Project description:Secretome-based therapies targeting angiogenesis are promising in ischemic heart disease (IHD). The effects of Chemokine C-C motif ligand (CCL) 28 on IHD remain unclear. This study investigated the role of CCL28 in angiogenesis in IHD. The myocardial infarction (MI) and hindlimb ischemia (HI) models were constructed on male wild-type and CCL28-knockout mice. AAV9-TIE1-sh-CCR10 induced endothelial specific CCR10 deficiency in mice by tail vein injection. Recombinant CCL28 protein was used to treat MI and HI in wild-type and db/db mice. Serum levels of CCL28 were evaluated in a cohort of 416 consecutive chronic total occlusions (CTO) patients. The upregulated CCL28/ C-C motif receptor (CCR)10 axis was observed in myocardial and hindlimb ischemia. Additionally, CCL28 was mainly secreted by macrophages and CCR10 was highly expressed in endothelial cells (ECs). Compared to CCR10- ECs, CCR10+ ECs exhibited robust proangiogenic capacity, which are induced by CCL28 through the CCR10/ERK/SOX5 positive feedback signaling. Deletion of CCL28 results in impaired angiogenesis in the ischemic models while the use of recombinant CCL28 protein has therapeutic potential for myocardial and hindlimb ischemia, including in diabetes. Endothelial specific CCR10 deficiency impairs angiogenesis and blocks the therapeutic effect of rCCL28 in the myocardial and hindlimb ischemia. Serum CCL28 levels exhibited the predictive effect on coronary collateral vessels (CCV) in CTO patients. CCR10+ endothelial cells (ECs) are characterized with the enhanced proangiogenic capacity and beneficial for angiogenesis. CCL28 promotes angiogenesis by increasing CCR10+ ECs via the CCR10/ERK/SOX5 positive feedback signaling. This study highlights the angiogenic role of CCL28 and recombinant CCL28 protein might be a potential therapeutic option to attenuate myocardial and hindlimb ischemia, including in diabetes.

Project description:Four major mucosal-associated chemokines, CCL25, CCL28, CXCL14, and CXCL17, play an important role in protecting mucosal surfaces from infectious pathogens. However, their role in protection against genital herpes remains to be fully explored. The CCL28 is a chemoattractant for the CCR10 receptor-expressing immune cells and is produced homeostatically in the human vaginal mucosa (VM). In this study, we investigated the role of the CCL28/CCR10 chemokine axis in mobilizing protective antiviral B and T cell subsets into the VM site of herpes infection. We report a significant increase in the frequencies of HSV-specific memory CCR10+CD44+CD8+ T cells, expressing high levels of CCR10, in herpes-infected asymptomatic (ASYMP) women compared with symptomatic women. Similarly, a significant increase in the CCL28 chemokine (a ligand of CCR10), was detected in the VM of herpes-infected ASYMP C57BL/6 mice, associated with the mobilization of high frequencies of HSV-specific effector memory CCR10+CD44+CD62L-CD8+ TEM cells and memory CCR10+B220+CD27+ B cells in the VM of HSV-infected ASYMP mice. Inversely, compared with wild-type C57BL/6 mice, the CCL28 knockout (CCL28-/-) mice (1) appeared to be more susceptible to intravaginal infection and reinfection with HSV type 2, and (2) exhibited a significant decrease in the frequencies of HSV-specific effector memory CCR10+CD44+CD62L-CD8+ TEM cells and of memory CD27+B220+ B cells in the infected VM. These findings suggest a critical role of the CCL28/CCR10 chemokine axis in the mobilization of antiviral memory B and T cells within the VM to protect against genital herpes infection and disease.

Samples of heart tissue were analyzed after myocardial infarction from WT and reg3beta knock-out mice. Samples from scar tissue and samples adjacent to the scar were analyzed. In the experiment we primarily compared infarction zone of wild-type to infarction zone of knock-out animals, and remote zone of wild-type to remote zone of knock-outs.

Project description:Angiogenesis plays a vital role in myocardial repair following myocardial infarction (MI). Y-box binding protein 1 (YBX1), an RNA-binding protein known for its role in cancer, is found to be significantly upregulated in endothelial cells within the peri-infarct zone and in hypoxic human umbilical vein endothelial cells (HUVECs). Using in vitro and in vivo models, we demonstrate that YBX1 enhances endothelial cell viability, proliferation, migration, and tube formation. Endothelial-specific overexpression of YBX1 in a murine MI model improved cardiac function and angiogenesis, while YBX1 knockdown aggravated cardiac injury. Transcriptome (RNA-seq) analysis revealed that YBX1 deficiency disrupts HIF-1α signaling. Mechanistically, YBX1 forms a complex with IGF2BP1/3 to stabilize HIF-1α mRNA through N⁶-methyladenosine (m⁶A) modification, thereby promoting angiogenesis and cardiac repair.

Project description:Tissue repair after myocardial infarction entails a vigorous angiogenic response that mitigates scarring and worsening of heart function and may represent a therapeutic target. Angiogenesis in the infarct wound is guided by incompletely defined myeloid cell–endothelial cell interactions. Here we identify the myeloid cell-expressed 75‑amino acid microprotein BRICK1 (short name: BRK1) as an indispensable driver of postinfarction angiogenesis. As a subunit of the intracellular actin-regulatory WAVE complex, BRK1 was not previously known to function outside the cell. We show that BRK1 translocates to the extracellular space after myocardial infarction in mice and humans. We find that BRK1 is not actively secreted but released from dying monocytes and macrophages. Cre-loxP-mediated myeloid cell-selective genetic deletion of Brk1 or antibody neutralization of extracellular BRK1 impaired microvessel formation in the infarct border zone and resulted in severe postinfarction heart failure in mice. Treatment with recombinant BRK1, conversely, preserved heart function after myocardial infarction. Mechanistically, BRK1 induced an angiogenic phenotype in human cardiac endothelial cells by signaling via the small GTPase RAP1 and mitogen-activated protein kinases 1 and 3 to promote retinoblastoma protein hyperphosphorylation and E2F transcription factor activation. BRK1 thus emerges as an angiogenic growth factor linking myeloid cell death to ischemic tissue repair and potentially enabling a protein-based therapy for myocardial infarction.

However, the dynamic regulatory mechanisms governing this network post-MI remain incompletely elucidated. Notably, the pivotal role of cardiac endothelial cells in the pathophysiological progression of MI calls for further comprehensive investigation.</p><p>METHODS: We examined alterations in Signal Peptide, CUB Domain And EGF Like Domain Containing 3 (SCUBE3) protein expression in the myocardium of MI-afflicted mice and human MI heart tissues. Utilizing genetically engineered mouse models in combination with diverse cellular and molecular biology techniques, we systematically dissected the functional role of SCUBE3 in MI and its underlying mechanisms in cardiac endothelial cell metabolism.</p><p>RESULTS: Proteomic profiling of serum samples from MI patients demonstrated a significant elevation in SCUBE3 protein levels. Immunohistochemical assessment revealed markedly increased SCUBE3 expression localized predominantly within fibroblasts in the infarct zone of cardiac tissues from MI patients. Consistently, in a murine MI model, SCUBE3 expression was also upregulated and primarily distributed in fibroblasts within the infarcted myocardial regions. Employing inducible, fibroblast-specific SCUBE3 overexpression and knockout mouse models, we observed that SCUBE3 overexpression robustly enhanced post-MI angiogenesis, improved microcirculatory network integrity, and attenuated myocardial injury. In contrast, SCUBE3 ablation exacerbated infarct-induced cardiac damage. Subsequent protein tracking and mass spectrometry analyses demonstrated that SCUBE3 selectively binds to and activates vascular endothelial growth factor receptor 1 (VEGFR1) on endothelial cells, thereby mediating downstream biological effects. Transcriptomic sequencing revealed that SCUBE3 significantly upregulates fatty acid metabolism–related pathways. Live-cell dynamic analysis further confirmed that SCUBE3 promotes fatty acid uptake and enhances the metabolic activity of endothelial cells. Metabolomic profiling indicated that SCUBE3 facilitates the conversion of unsaturated fatty acids into phosphosugars and nucleotides, supplying essential substrates and energy to support endothelial cell proliferation. In SCUBE3-overexpressing mice, endothelial cell–specific inducible VEGFR1 knockout completely abrogated SCUBE3-driven fatty acid metabolism and its associated cardioprotective effects. Moreover, beyond its role in angiogenesis SCUBE3 also markedly enhances cardiomyocyte regenerative capacity, highlighting its potential as a novel therapeutic target for myocardial repair.</p>

Project description:Compensatory angiogenesis is crucial for recovery in patients with diabetic myocardial infarction (DMI), yet effective interventions are still unclear. This study employed liquid chromatography-tandem mass spectrometry to identify lactylated proteins in the infarct border zone of DMI mouse hearts. Our findings revealed that IDH2 is lactylated at lysine 272, enhancing its binding to Cav1 while inhibiting the Cav1-eNOS interaction. This modification promotes eNOS activity and facilitates the proliferation, migration, and angiogenesis of cardiac microvascular endothelial cells under high glucose and hypoxic conditions. In endothelial cell-specific IDH2-K272R knock-in mice, the loss of K272 lactylation impairs cardiac function and exacerbates pathological remodeling due to disrupted angiogenesis.

Project description:Gene expression alterations in myocardial tissues following acute myocardial infarction (AMI) are of great significance for studying the pathophysiological mechanisms of this disease. RNA-seq was used to analyse the gene expression levels in myocardial tissues both in a mouse left anterior descending coronary artery (LAD) ligation model at the 4th day after MI and in sham-operated mice.We uncovered the upregulation of several angiogenesis-related genes after MI, which provided a basis for researches on cardiac repair following AMI.
