Project description:Micro and nanoplastics (MNPs) are widespread contaminants of our environment and food web that can be absorbed by the intestine and distributed to most or all organs, and may cause significant toxicity and dysfunction. In our recent study, we investigated the toxicity and mechanisms underlying uptake and translocation of 26 nm carboxylated polystyrene MNPs (PS26C) using a combination of simulated digestion and a static, immortalized cell-based, three cell type (triculture) small intestinal epithelial model. Results revealed that PS26 caused minimal toxicity, and inhibitor studies revealed that uptake mechanisms included passive diffusion, phagocytosis, clathrin-mediated endocytosis (CME), and fast endophilin-mediated endocytosis (FEME). However, the triculture model is somewhat lacking in physiological relevance, and studies in a more relevant and accurate model are required to validate and expand upon those results. In the current study, we therefore employed a microfluidic intestine-on-a-chip model developed using primary human intestinal epithelial organoids from both healthy and Crohn’s disease donors, and primary human microvascular endothelial cells to evaluate the toxicological impacts and mechanisms effectuating uptake and translocation of 25 nm polystyrene shell-gold core tracer (quantifiable by ICP-MS) MNPs (AuPS25). As in our previous study, AuPS25 caused minimal toxicity after 24 h exposure in either the healthy or Crohn’s IOC model. RNAseq analysis of epithelial cell lysates identified only 9 dysregulated genes, which notably included downregulation of IFI6 (interferon alpha-induced protein 6), which has antiviral and negative innate immune regulatory functions in the intestine. Inhibitor studies revealed that AuPS25 uptake occurred by both passive and active mechanisms, including phagocytosis and/or macropinocytosis, and CME and/or FEME. Interestingly, uptake and translocation of AuPS25 was greater in healthy than in unstimulated Crohn’s disease epithelium.

Project description:Nanoplastics are produced by breakdown of plastics in environmental contamination or commercial use for cosmetics or daily expenses. Emerging evidence indicate that ingested nanoplastics with a size smaller than 100 nm have the potential to reach the brain and induces neurotoxicity. However, the potential toxicity of nanoplastics on brain are limited because of difficulties in synthesize of nanoplastics. In present study, we synthesized the fluorescent polystyrene nanoplastics (PSNPs) and examined the toxicity of PSNPs in brain in vivo and in vitro analyses by comparison to IR-813 fluorophore. Synthesized PSNPs were characterized by fluorescence imaging system, scanning electron microscopy, and Fourier-transform infrared spectroscopy. PSNPs were detected in adult mice brain by oral ingestion. In addition, a series of behavioral analyses showed that oral ingestion of PSNPs induced memory impairments. Among brain cells, PSNPs were predominantly internalized in microglia, and uptake of PSNPs induced microglial activation. In addition, the conditioned medium derived from microglia exposed to PSNPs repressed hippocampal neuronal activity. Furthermore, transcriptome analysis showed that PSNPs changed gene expressions in microglia, elevation of neuroinflammation in contrast to suppression of neurotrophic factors. These results indicated that predominant uptake of PSNPs in microglia induced elevation of neuroninflammatory responses whereas suppression of neurotrophic factors that may contribute to the cognitive impairment. Our findings indicate the toxic mechanism and potential detrimental effect of nanoplastic in brain and suggest a potential risk of cognitive impairment by exposure to nanoplastics.

Project description:<p>The uptake and accumulation of nanoplastics by plants have raised particular concerns. However, molecular mechanisms underlying nanoplastic phytotoxicity are still vague and insufficient. Thus, its potential environmental risks and biosafety are receiving increased attention. In this study, cucumber plants were exposed to polystyrene nanoplastics (PSNPs) under different concentration. The results demonstrated that phenotypic parameters (total photosynthetic pigment content, relative membrane permeability, total phenolic content, and lipid peroxidation) were not significantly affected. However, metabolomic and transcriptomic analysis revealed that both doses of PSNPs induced the expression regulation of different key genes, exhibiting considerable distinction on metabolic responses in cucumber. The high dose of PSNPs induced stronger alteration in cucumber leaves than low dose of PSNPs. Low dose exposure significantly affected four metabolic pathways involved in the carbohydrate metabolism. High dose exposure significantly affected five metabolic pathways involved in the carbohydrate metabolism and four metabolic pathways involved in the amino acid metabolism. These findings shed new light on the phytotoxic mechanism and environmental implication behind the interaction of nanoplastics and plants, and advance our understanding of the potential adverse effect induced by the presence of nanoplastics in agricultural systems.</p>

Project description:Bariatric surgeries remain the most effective treatments for sustained weight loss and remission of type 2 diabetes. In addition to decreased body weight and improved glucose regulation, these procedures also dramatically increase secretion of several gut hormones including GLP-1. Despite these benefits, there are deleterious side effects to these procedures that include an increased incidence of iron-deficiency related anemias. The transcription factor HIF2a is heavily expressed in the duodenum and regulates the molecular machinery of iron absorption from the lumen. Lower iron levels after both gastric bypass and sleeve gastrectomy procedures occur despite dramatic upregulation of HIF2a signaling in the duodenum and are likely the result of increased hepcidin levels. Low iron diets also stimulate increased HIF2a signaling in the duodenum and produce effects similar to bariatric surgery that include reduced body weight/fat, improved glucose regulation and increased secretion of the GLP-1. Gut-specific deletion of VHL results in a constitutive upregulation of HIF2a signaling in the small intestine and produces a dramatic phenotype that includes reduced body weight/fat, improved glucose tolerance and increased GLP-1 secretion from the intestine. These data demonstrate an important role of HIF2a signaling in the duodenum to regulate multiple aspects of systemic metabolism and gut hormone secretion pointing towards critical cross-talk between the systems that regulate iron and other aspects of systemic physiology important to prevalent metabolic diseases.

Project description:Nanoplastics pollution is a rising environmental concern whose impacts on biodiversity and human health are far from being understood. This is particularly salient in aquatic ecosystems, where the majority of species depend on external fertilization for reproduction. Here we evaluated the effects of a short-term exposure to engineered polystyrene nanoplastics (NPs) in the zebrafish germ line to further explore their impact on reproduction. To this end, zebrafish (Danio rerio) were exposed to 5 mg/L of 45 nm polystyrene (PS)-NPs via water for 96h. We show that, in males, nanoplastics induced testicular histological alterations with abnormal sperm clustering and chromatin compaction, resulting in viable spermatozoa but with reduced motility. Moreover, in females we observed an alteration in oocyte stages frequencies during oogenesis, possibly reflecting alterations in oocyte growth. RNA-sequencing analysis in male testis links nanoplastic induced alterations in the expression of genes involved in chromatin structure, meiosis and DNA double-strand break formation and repair progression, and gametes recognition. Flow cytometry analysis revealed that the observed effects in males were directly due to nanoplastics penetrating the testicular barrier and being internalized within germline cells. Overall, our results demonstrate that acute exposure to NPs can compromise reproductive fitness, underscoring the environmental and health impacts of NPs pollution.
