Project description:Diverse types of GABAergic projection neurons and interneurons of the telencephalon derive from progenitors in a ventral germinal zone, called the ganglionic eminence. Using single-cell transcriptomics, chromatin accessibility profiling, lineage tracing, birthdating, heterochronic transplantation, and perturbation sequencing in mouse embryos, we investigated how progenitor competence influences the maturation and differentiation of these neurons. We found that the progression of neurogenesis over developmental time shapes maturation competence in ganglionic eminence progenitors, influencing how they progress into mature states. In contrast, differentiation competence, which defines the ability to produce diverse transcriptomic identities, remains largely unaffected by the stages of neurogenesis. Chromatin remodeling alongside a NFIB-driven regulatory gene module influences maturation competence in late-born neurons. These findings provide key insights into how transcriptional programs and chromatin accessibility govern neuronal maturation and the diversification of GABAergic neuron subtypes during neurodevelopment.

Project description:Inhibitory neurons of the telencephalon are generated from progenitors in the ganglionic eminences that mature and differentiate into specialized cell types. Here, we used scRNA-seq and scATAC-seq together with lineage tracing and birthdating techniques to investigate the influence of neurogenesis timing on the development of GABAergic precursors. We found that the timing of neurogenesis substantially influences the progenitors' maturation competence (the capacity to develop toward a fully functional state), but not their differentiation competence (the ability to evolve into transcriptomically diverse states). The underlying mechanism was chromatin priming, orchestrated by the transcription factor Nfib in collaboration with regulators of inhibitory neuron development. Finally, the heterochronic transplantation of progenitors revealed an interplay between both intrinsic and extrinsic cues acting upon maturation competence. These findings identify a mechanism that coordinates inhibitory neuron development by changing their maturation to achieve maximum adaptability.

Elucidating ADNP Syndrome Gene Dysregulation through a Novel Approach in iPSC-Derived GABAergic Neuron Development [ADNP_scRNA-seq]

Project description:GABAergic interneurons are essential for maintaining neural circuit function, and their dysfunction is linked to a range of neurodevelopmental and neurodegenerative disorders. Traditional approaches for differentiating human pluripotent stem cells (PSCs) into neuronal cells are often hindered by challenges such as incomplete neural differentiation, prolonged culture times, and variability across PSC lines. To overcome these limitations, we developed an innovative strategy by combining the overexpression of transcription factors Ascl1 and Dlx2 with dual-SMAD and WNT inhibition, effectively driving the differentiation of PSCs into diverse, region-specific GABAergic neuron subtypes. Through single-cell sequencing, we thoroughly characterized the heterogeneity of these induced neurons (iNs) and uncovered the regulatory mechanisms underlying their fate specification. Our findings demonstrate the successful generation of multiple disease-relevant GABAergic neuron types. Furthermore, we investigated the impact of the ADNP syndrome-associated p.Tyr719* variant on GABAergic neuron differentiation, revealing that this mutation significantly alters neuronal subtype composition and synaptic transmission. This study advances our understanding of the diversity of PSC-derived GABAergic neurons and underscores their potential as models for exploring neurological disorders.
