluding 178 genes within the BICS that were also up-regulated under no-carbon conditions, the remaining 252 genes were defined as the Biomass Regulon (BR). Interestingly, 88 genes were only induced by plant biomass and not by three individual polysaccharides (Avicel, xylan, and pectin); these were denoted as the Biomass Unique Set (BUS). Deletion of one BUS gene, the transcriptional regulator rca-1, significantly improved lignocellulase production using plant biomass as the sole carbon source, possibly functioning via de-repression of the regulator clr-2. Thus, this result suggests that rca-1 is a potential engineering target for biorefineries, especially for plant biomass direct microbial conversion processes.

Project description:Bisphenol A (BPA), which is used in a variety of consumer-related plastic products, was reported to cause metabolic disruption. Substitute compounds are increasingly emerging, but are not sufficiently investigated. In this study, we aimed to investigate the mode of action of BPA and four of its substitutes during the differentiation of human preadipocytes. Human preadipocytes were exposed to BPA, BPS, BPB, BPF, and BPAF, as well as the PPARγ-antagonist GW9662. After 12 days of differentiation, global protein profiles were assessed using LC-MS/MS-based proteomics.

Project description:BackgroundPiglets at weaning suffer many stressors such as sudden change of feed, change in group composition and the end of lactogenic immunity. These stressors may cause poor growth performance. There is a need for alternatives to support piglets during the weaning period. Organic acids are known to have a positive effect on performance through reducing the pH and their antimicrobial action.Context & purposeThe purpose was to study the effect of the inclusion of a free and buffered organic acid blend in drinking water on performance of weaned pigs. Four-hundred and twenty pigs in a conventional herd were allocated after weaning to one of three treatments and monitored during 4 weeks: group (1) Full medication, group (2) organic acid blend + full medication, group (3) organic acid blend + reduced medication. Average daily gain, feed intake and water consumption was recorded at group level.ResultsDuring the overall study period live weight and average daily gain of the piglets was significantly higher (P <0.001) for treatment (3) compared to (1) and (2) (Table 1). Live weight was significantly higher for treatment (3) compared to (1) from week 2 of the study (Fig. 1). No significant differences were found for average daily feed intake. FCR for treatment (3) improved by 1.0 compared to treatment (1) in week 1 (P <0.05), while in week 2 and 3 no significant differences were found (Table 2). Overall, FCR was with 0.3 difference significantly lower (P = 0.001) for treatment (3) than for (1) and (2) (Table 1). Pigs receiving organic acids in drinking water had significantly (P <0.05) higher water consumption than group (1) in weeks 3 and 4 (Table 2).Conclusion & potential implicationsThe use of a blend of free and buffered organic acids together with a reduced medication program improves growth performance during the first month after weaning compared to a control with full medication and a combination between organic acids and full medication. This implies that organic acids could be used as a valid alternative for antibiotic reduction in post-weaning pigs. The treatment also increased the drinking water intake.

Project description:The adverse effects of unsustainable behaviors on human society are leading to an increasingly urgent and critical need to change policies and practices worldwide. This requires that citizens become informed and engaged in participatory governance and measures leading to sustainable futures. Citizens' understanding of the inherent complexity of sustainable systems is a necessary (though generally not sufficient) ingredient for them to understand controversial public policies and maintain the core principles of democratic societies. In this work, we present a novel, open-ended experiment where individuals had the opportunity to solve model urban sustainability problems in a purposeful game. Participants were challenged to interact with familiar LEGO blocks representing elements in a complex generative urban economic indicators model. Players seeks to find a specific urban configuration satisfying particular sustainability requirements. We show that, despite the intrinsic complexity and non-linearity of the problems, participants' ability to make counter-intuitive actions helps them find suitable solutions. Moreover, we show that through successive iterations of the experiment, participants can overcome the difficulties linked to non-linearity and increase the probability of finding the correct solution to the problem. We contend that this kind of what-if platforms could have a crucial role in future approaches to sustainable developments goals.

Project description:The circular bioeconomy has become a crucial strategy for sustainable development, especially by upcycling organic-rich by-products from various industrial processes. Purple non-sulphur bacteria (PNSB) have emerged as excellent candidates in this field, demonstrating exceptional metabolic versatility. While the growth of PNSB on sugar-rich streams has been extensively explored, the sugar assimilation metabolism remains poorly understood. Here, we explore the metabolic mechanisms of sucrose utilisation in two phototrophic purple non-sulphur bacteria (PNSB), Rhodospirillum rubrum and Rhodobacter capsulatus. Our findings demonstrate distinct carbohydrate hydrolysis and assimilation capacities, as well as the use of different redox strategies for each species. Moreover, Rhodospirillum rubrum could only grow on sucrose when co-cultivated with Rhodobacter capsulatus. This trophic link between Rhodospirillum rubrum and Rhodobacter capsulatus in sucrose containing co-culture was characterised and resulted in significantly enhanced productivity compared to pure cultures. Finally, we demonstrate that the synergy observed between Rhodospirillum rubrum and Rhodobacter capsulatus can be successfully scaled up in a photobioreactor system. Our study highlights how fundamental knowledge of the metabolism, trophic link and general microbial ecology concept might be useful for the development of biobased resource recovery strategies.
