Project description:Adipocyte-secreted factors (ASFs) induce lipid accumulation in breast cancer cells. These lipid droplets are thought to serve a cell-protective, pro-oncogenic role. ASFs including insulin, CCL5, and apelin interact with receptors that stimulate mTOR signaling in neighboring cells. One of the major transcription factors that acts downstream of this signaling pathway is SREBP1, which translocates into the nucleus and activates the expression of metabolic enzymes and other proteins involved in fatty acid synthesis, transport, and storage. Their associated enzymatic activities increase or decrease pools of metabolites that serve as substrates and cofactors for chromatin remodeling, such as acetyl-CoA, alpha-ketoglutarate (α-KG), flavin adenine dinucleotide (FAD+), and nicotinamide adenine dinucleotide (NAD+), but the impact on gene regulation in this context is not completely understood. To model biochemical signaling from adipocytes to breast cancer epithelial cells, we cultured differentiated mouse OP-9 cells, collected the adipocyte conditioned medium (ACM) after three days, and treated human breast epithelial BT-549 cells with ACM. Three days after treatment, we observed accumulation of lipid droplets in the cytoplasm of most BT-549 cells, whereas none were visible in cells grown in unconditioned medium (UCM). Transcription profiling verified the activation of lipid droplet biosynthesis genes, and revealed repression of loci at every chromosome, consistent with the idea that lipogenesis affects the availability of substrates and cofactors for global chromatin remodeling.

Transcriptomic profiling of adipocytes in response to cold reveals unique properties of Cxcl12 as a brown adipocyte secreted chemokine

Project description:Beyond its thermogenic potential, brown adipose tissue (BAT) performs important endocrine functions that regulate metabolism. However, the BAT microenvironment and the factors involved in BAT homeostasis and adaptation to cold remain poorly characterized. We therefore aimed to study secreted factors from active brown adipocytes that may be involved in adipocyte function and/or may orchestrate inter-cellular communications. For this, mRNA levels in mature adipocytes of brown, beige and white adipose depots from mice exposed to 21 days of cold were evaluated using RNA sequencing, and bioinformatic analysis was used to predict for potentially secreted factors. Cxcl12 was found to be the most cold-induced C-X-C chemokine in BAT, and Cxcl12 mRNA expression analysis by qPCR and fluorescence in-situ hybridization revealed its enrichment in brown adipocytes upon cold. Cold increased CXCL12 secretion from BAT, yet its level in plasma remained unchanged indicating a potential local action. Cxcl12 knockdown in mature brown adipocytes impaired thermogenesis, estimated by norepinephrine (NE)-induced Ucp1 gene expression, glycerol release and mitochondrial respiration despite unaltered β-adrenergic signaling, suggesting Cxcl12 regulates adipocyte function independently from the β-adrenergic pathway. Importantly, unaltered adipocyte characteristics upon Cxcl12 loss may indicate CXCL12 primarily regulates the NE-induced adipocyte activation. Furthermore, CXCL12 might exert inter-cellular crosstalk via its capacity to promote macrophage chemotaxis and neurite outgrowth. Here we present CXCL12 as a novel brown adipocyte, cold-induced secreted factor involved in adipocyte function and inter-cellular crosstalk within BAT.

Insulin-Independent Regulation of Type 1 Diabetes via Large Brown Adipocyte-Secreted Proteins and the Novel Glucagon Regulator Nidogen-2

Project description:Current treatments for type 1 diabetes (T1D) focus on optimizing insulin replacement. We demonstrate the therapeutic potential of the large secreted protein fraction from brown adipose tissue (BAT), independent of insulin. This secreted fraction mediates insulin receptor-dependent recovery of euglycemia in diabetic nonobese diabetic (NOD) mice by suppressing glucagon secretion. It also promotes white adipocyte differentiation and browning, and enhances glucose uptake in adipose tissue, skeletal muscle, and liver. From this fraction, we identify nidogen-2 as a brown adipocyte-secreted factor that reverses hyperglycemia in T1D NOD, inhibits glucagon secretion from pancreatic α-cells, and mimics other actions of the secreted fraction. These findings provide proof of principle that the pleiotropic effects of BAT-derived peptides represent a novel approach to diabetes management.

Effect of Large Brown Adipocyte-secreted Proteins on Type 1 Diabetic Mice (NOD) inguinal white adipose tissue

Project description:Brown adipose tissue has been recognized as an important endocrine organ for metabolic health. In our study, we found that large proteins secreted by brown adipocyte (CB-100) promote the recovery of euglycemia in type 1 diabetic mice (NOD) when injected at an early stage of the disease. To further elucidate the in vivo effects of CB-100, we analyzed brown adipose tissue from both CB-100 and sham-treated mice. Differential gene expression analysis revealed that CB-100 treatment promote the adipocyte proliferation and browning wihtin inguinal white adipose tissue. This suggests that the recovery of euglycemia in CB-100-treated mice may be attributed to the recovery of healty white fat and its browning, which also contributes to glucose clearance

MDA-MB-231 cells were treated with medium derived from adipocytes culture supernatants. Using two different RNA libraries, we sequenced the complete transcriptome(including mRNA, lncRNA, circleRNA and small RNA) of MDA-MB-231 treated with medium derived from adipocytes culture supernatants.

Transcriptomic analysis of osteoblastic cells derived from human mesenchymal stem cells, incubated with secreted factors release by adipocytes against non-incubated controls

Project description:Our preliminary findings lead us to propose bone marrow adipocyte secretions as new actors of bone loss. Indeed, using a coculture model based on human bone marrow stromal cells, we previously showed that soluble factors secreted by adipocytes induced the conversion of osteoblasts towards an adipocyte-like phenotype. To further decipher the molecular changes triggered by the coculture, we performed transcriptional analyses using Agilent 60-mer Sure Print G3 Human Gene Expression Microarray Kit containing a 50, 599 probes. Transcriptional changes were monitored in 7 biological replicates each consisting of osteoblasts grown alone (OB) and cocultured osteoblasts (OB-CC) upon 9h or 48h and adipocytes (AD) at day 14 of differentiation placed in serum-free medium for 9h or 48h.
