Peripheral Blood Monocyte Gene Expression in Recent-Onset Type 1 Diabetes

Project description:OBJECTIVE: Novel biomarkers of disease progression after type 1 diabetes onset are needed. RESEARCH DESIGN AND METHODS: We profiled peripheral blood (PB) monocyte gene expression in 6 healthy subjects and 16 children with type 1 diabetes diagnosed ~3 months previously, and analyzed clinical features from diagnosis to 1 year. RESULTS: Monocyte expression profiles clustered into two distinct subgroups, representing mild and severe deviation from healthy controls, along the same continuum. Patients with strongly divergent monocyte gene expression had significantly higher insulin dose-adjusted HbA1c levels during the first year, compared to patients with mild deviation. The diabetes-associated expression signature identified multiple perturbations in pathways controlling cellular metabolism and survival, including endoplasmic reticulum and oxidative stress (e.g. induction of HIF1A, DDIT3, DDIT4 and GRP78). qPCR quantitation of a 9-gene panel correlated with glycaemic control in 12 additional recent-onset patients. The qPCR signature was also detected in PB from healthy first-degree relatives. CONCLUSIONS: A PB gene expression signature correlates with glycaemic control in the first year after diabetes diagnosis, and is present in at-risk subjects. These findings implicate monocyte phenotype as a candidate biomarker for disease progression pre- and post-onset, and systemic stresses as contributors to innate immune function in type 1 diabetes. CD14+ monocytes from a total of 16 children with recent-onset type 1 diabetes and 6 adult healthy controls were profiled in 2 independent microarrays.

Blood biomarkers of pancreatic cancer associated diabetes identified by peripheral blood-based gene expression profiles

Project description:The prognosis of pancreatic cancer is still very poor, how to detect pancreatic cancer from high-risk group in an early stage is essential for improving its long-time survival. Therefore, the purpose of this study was to explore specific biomarkers that can differentiate pancreatic cancer-associated diabetes from type-2 diabetes for the early detection of pancreatic cancer. In the current study, we used global gene transcription analysis with affymetrix gene chip to identify genes specifically expressed in pancreatic cancer-associated diabetes mellitus from peripheral blood samples in stead of from tissue samples. 32 peripheral blood samples were collected for microarray experiments to find differentially expressed genes specific for pancreatic cancer associated diabetes, which included 8 patients diagnosed as pancreatic cancer with diabetes, 8 patients of pancreatic cancer without diabetes, 8 patients with diabetes mellitus>5years and 8 healthy controls. the comparision was done between pancreatic cancer with diabetes vs normal, pancreatic cancer vs normal and diabetes vs normal to identify a small group of genes that differently expressed in the pancreatic cancer with diabetes group.

High-throughput sequencing of the IGH repertoire in human peripheral blood and bone marrow

Project description:We applied high-throughput sequencing of the IGH repertoire in peripheral blood and bone marrow (BM) of a primary immunodeficiency patient. We obtained 325994 IGH sequences from the BM sample and 72597 IGH sequences from the peripheral blood. After comparing with IMGT/GENE-DB database (http://www.imgt.org), we found obviously increased CDR3 clonality in the patient BM as compared to his peripheral blood. Furthermore, the frequency of IgG transcripts in the patient BM was higher than that in the peripheral blood while the frequency of IgM transcripts was similar.

T cell receptor β-chains display abnormal shortening and repertoire sharing in type 1 diabetes

Project description:Defects in T cell receptor (TCR) repertoire are proposed to predispose to autoimmunity. Here we show, by analyzing >2×108 TCRB sequences of circulating naive, central memory, reg- ulatory and stem cell-like memory CD4+ T cell subsets from patients with type 1 diabetes and healthy donors, that patients have shorter TCRB complementarity-determining region 3s (CDR3), in all cell subsets, introduced by increased deletions/reduced insertions during VDJ rearrangement. High frequency of short CDR3s is also observed in unproductive TCRB sequences, which are not subjected to thymic culling, suggesting that the shorter CDR3s arise independently of positive/negative selection. Moreover, TCRB CDR3 clonotypes expressed by autoantigen-specific CD4+ T cells are shorter compared with anti-viral T cells, and with those from healthy donors. Thus, early events in thymic T cell development and repertoire generation are abnormal in type 1 diabetes, which suggest that short CDR3s increase the potential for self-recognition, conferring heightened risk of autoimmune disease.
