Project description:Dental pulp stem cells (DPSC) constitute a neural crest-derived stem cell population endowed with multipotency and self-renewal. While the process of DPSC differentiation has been studied extensively in vitro, very little is known about mechanisms underpinning the differentiation of human DPSC in vivo. Here, we induced vasculogenic, odontoblastic, or neurogenic differentiation of human DPSC for 7 days in vitro and performed single cell sequencing. Then, human DPSC tagged with GFP (DPSC-GFP) seeded in human tooth slice/scaffolds were transplanted into immunodeficient mice. Single cell sequencing of DPSC-GFP sorted by flow cytometry was performed 7 and 21 days after transplantation. We observed major shifts in patterns of gene expression when DPSC were induced to undergo vasculogenic, odontoblastic, or neurogenic differentiation in vitro. Although some DPSC retained mesenchymal stem cell (MSC) markers (indicating asymmetric cell division and self-renewal), each differentiation protocol resulted in a unique gene expression signature in vitro. In vivo, stem cell markers that were highly expressed in DPSC pre-transplantation gradually decreased in expression after 7 and 21 days. In contrast, vascular endothelial cell markers were highly expressed 7 days after transplantation, while neuronal markers were highly expressed 21 days after transplantation. In conclusion, DPSC cells are heterogeneous with clearly distinct cell clusters, all of which contain cells with unique differentiation potential. Notably, the microenvironment created when human DPSC are transplanted inside a human root canal in vivo induces vasculogenic differentiation first, which is then followed by neurogenic differentiation.

We previously reported the presence of vasculogenic mimicry in osteosarcoma patients. The mechanistic basis for osteosarcoma VM remains unclear. We used microarrays to detail the global programme of gene expression between 143B cells and HOS cells exposed to Matrigel which showed greatly different vasculogenic mimicry formation potential and identified distinct classes of vasculogenic mimicry-realted genes during this process.

Project description:Neurogenic astroglia-like cells of the prospective white matter (PWM) generate interneurons and astroglia during cerebellar development. However, the characterization of PWM subpopulations is still insufficient. Here we show that ACSA-2 (Astrocyte Cell Surface Antigen-2) is an intrinsic marker of astrocyte-committed precursors. In the developing cerebellum, the ontogeny of ACSA-2+ astrocytes revealed this epitope to be exclusively expressed by PWM cells. By contrast, in the adult brain, ACSA-2 is observed on parenchymal astrocytes, fibrous astrocytes of the white matter and at low level on Bergmann glia. In combination with GLAST, a marker for astroglia-like progenitors, we addressed the characteristics of neurogenic (ACSA-2-/GLAST+) and astrocyte-committed (ACSA-2+/GLAST+) precursors using transcriptomics and cell transplantation assays. Gene expression analyses identified major differences in genes related to the maturation status of astrocytes, their potential to generate interneurons and genes required for Bergmann glia specification. Transplantation assays further confirmed a divergent differentiation potential: ACSA-2+/GLAST+ cells, on the one side, differentiate into astrocytes and oligodendrocytes but not into Bergmann glia or neurons when transplanted into the neonatal cerebellum. On the other side, ACSA-2-/GLAST+ progenitors were able to generate interneurons and all the glial phenotypes. Moreover, ACSA-2-/GLAST+ progenitors even maintained the neurogenic potential when transplanted into the adult cerebellum. In conclusion, this work reports about ACSA-2, a marker for glial-restricted precursors of the PWM that in combination with GLAST enables for the discrimination and isolation of neurogenic and astrocyte-committed progenitors of the neonatal cerebellum.

Project description:Stem cell-based therapy is an alternative strategy for brain repair. Various cell types have been investigated and dental pulp stem cells (DPSC) have been identified as promising candidates. These multipotent stem cells are found in the dental pulp tissue of molar teeth. They are clinically easily obtained, have a high proliferative rate, and possess neurogenic potential due to their mesoectodermal origin. Here, overexpression of octamer-binding transcription factor 4 (OCT4) in combination with neural conditions was used to reprogram human DPSC along the neural lineage. Transcriptomic analysis of differentially-expressed genes highlighted the expression of genes associated with neural and neuronal functions in the OCT4-overexpressing DPSC following neural induction.

Project description:The mammalian heart contains multiple cell types that appear progressively during embryonic development. Advances in determining cardiac lineage diversification has often been limited by the unreliability of genetic tracers. Here we combine clonal analysis, genetic lineage tracing, tissue transplantation and mutant characterization to investigate the lineage relationships between epicardium, arterial mesothelial cells (AMCs) and the coronary vasculature. We report a contribution of the second heart field (SHF) to a vasculogenic niche composed of AMCs and sub-mesothelial cells at the base of the pulmonary artery. Sub-mesothelial cells from this niche differentiate into lymphatic endothelial cells and, in close association with AMC-derived cells, contribute to, and are essential for the development of ventral cardiac lymphatics. In addition, regionalized epicardial/mesothelial retinoic acid signaling regulates lymphangiogenesis, contributing to the niche properties. These results uncover a SHF vasculogenic contribution to coronary lymphatic development through a local niche at the base of the great arteries.

In vitro assays suggest that not all cells within these lines a VM-competent.
