Project description:Skeletal muscle diseases often exhibit fiber type-specific characteristics and posing significant clinical challenges, such as Duchenne muscular dystrophy (DMD), sarcopenia, and volumetric muscle loss injuries. Current treatments are primarily palliative, highlighting the need for innovative therapies. The extracellular matrix (ECM) plays a pivotal role in muscle physiology and regeneration by influencing cell differentiation and repair. Understanding the mechanism of specific ECM coatings affect muscle fiber type differentiation in vitro could elicit the targeted therapies for muscle regeneration. In this study, C2C12GFP myoblasts were differentiated into myotubes on the plates coated with Fibronectin, Collagen I, and Geltrex™. The myoblasts differentiated successfully on all ECM substrates, forming myofiber. qPCR confirmed myogenic marker expression patterns, with decreased Pax7 and increased Myog by day 7. Protein analysis (Western blot and immunofluorescence) and transcriptome profiling (RNA-Sequencing) consistently demonstrated that Collagen I enhanced the development of slow-type fibers, evidenced by increased slow myotube protein expression and the upregulation of slow fiber-associated genes (e.g., Myh7, Myl2), potentially via Calcineurin/NFAT, MEF2, MYOD, AMPK, PI3K/AKT and ERK1 pathways. In contrast, Fibronectin and Geltrex™ favored fast-type fiber development, as shown by elevated fast-type fiber protein levels and upregulation of fast fiber-associated genes (e.g., Fyn, Hk2), possibly through HIF1A, FOXO1, NFKB and ERK2 activation.

Project description:Analysis of C2C12 myoblast induced with tetracycline to enhance integrin alpha7 expression. Integrin alpha7 is the major laminin binding integrin in muscle cells. Enhancing its expression has been demonstrated to alleviate pathology in a murine model of Duchenne muscular dystrophy. Results of this study provide insights into the effects of increasing integirn alpha7 expression on muscle cells and possible side effects associate with enhancing integrin alpha7 in muscle cells. Project description:The purpose of this study is to investigate how GDF10 regulates early myoblast differentiation and fusion. We found that melatonin can enhance fibro-adipogenic progenitors (FAPs) paracrine signaling to support muscle regeneration. Through combining transcriptome analysis and single-cell transcriptome analysis, we identified GDF10 as a type of myogenic factor specifically secreted by FAPs, which plays a beneficial role in promoting muscle fiber development and regeneration. To determine the underlying mechanism by which GDF10 regulates myogenesis, we performed RNA-seq on myoblasts treated with GDF10 or vehicle for 6 hours in vitro. We found that GDF10 promoted myoblast fusion by regulating LKB1-AMPK-MYOG-MYMK signaling during differentiation. In summary, our study revealed novel insights into the mechanism by which melatonin promotes muscle regeneration by mediating the interplay between FAPs and muscle stem cells (MuSCs).

Project description:The development of an antithrombotic surface that prevents coagulation activation without interfering with hemostasis is the “holy grail” for blood-contacting devices. Such an advance would restrain device-induced thrombogenesis, and decrease the need for anticoagulant use in conjunction with devices, which would in turn reduce the bleeding risk. Herein, by optimizing factor (F)XII-surface binding rather than preventing its surface adsorption, we developed a substrate-independent antithrombotic coating with sheltered positive charges, the function of which was verified in vitro in human blood and using a carotid artery-jugular vein shunt model in rabbits. The new coating exhibits a strong interaction with FXII but results in a low reciprocal activation of the contact pathway. These findings contradict the prevailing strategy of designing antithrombotic materials through protein-repelling surfaces. Besides providing a universal coating which can benefit most blood-contacting devices, this work also pioneers a simple, but powerful and counterintuitive, guideline for designing antithrombotic surfaces.

Project description:Cells dynamically change their internal organization via continuous cell state transitions to mediate a plethora of physiological processes. Understanding such continuous processes is severely limited due to a lack of tools to measure the holistic physiological state of single cells undergoing a transition. We combined live-cell imaging and machine learning to quantitatively monitor skeletal muscle precursor cell (myoblast) differentiation during multinucleated muscle fiber formation. Our machine learning model predicted the continuous differentiation state of single primary murine myoblasts over time and revealed that inhibiting ERK1/2 leads to a gradual transition from an undifferentiated to a terminally differentiated state 7.5-14.5 hours post inhibition. Myoblast fusion occurred ~3 hours after predicted terminal differentiation. Moreover, we showed that our model could predict that cells have reached terminal differentiation under conditions where fusion was stalled, demonstrating potential applications in screening. This method can be adapted to other biological processes to reveal connections between the dynamic single-cell state and virtually any other functional readout.
