Project description:Rheumatoid arthritis (RA) and other inflammatory arthritides are chronic joint diseases marked by inflammation and bone erosion, leading to progressive damage. While smaller joints are typically affected early, larger joints often become involved over time. Temporomandibular joint (TMJ) involvement occurs in over 50% of arthritis patients, causing severe jaw pain and functional impairment, yet it remains underrecognized and underresearched. Due to its unique mechanical and biological features, the TMJ may exhibit distinct pathomechanisms compared to other joints. To investigate these differences, we analyzed transcriptomic profiles of the TMJ, ankle joint, and alveolar bone in polyarthritic hTNF-α transgenic mice (hTNF-tg) and controls.

Expression profiling of ankle joint of a mouse model of gouty arthritis uncovers potential mechanisms involved in pain and inflammation

Project description:Gout arthritis (GA) is a prevalent and painful inflammatory arthritis, and its global burden continues to rise. Intense pain induced by gout attacks is a major complication of gout. Treatment and long-term management for gout patients remain challenging. However, the pathogenesis of GA still remained largely unknown. The purpose of this study is to perform an expression profiling to identify genes related with GA in an animal model. Using a monosodium urate (MSU) crystals-induced GA model, we performed RNA-sequencing for the ankle joint of gout mice in different time points. Our results revealed transcriptional changes in the joint. We provided by far the first study to profile gene expression changes and pathway analysis of ankle joint in different time for investigating GA mechanisms. This work may provide novel insights into understanding the mysterious mechanisms underlying GA.

Project description:Adult mammalian synovial joints have limited regenerative capacity, where injuries heal with mechanically inferior fibrotic joint tissues. Here we developed a unilateral whole-joint resection model in adult zebrafish to advance our understanding of how to stimulate regrowth of native synovial joint tissues. Using single-cell RNA sequencing, we profile RNA expression from live-sorted jaw joint cells throughout the time course of joint regeneration (1 to 70 days-post-joint-resection (dpjr)). Our findings reveal latent molecular and cellular programs within the adult skeleton that are deployed to regenerate a complex joint with lubricated articular cartilage.

Project description:Rheumatoid arthritis is an autoimmune disease in which joint inflammation lead to progressive cartilage and bone destruction. Matrix metalloproteinases (MMP) implicated in homeostasis of extracellular matrix (ECM) play a central role in cartilage degradation. The aim of this study was to investigate the role of MMP-8 (collagenase-2) suppression in the K/BxN serum-transfer arthritis model. Three male mice of each following groups: MMP-8 wild type and arthritic mice, MMP-8 wild type without arthritis (wild type control), MMP-8 KO and arthritic mice and MMP-8 KO without arthritis (KO control) were selected for RNA extraction, from ankle joints, and hybridation on Affymetrix microarrays. Male mice were used because they showed a trend to higher arthritis severity compared to female mice. In arthritic mice, ankle joints were taken 7 days after arthritis induction.

Arthritis Flares Mediated by Tissue Resident Memory T Cells in the Joint

Project description:Rheumatoid arthritis is a systemic autoimmune disease, but disease flares typically affect only a subset of joints, distributed in a pattern that is distinctive for each patient. Pursuing this intriguing pattern, we show that arthritis recurrence is mediated by long-lived synovial resident memory T cells (TRM). In three murine models, CD8+ cells bearing TRM markers remain in previously inflamed joints during remission. These cells are bona fide TRM, exhibiting failure to migrate from joint to joint, preferential uptake of fatty acids, and long-term residency. Disease flares result from TRM activation by antigen, leading to CCL5-mediated recruitment of circulating effector cells. Correspondingly, TRM depletion ameliorates recurrence in a site-specific manner. Human rheumatoid arthritis joint tissues contain a comparable CD8+-predominant TRM population, most evident in late-stage leukocyte-poor synovium, exhibiting limited T cell receptor diversity and a pro-inflammatory transcriptomic signature. Together, these findings establish synovial TRM cells as a targetable mediator of disease chronicity in autoimmune arthritis.
