Project description:Cardiovascular progenitors (CPs) are responsible for generating the diverse cell populations in hearts, exhibiting significant transcriptional heterogeneity. However, the regulation of fate plasticity among heterogeneous CP subpopulations, as well as the intrinsic regulatory factors, remains poorly understood. In this study, we characterized three CP subpopulations derived from pluripotent stem cells and found that the deletion of ZBTB family protein Zbtb16 led to a profound imbalance in the branching transition trajectory of Early-CPs, redirecting them toward CPs committed to endothelial cells and cardiac fibroblasts (EC/CF-CPs) rather than that to cardiomyocytes (CM-CPs). Mechanistic studies revealed that Zbtb16 interacted with the m6A reader IGF2BP3 to post-transcriptionally recognize and stabilize the mRNAs of key genes critical for the CM-CP subpopulation determination. Collectively, our findings established Zbtb16 as a key regulator of fate plasticity of heterogeneous CP subpopulations and revealed the interplay with IGF2BP3 to impact the m6A-modified mRNA stabilization, which provided new insights into the intrinsic connections between CP subpopulation plasticity and cardiovascular multi-lineage fate determination.

RIP-sequence of IGF2BP3 in Ishikawa cells

Project description:To explain the possible molecular mechanism underlying the oncogenic roles of IGF2BP3 in EC, we employed RNA immunoprecipitation (RIP) assays to identify the lncRNAs involved in the regulation of IGF2BP3 function. RIP experiments, high-throughput sequencing and data analysis were performed by Seqhealth Tech (Wuhan, China). RIP assays were carried out on Ishikawa cells. The cells were lysed, and the lysis samples for immunoprecipitation reactions were incubated with anti-IGF2BP3 antibody (ab177477, Abcam, USA) or rabbit IgG (Cell Signaling Technology). The library products were enriched, quantified and finally sequenced on the Illumina PE150 platform.One hundred ninety-one candidates as IGF2BP3-interacting lncRNAs were identified in the RIP-seq results.

IGF2BP3-mediated m6A modification of RASGRF1 promoting joint injury in rheumatoid arthritis [RIP-seq]

Project description:With the deepening of epigenetic research, more and more studies have shown that N6- methyladenosine (m6A) is closely related to the occurrence and development of rheumatoid arthritis (RA), but the specific mechanism still needs to be further elucidated. In the study, we collected synovial tissues from normal controls and patients with OA or RA. The levels of m6A and inflammation were analyzed by immunofluorescence and western blot. The role of IGF2BP3 in cell proliferation and inflammatory activation was explored using transfection and RNA immunoprecipitation assays. IGF2BP3-/- mice were established, which were used to construct a mouse model of arthritis by transferring serum from adult arthritis K/BxN mice. Histological analysis and micro-CT were performed to determine joint damage. m6A level was markedly increased in RA patients and mouse models, and the expression of IGF2BP3 was upregulated in RA and related to inflammatory markers. IGF2BP3 played an important part in RA-fibroblast-like synoviocytes (FLS) by promoting cell proliferation, migration, invasion, inflammatory cytokine release, and inhibiting autophagy. In addition, IGF2BP3 inhibited autophagy to reduce the production of ROS, thereby decreasing inflammatory activation in macrophages. More importantly, RASGRF1-mediated mTORC1 activation played a crucial role in the ability of IGF2BP3 to promote cell proliferation and inflammatory activation. Based on the arthritis model of IGF2BP3-/- mice, IGF2BP3 knockdown Inhibited RA-FLS proliferation and inflammatory infiltration, and further improved RA joint injury. Our study revealed the important role of IGF2BP3 in RA progression. The targeted inhibition of IGF2BP3 reduced cell proliferation and inflammatory activation and limited RA development in a mouse model of arthritis, providing a potential strategy for RA therapy.

Here we report the post-transcript regulation of IGF2BP3, which belongs to the insulin-like growth factor 2 mRNA binding protein family. We used iRIP-seq and RNA-seq to analyze the transcript regulation and alternative splicing on IGF2BP3 treated with overexpression cells and control. Overexpressed IGF2BP3 has broadly increased genes expression which involved in G-protein coupled receptor signaling pathway, positive regulation of cell proliferation and signal transduction. IGF2BP3 regulated alternative splicing of multiple genes mainly clustered at response to hypoxia, negative regulation of transcription, and embryonic development. This study first provides alternative splicing analysis on transcription level of IGF2BP3 regulation, which laid the foundation for later research on IGF2BP3 critical functions
