Project description:Neonatal hypoxic-ischemic brain damage (HIBD) is initiated by perinatal asphyxia, leading to brain injury triggered by reduced blood and oxygen flow, resulting in neurological impairments such as cerebral palsy, epilepsy, cognitive deficits, and behavioral disorders.Recent insights collectively suggest the important roles of lysyl oxidase (LOX) in the pathological processes of several acute and chronic neurological diseases, but the molecular regulatory mechanisms in HIBD remain elusive.In this study, we found through the study of HIBD rat models and the oxygen-glucose deprivation/re-oxygenation (OGD/R) cell models that LOX significantly increased after HIBD or OGD/R. RNA seq results showed a significant increase in piezo1 after primary neuron OGD/R, and iron death data was also enriched. Moreover, inhibiting LOX or piezo1 can rescue neuronal ferroptosis and further improve cognitive function in rats. In addition, we also found that traumatic acid can inhibit the enzyme activity of LOX and improve a series of pathological features of neuronal damage caused by increased LOX.

Project description:The ovarian follicle is devoid of direct blood supply because of a blood follicle barrier. The intra follicular compartments are hypothesized to have low oxygen level. As the diameter of an ovarian follicle substantially increases during follicular development, the oxygen concentration is likely to decrease in the follicular fluid and reaches lowest levels in the ovulatory follicles. During the present investigation, transcriptome differences induced by low oxygen levels were comprehensively analyzed to understand the specific significance of low oxygen levels in ovulatory follicles.

Oxygen concentration affects de novo DNA methylation and transcription in in vitro cultured oocytes

Project description:We optimised an in vitro culture model for mouse oocytes starting from immature oocytes to get mature oocytes and investigated the effect of oxygen concentrations on the cultured oocytes (5 % vs 20% oxygen). The cultured oocytes were size-selected in both conditions. We generated expression and methylation profiling (RNA-Seq, RRBS, PBAT) by high throughput sequencing from these size selected oocytes and compared the results with in vivo size oocytes. Our observations reveal changes in DNA methylation and transcripts between oocytes cultured in vitro with different oxygen concentrations and in vivo grown murine oocytes. Oocytes grown under 20% O2 had a higher correlation with in vivo oocytes for DNA methylation and transcription demonstrating that higher oxygen concentration is beneficial for the oocyte maturation in ex-vivo culture condition.
