Project description:This study aimed to elucidate the mechanisms underlying the anti-diabetic effects of Mathurameha against high glucose-induced endothelial dysfunction by SWATH-MS. The EA.hy926 cells were treated with normal glucose, high glucose, or high glucose plus Mathurameha for 24 h. Proteins were extracted and resolved on 12% SDS-PAGE. Coomassie blue-stained gel were excised in the whole lane and subjected to in-gel tryptic digestion and SWATH-MS.

Transcriptional profiling of mouse cavenous endothelial cells in normal and high glucose condition

Project description:Purpose: To investigate the potential target genes associated with diabetic condition in mouse cavernous endothelial cells (MCECs) for diabetic induced erectile dysfunction (ED) treatment. Materials and Methods: Mouse cavernous tissue was embedded into Matrigel and sprouted cells were subcultivated for other studies. To mimic diabetic condition, the MCECs were exposed to normal-glucose (NG, 5 mmoL) and high-glucose (HG, 30 mmoL) condition for 72 hours. RNA-sequencing assay was performed to evaluate gene expression profiling and RT-PCR was used for sequencing data validation.

Project description:Here, the effects of high glucose levels on influenza severity were investigated using an in vitro model of the pulmonary epithelial-endothelial barrier. We show that compared to low glucose levels, high glucose conditions prior to influenza A virus infection increased virus-induced barrier damage. Increased barrier damage was not associated with increased cell death nor increased virus replication, but rather an increased pro-inflammatory response in endothelial cells and the subsequent damage of the epithelial junctional complex.

RNA-seq of young and senescent human umbilical vein endothelial cells (HUVECs) under normal or high glucose conditions

Project description:Human endothelial cellular models are useful to disentangle the pathophysiological role of dysfunctional endothelium in the development of cardiovascular (CV) disease and organ damage in T2D. Here, we performed a RNAseq of human umbilical vein endothelial cells (HUVECs) undergoing replicative senescence and exposed to high glucose (25 mM) to investigate the combined effects of replicative senescence and high glucose on the transcriptional landscape of these cells. To gain insight into the molecular mechanisms driving the acquisition of a senescent phenotype following exposure to HG, we performed a RNA-seq assay on control (Ctr) and replicative senescent (Sen) HUVECs cultivated in presence/absence of HG culture medium (total number of samples = 12; number of samples in each cell type-medium condition group = 3) using the NovaSeq 6000 Illumina system. Differential expression analysis was performed in R environment (version 4.2.2) using the limma and edgeR Bioconductor R packages.
