Project description:Enhanced nutrition during the early calfhood period has been shown to hasten the onset of puberty in bull calves. The hypothalamic-pituitary-ovarian biochemical signalling axis regulates reproductive development in bulls, with the dynamics of the arcuate nucleus region of the hypothalamus, in particular, central to mediating the relationship between metabolic status with reproductive development. However, the precise molecular mechanisms regulating the influence of metabolic status on this signalling axis in bull calves is yet to be fully elucidated. The objective of this study was to determine the effect of an enhanced plane of nutrition during early life, up to 12 weeks of age, on the proteomic profile of the arcuate nucleus of bull calves. Bull calves were offered either a high or moderate plane of nutrition.

Holstein bull calves were designated to high (HI; n=15) or moderate (MOD; n=12) dietary groups, with diets designed to provoke growth rates of 1.0 and 0.5 kg/day, respectively. At 12 wk of age, all calves were euthanized, and hypothalamic arcuate nucleus tissue harvested. RNA was extracted from the hypothalamic arcuate nucleus and used for miRNAseq.

Effect of early-life plane of nutrition on the transcriptional profile of the hypothalamic arcuate nucleus region in heifer calves

Project description:The aim of this study was to examine alterations in the transcriptional profile of the arcuate nucleus region of the hypothalamus between heifer calves offered varying planes of nutrition during early life. Angus X Holstein-Friesian heifer calves (19 ± 5 days of age, BW: 51.2 ± 7.8 kg, mean ± SD) were offered a high (HP, n=14) or moderate plane of nutrition (MP, n=15) from 3 to 21 weeks of age to achieve a target growth rate of 1.2 kg/d and 0.5 kg/d, respectively. At 21 weeks of age, all calves were euthanized and the arcuate nucleus region of the hypothalamus of each calf recovered. RNA was extracted transcriptional profiles examined using RNAseq. Compared with calves on the moderate plane of nutrition, a High plane of nutrition altered the expression of 80 genes within the arucate nucleus. Functional analysis of differentially expressed genes revealed functions including organismal development, behaviour, endocrine system development and function, lipid metabolism, molecular transport, small molecule biochemistry, cellular development, cellular growth and proliferation, and nervous system development and function to be affected by early life plane of nutrition in the arucate nucleus of heifer calves.

At 12 wk of age, all calves were euthanized, and testes parenchyma harvested. RNA was extracted from the testes and used for miRNAseq. Bioinformatic analysis identified 7 miRNA as differentially expressed between the HI and MOD treatment groups, with target mRNA genes invovled in pathways related to AMPK and IGF-1 signaling.

Proteomic analysis underlying the impact of early life nutrition on the molecular control of testicular development in the young bull calf.

Project description:It has been established that enhanced early life nutrition progresses sexual development in the bull calf through neuroendocrine signalling via the hypothalamic-pituitary-testicular axis. However, the underlying molecular mechanisms regulating this process have not been fully elucidated. This study measured the impact of contrasting feeding regimes in the first 12 wk of life, known to impact age at puberty, on the proteomic landscape of the testes of bull calves. Holstein bull calves with a mean (±SD) bodyweight and age of 48.8 (± 5.3) kg and 17.5 (± 2.8) days, were designated to high (HI; n=10) or moderate (MOD; n=10) dietary groups, with diets designed to provoke growth rates of 1.0 and 0.5 kg/day, respectively. At 12 wk of age, all calves were euthanized, and testes parenchyma harvested. HI calves were heavier at slaughter (112.4 v 88.7 (2.98) kg, P<0.001), and had a greater average daily gain (ADG) of (0.88 v 0.58 kg, P<0.001). The turquoise network from the protein analyses contained the protein CDH13 which is involved in testes development. Gene ontology analysis of the turquoise network revealed enrichment of genes with functions related to cholesterol biosynthesis, IGF-1 signalling, insulin receptor/secretion signalling, androgen signalling and Sertoli cell junction signalling.

Project description:The objective of this study was to examine changes in hypothalamic gene expression of heifer calves offered varying planes of nutrition during early life. Angus X Holstein-Friesian heifer calves (19 ± 5 days of age, BW: 51.2 ± 7.8 kg, mean ± SD) were offered a high (HP, n=14) or moderate plane of nutrition (MP, n=15) from 3 to 21 weeks of age to achieve a target growth rate of 1.2 kg/d and 0.5 kg/d, respectively. At 21 weeks of age, all calves were euthanized and the hypothalamus of each calf recovered. RNA was extracted and hypothalamic gene expression examined using RNAseq. Compared with calves on the moderate plane of nutrition, a HP of nutrition altered the expression of 39 transcripts in the hypothalamus. Biological pathway analysis revealed these differentially expressed genes to be involved in pathways including acute phase response signalling, STAT3 pathway, and tight junction signalling. Additionally, biological functions affected by early life plane of nutrition included cell-to-cell signalling and interaction, molecular transport, small molecule biochemistry, behaviour, nervous system development and function, nucleic acid metabolism, and cellular function and maintenance.

Effect of early calf-hood nutrition on the transcriptomic profile of the HPT axis in Holstein-Friesian bulls

Project description:The aim of this study was to investigate the effect of early calf-hood nutrition on the transcriptomic profile of the arcuate nucleus of the hypothalamus, the anterior pituitary and the testes in Holstein-Friesian bulls and possible downstream effects on reproduction. Holstein Friesian bull calves with a mean (±S.D.) age and bodyweight of 19 (±8.2) days and 47.5 (±5.3) kg, respectively, were assigned to either a high (n=10) or low (n=10) plane of nutrition. Calves were fed in order to achieve an overall growth rate of 1.2 and 0.5 kg/day for the high and low plane of nutrition treatments, respectively. At 126 days of age, the bulls were euthanized using an intravenous overdose of sodium pentobarbitone and arcuate nucleus of the hypothalamus, anterior pituitary and the parenchyma of the testes tissue samples were harvested and RNAseq analysis was performed.
