Metabolic Reprogramming in Primary Endocervical Cells Infected by Chlamydia trachomatis

Project description:Genital infection by Chlamydia trachomatis is the most common bacterial sexually transmitted disease worldwide, posing a significant threat to reproductive health. Its ability to persist silently in the host often delays treatment, leading to chronic inflammation and complications such as pelvic inflammatory disease, ectopic pregnancy, and infertility in females. Despite progress in understanding C. trachomatis pathogenesis, studies in primary cells remain limited. This study explored the interaction between C. trachomatis and primary human endocervical cells, revealing extensive host transcriptional changes, including strong inflammatory responses (IFN-α, IFN-γ, TNF-α), suppression of E2F targets, DNA repair, G2M checkpoint, and oxidative phosphorylation, indicating mitochondrial dysfunction. Downregulation of electron transport chain genes and phenotypic analysis showed selective TCA cycle impairments in succinate and citrate utilization. Further research is needed to uncover the mechanisms behind C. trachomatis-mitochondria interactions.

Project description:Chlamydia trachomatis (C. trachomatis) is an intracellular bacterium, and is one of the main pathogens that cause sexually transmitted infections worldwide. Long non-coding RNAs (lncRNAs) have become vital regulators in many biological processes. However, few studies have shown that lncRNAs take part in the pathogenesis of C. trachomatis. Here, we used microarrays to study the expression profiles of lncRNAs and mRNAs in HeLa cells at 12, 24, and 40 hours pot-infection (hpi). Our study provides evidence that lncRNAs are involved in the interaction between C. trachomatis and hosts.

Impact of HPV16 E6 and E7 oncogenes together with Chlamydia trachomatis infection on proteome changes in immortalized oral keratinocytes

Project description:Certain types of Human papilloma viruses (HPV) are the etiological agents for cervical cancer. However, not all infections of high-risk HPVs will finally lead to cancer since most HPV infections are cleared without any consequences. Chlamydia trachomatis is the most prevalent sexual transmitted bacteria and is an obligatory intracellular pathogen exhibiting tropism in endocervical epithelial cells. Over the past decades, C. trachomatis is thought to be a potential co-factor for cervical cancer formation, but there are also studies that did not show such a correlation. To address this question in molecular terms, we stably expressed HPV16 E6 and E7 in spontaneously immortalized NOKs (normal oral keratinocytes) and performed SILAC (stable isotope labeling by amino acids in cell culture) with or without C. trachomatis infection to study the impact of HPV16 oncogene expression and C. trachomatis infection on host proteome changes.

Early transcriptional landscapes of Chlamydia trachomatis-infected epithelial cells at single cell resolution

Project description:Here we examined host cell gene expression in response to Chlamydia trachomatis infection by applying scRNA-Seq to in vitro C. trachomatis-infected HEp-2 epithelial cells and time-matched uninfected cells over the early chlamydial developmental cycle (3, 6 and 12 hours). We collected 264 single cells across both conditions all time points. The aims of the experiment were examining host cell responses to infection at single cell resolution, and identifying early host cell signatures of infection.
