Artificial reforestation has less diverse soil nitrogen-cycling genes than natural restoration

Project description:Reforestation is effective in restoring ecosystem functions and enhancing ecosystem services of degraded land. The three most commonly employed reforestation methods of natural reforestation, artificial reforestation with native Masson pine (Pinus massoniana Lamb.), and introduced slash pine (Pinus elliottii Engelm.) plantations were equally successful in biomass yield in southern China. However, it is not known if soil ecosystem functions, such as nitrogen (N) cycling, are also successfully restored. Here, we employed a functional microarray to illustrate soil N cycling. The composition and interactions of N-cycling genes in soils varied significantly with reforestation method. Natural reforestation had more superior organization of N-cycling genes, and higher functional potential (abundance of ammonification, denitrification, assimilatory, and dissimilatory nitrate reduction to ammonium genes) in soils, providing molecular insight into the effects of reforestation.

Expression data from Northeastern Indigenous and Large White longissimus dorsi

Project description:To understand the molecular basis of distinct pork quality in Chinese indigenous and Western breed, longissimus dorsi samples were collected from three adult Northeastern Indigenous and from three adult Large White. Total RNA was extracted and subjected to porcine Affymetrix Genechip. The study helps to elucidate the genetic mechnism of divergent pork quality and provide the theory basis for selection and genetic improvement of meat quality traits in porcine. Six longissimus dorsi samples were collected from three Northeastern Indigenous and from three Large White. Three Large White were control samples. Total RNA was extracted from each sample.Gene-expression profiling was performed for each RNA sample separately on the GeneChipÂ® Porcine Genome Array at CapitalBio Corporation (Beijing, China).

Young differentiating xylem was harvested on the tension wood side of 6 month-old poplar trees grown in greenhouse conditions. The samples were harvested at 6 hour intervals throughout the day. High productivity in hybrid-poplar trees without isoprene emission to the atmosphere

Project description:The use of hybrid-poplar tree plantations as a source for biofuels and biomass production in temperate regions of the Northern Hemisphere has also, unintentionally, increased forest isoprene emissions to the atmosphere. The consequences of increased isoprene emissions include higher rates of tropospheric ozone production and increases in atmospheric aerosol production. Using RNA interference (RNAi) to suppress isoprene emission in several gene insertion events of hybrid-poplars, we show that this trait, which has been assumed as a requisite for the tolerance of abiotic stress, is not required for high rates of woody biomass production, even in extremely hot and dry climates. Biomass production over four years in experimental poplar plantations in Arizona and Oregon was similar among genetic lines that emitted or did not emit significant amounts of isoprene. Lines that had substantially reduced isoprene emission rates also showed decreases in flavonol pigments, which reduce oxidative damage during extremes of abiotic stress; a pattern that would be expected to amplify metabolic dysfunction during abiotic stress. Compensatory increases in the expression of other proteomic components, however, such as those that disable superoxide and other free radicals, and the fact that most biomass is produced during the spring, prior to the hottest and driest part of the growing season, explains the apparent paradox of high biomass production with low isoprene emission. The results of this study provide optimism for designing agroforest plantations of the future that provide high rates of lignocellulose production while eliminating detrimental effects of isoprene emission on atmospheric quality.

Growth-limiting drought stress induces time-of-day dependent transcriptome and physiological responses in hybrid poplar

Project description:Episodic drought stress negatively impacts the health of long-lived trees. Understanding the genetic and molecular mechanisms that underpin response to drought stress is requisite for selecting or enhancing climate change resilience. Here we aim to establish standardized drought stress protocols for transcriptome studies in poplar trees, to determine how hybrid poplars respond to prolonged and uniform exposure to drought; to determine if the responses to moderate and more severe growth-limiting drought stresses were qualitatively or quantitatively different; and, to determine how response to drought changes throughout the day. We established hybrid poplar trees (Populus x ’Okanese’) from unrooted stem cutting with abundant soil moisture for six weeks. We then withheld water to establish three soil water contents reflecting well-watered, moderate, and severe growth-limiting drought conditions. Plants were rewatered as needed for three weeks to maintain the soil water conditions. The mild and severe drought treatments elicited distinct changes in growth and development, photosynthetic rates and global transcriptomic changes. Notably, the time of day of sampling was strongest signal in the transcriptome data and it quantitatively and qualitatively affected drought responsive changes in gene expression. These analyses emphasize the complex nature of drought regulation in long-lived trees.

Project description:Anthropogenic nitrogen (N) deposition may affect soil organic carbon (SOC) decomposition, thus affecting the global terrestrial carbon (C) cycle. However, it remains unclear how the level of N deposition affects SOC decomposition by regulating microbial community composition and function, especially C-cycling functional genes structure. We investigated the effects of short-term N addition on soil microbial C-cycling functional gene composition, SOC-degrading enzyme activities, and CO2 emission in a 5-year field experiment established in an artificial Pinus tabulaeformis forest on the Loess Plateau, China.
