Vaccination Against MSI Colorectal Cancer

Project description:Patients with advanced microsatellite unstable (MSI-H) colorectal cancer will be vaccinated with three so called frame shift peptides (FSPs), AIM2(-1), HT001(-1) and TAF1B(-1) combined with Montanide ISA-51 VG. By this, an immune response directed against MSI-induced FSPs that are shared by the majority of MSI-H colorectal cancers can be induced. The aim is to show that vaccination against MSI-induced FSPs is safe and can induce or enhance immune responses against MSI-H colorectal cancer-associated antigens.

Project description:Mechanisms of poor responses to vaccines remain unknown. Hepatitis B virus-naïve elderly subjects received three vaccines, including a vaccine against hepatitis B virus (HBV). Transcriptomic profilling of blood collected pre-vaccination and post-vaccination was performed in order to identify candidate biomarkers of antibody response to the different vaccines.

Project description:Tumors display oncofetal chondroitin sulfate (ofCS) glycosaminoglycans that support disease progression and dissemination. ofCS can be detected by the the Plasmodium falciparum derived VAR2CSA protein but the development of recombinant VAR2CSA as a clinical tool has proven challenging. Here, we performed AP-MS experiments with newly developed antibodies with ofCS specificity comparable to VAR2CSA, as well as recombinant VAR2CSA bait.

Affymetrix SNP array data for chicken responses to glycoconjugate vaccination against Campylobacter jejuni

Project description:Campylobacter jejuni is a common cause of diarrheal disease worldwide. Human infection typically occurs through the ingestion of contaminated poultry products. We previously demonstrated that an attenuated Escherichia coli live vaccine strain expressing the C. jejuni N-glycan on its surface reduces the Campylobacter load in more than 50% of vaccinated leghorn and broiler birds to undetectable levels (responder birds), whereas the remainder of the animals were still colonized (non-responders). To understand the underlying mechanism, we conducted 3 larger scale vaccination and challenge studies using 135 broiler birds and found a similar responder/non responder effect. The submitted data were used for a genome-wide association study of the chicken responses to glycoconjugate vaccination against Campylobacter jejuni.
