Project description:Understanding mammalian preimplantation development, particularly in humans, at the proteomic level remains limited. Here, we applied our comprehensive solution of ultrasensitive proteomic technology to measure the proteomic profiles of oocytes and early embryos and identified nearly 8,000 proteins in humans and over 6,300 proteins in mice. We observed distinct proteomic dynamics before and around zygotic genome activation (ZGA) between the two species. Integrative analysis with translatomic data revealed extensive divergence between translation activation and protein accumulation. Multi-omic analysis indicated that ZGA transcripts often contribute to protein accumulation in blastocysts. Using mouse embryos, we identified several transcriptional regulators critical for early development, thereby linking ZGA to the first lineage specification. Furthermore, single-embryo proteomics of poor-quality embryos from over 100 patient couples provided insights into preimplantation development failure. Our study may contribute to reshaping the framework of mammalian preimplantation development and opening avenues for addressing human infertility.

Project description:Lactate is present at a high level in the microenvironment of mammalian preimplantation embryos in vivo and in vitro. However, its role in preimplantation development is unclear. Here, we report that lactate is highly enriched in the nuclei of early embryos when major zygotic genome activation (ZGA) occurs in humans and mice. The inhibition of its production and uptake results in developmental arrest at the 2-cell stage, major ZGA failure, and loss of lactate-derived H3K18lac, and the abnormal phenotypes could be recapitulated by overexpression of H3K18R mutation and rescued by addition of Lac-CoA. By profiling the landscape of H3K18lac in human and mouse preimplantation embryos, we show that H3K18lac is enriched on the promoter regions of most major ZGA genes and corelates with their expressions. Taken together, we demonstrate the important role for lactate in major ZGA via H3K18lac, showing a conserved metabolic mechanism underlies preimplantation development of mammalian embryos.

Project description:Proteins play a crucial role in the central dogma as the end product of gene expression. However, a deep proteomic landscape is still the missing puzzle piece of the comprehensive understanding of mammalian preimplantation development. Here, using our recently developed Comprehensive Solution of Ultrasensitive Proteome Technology (CS-UPT), we measured the proteomic profiles of oocytes and early embryos for ~5000 proteins in humans and over 6200 proteins in mice. Comparative analysis identified divergent proteomic dynamics before and around zygotic genome activation (ZGA) between the two species. Integrative analysis with transcriptome and translatome data revealed extensive and stage-specific protein turnover events. Remarkably, our comprehensive multi-omics analysis indicates that ZGA transcripts significantly contribute to protein accumulation at the blastocyst stage. Additionally, we experimentally confirmed several pf these key regulators are essential for early mouse development. Our data provide a rich resource for mammalian preimplantation development and laid a solid foundation for future investigations.

Project description:Translational regulation plays a critical role during oocyte and preimplantation embryo development. Here, we utilized high resolution ribosome fractionation and low-input ribosome profiling of mouse oocytes and preimplantation embryos to explore the translational landscapes of early embryo development. The inconsistency was observed when comparing polysome-bound, monosome-bound, and free RNA profiles, indicating the role of ribosomes in manipulating RNA fate, especially for oocyte-to-embryo transition (OET) and zygotic genome activation (ZGA). Moreover, eif1ad3 with its homologs, including eif1ad4, eif1ad6, eif1ad7, etc. are highly translated during ZGA and regulating the translational activity of other genes. Knocking down these genes lead to defective ZGA and preimplantation development. These data reveal that the transcriptome and translatome manipulate RNA fate collectively to regulate mouse preimplantation development.

Project description:The increasing prevalence of nanoplastics (NPs) in the environment has raised concerns regarding their potential risks to ecosystems and human health. NP exposure has been shown to interfere with cellular processes; however, their effects on preimplantation embryonic development remain unclear. Given the critical role of zygotic genome activation and early cell cycle progression, disruption during this period can significantly affect embryonic viability. To investigate the molecular effects of polymethylmethacrylate (PMMA)-NPs on early embryonic development, mouse zygotes were exposed to 0.1 or 1 mg/mL PMMA-NPs, and RNA sequencing was performed on four-cell stage embryos. Differential gene expression analysis revealed that 0.1 mg/mL NP exposure affected genes related to DNA damage repair, transcriptional regulation, and cellular homeostasis while 1 mg/mL NP exposure altered the expression of genes associated with apoptotic signaling, cell cycle arrest, and failure of blastocyst formation, suggesting a concentration-dependent effect on developmental progression. Thus, PMMA-NP exposure disrupts early embryonic development by inducing specific transcriptomic changes that potentially affect genome integrity and developmental competency. This study provides new insights into the molecular mechanisms underlying NP-induced embryo toxicity and elucidate the effect of NPs on preimplantation embryo development.
