Project description:Extracellular vesicles (EVs) are important mediators of embryo attachment and outgrowth critical for successful implantation. While EVs have garnered immense interest in their therapeutic potential in assisted reproductive technology by improving implantation success, their large-scale generation remains a major challenge. Here, we report a rapid and scalable production of nanovesicles (NVs) from human trophectoderm cells (hTSCs) via membrane extrusion; these NVs can be generated in approximately 6 hours with a 20-fold higher yield than EVs isolated from the same number of cells. NVs display similar biophysical traits (morphologically intact, spherical, 90-130 nm) to EVs, and are laden with hallmark players of implantation (ITGA2/V, ITGB1, PRDX1, SOD2, and MFGE8) that include cell-matrix adhesion and extracellular matrix organisation proteins. Functionally, NVs are readily taken up by low-receptive endometrial HEC1A cells and reprogram their proteome towards a receptive phenotype that support hTSC spheroid attachment. Moreover, a single dose treatment of NVs significantly enhanced adhesion and spreading of mouse embryo trophoblast on fibronectin matrix. We show functional potential of NVs in enhancing embryo implantation and highlight their rapid and scalable generation, amenable to clinical utility.

Project description:Ineffective endometrial matrix remodeling, a key factor in infertility, impedes embryo implantation in the uterine wall. Our study reveals the cellular and molecular impact of human collagenase-1 administration in mouse uteri, demonstrating enhanced embryo implantation rates. Collagenase-1 promotes remodeling of the endometrial extracellular matrix (ECM), degrading collagen fibers and proteoglycans. This process releases matrix-bound bioactive factors, (e.g. VEGF, decorin), facilitating vascular permeability and angiogenesis. Collagenase-1 elevates embryo implantation regulators, including NK cell infiltration and the key cytokine LIF. Remarkably, uterine tissue maintains structural integrity despite reduced endometrial collagen fiber tension. In-utero collagenase-1 application rescues implantation in the heat stress and embryo transfer models, known for low implantation rates. Importantly, ex-vivo exposure of human uterine tissue to collagenase-1 induces collagen de-tensioning and VEGF release, mirroring remodeling observed in mice. Our research highlights collagenase potential to induce and orchestrate cellular and molecular processes enhancing uterine receptivity for effective embryo implantation. Project description:Endometrial receptivity plays a vital role in the success of embryo implantation. However, the temporal proteomic profile of porcine endometrium during embryo implantation is still unclear. In this study, the expression of proteins in endometrium on days 9, 10, 11, 12, 13, 14, 15 and 18 of pregnancy (D9, 10, 11, 12, 13, 14, 15 and 18) was profiled via iTRAQ technology. The results showed that 25, 55, 103, 91, 100, 120, 149 proteins were up-regulated, and 24, 70, 169, 159, 164, 161, 198 proteins were down-regulated in porcine endometrium on D10, 11, 12, 13, 14, 15 and 18 compared with that on D9, respectively. Among these differentially expressed (DE) proteins, MQM results indicated that S100A9, S100A12, HRG and IFI6 were differentially expressed in endometrial tissues during embryo implantation period. Bioinformatics analysis showed that the proteins differentially expressed in the 8 comparisons (D10 vs D9, D11 vs D9, D12 vs D9, D13 vs D9, D14 vs D9, D15 vs D9 and D18 vs D9) were involved in important processes and pathways related to immunization, endometrial remodeling, which have a vital effect on embryonic implantation. Our results reveal that RBP4 could regulate the cell proliferation, migration and apoptosis of endometrial epithelial cells and endometrial stromal cells to affect embryo implantation. This research also provides resources for studies of proteins in endometrium during early pregnancy.

Some differentially expressed genes, including upregulated Areg, Ihh, Lifr and downregulated Msx1, Pgr, Gata2, have been reported to regulate the establishment of uterine receptivity. Besides, we found that blastocysts induced an increase in both the number and acidification of lysosome, consistent with enhanced lysosomal hydrolase activity in uterine Le. Further exploration uncovered that blastocyst-derived IGF2 was involved into the activation of epithelial STAT3 to induce lysosomal hydrolase expression, and inhibition of lysosomal function derails normal uterine receptivity and embryo implantation. Finally, based on the proteomic data of both epithelia and the separated lysosome, it was revealed that CLDN1 and MUC1, two well-known downregulated molecules for successful implantation, are degraded by epithelial lysosome. In brief, our data demonstrated that blastocysts induced normal epithelium differentiation with lysosome activation to promote the establishment of uterine receptivity for embryo implantation.

Project description:The success of early pregnancy depends largely on the successful implantation of the embryo. Dynamic crosstalk between embryo and mother plays a crucial role in this process, but our understanding is still limited. Here, using the pig as a model, we first comprehensively profile the single-cell transcriptome of early free embryos and corresponding maternal endometrium, identifying embryonic and matched endometrial cells with heterogeneity. We developed a novel tool, ExtraCellTalk, to predict crosstalk between the early embryo and the maternal uterus using uterine luminal proteins as bridges, generating an overall dynamic map of maternal-fetal crosstalk. Through cross-species comparisons, we identified a conserved RBP4/STRA6 pathway in which embryonic-derived RBP4 targets the STRA6 receptor on stromal cells in the endometrium, thereby interacting with endothelial and perivascular cells to regulate their interconversion to promote endometrial remodeling. These maternal-fetal crosstalks and corresponding endometrial changes that we uncovered are essential for subsequent embryo implantation and are the basis for further studies to improve embryo implantation.

Project description:Currently, in vitro fertilization (IVF) is one of the most common approaches towards treating infertility. However, the success of this approach is still relatively low an the outcome of the IVF procedure is challenging to predict. At present, there is no highly successful marker that could reliably predict the pregnancy outcome in the process of assisted reproduction (AR). Currently, multiple embryo transfers are performed to warrant a successful pregnancy outcome, but this commonly results in multiple pregnancies, narrowing the feasibility of this method for a subset of couples affected by infertility. Recently IVF cycles with single-embryo transfers have become an option of interest in AR approaches. Such cycles, however, are significantly less successful and for this reason, there is an incentive to improve the pregnancy chance per IVF cycle by incorporating more criteria for selection of the embryo with the highest implantation potential. Gene expression in follicular cells, including granulosa cells (GC) and cumulus cells (CC) has previously been considered as a tool to predict the quality of the oocytes in several studies in addition to assessing the morphologic criteria of the embryos. Gene expression estimation in these cells, especially of the CC population, carries along several benefits as their harvesting is non-invasive, they are in direct contact with the oocyte and are usually discarded during the process of intracytoplasmatic sperm injection procedure (ICSI), making them any easily accessible surrogate tissue for gene expression analyses. Considering that several studies reporting various rates of success in prediction of AR cycle outcome with expression as a biomarker have been published, we attempted to perform the largest study of global gene expression alterations in finding novel biomarkers to outcome of in- vitro fertilization, which would encompass discovery of new biomarker genes and subsequent confirmational study of the selected biomarker set on an independent validation group. In this experiments 4 samples were hybridized twice, to allow for inspection of technical validity of microarray experiments.

Project description:Successful embryo implantation requires coordinated changes in the uterine luminal epithelium, including structural alterations, apical-basal polarity shifts, intrauterine fluid resorption, and cellular communication. These processes, collectively termed plasma membrane transformation (PMT), are crucial but poorly understood, especially regarding their regulation by planar cell polarity (PCP) proteins. PCP proteins, essential for cell organization, are understudied in the context of uterine physiology. Prickle proteins, particularly Prickle1, play critical roles in cellular polarization and tissue morphogenesis. However, their function in the endometrial epithelium during embryo implantation is unknown. To investigate Prickle1's role in uterine physiology, we developed an endometrial epithelial conditional knockout (cKO) of Prickle1 using Lactoferrin-icre. PRICKLE1 ablation in the endometrial epithelium resulted in decreased fertility and embryo implantation by gestational day 4.5. Three-dimensional imaging revealed abnormal luminal folding, impaired luminal closure, and altered glandular structure in mutant uteri. Additionally, we observed decreased aquaporin-2 expression, disrupted cellular architecture, and protein expression and localization in mutant uteri. Evidence of epithelial-to-mesenchymal transition (EMT) was found within luminal epithelial cells, further linking PRICKLE1 loss to uterine pathologies. Furthermore, altered cellular division planes and increased multinucleated cells suggested a connection to EMT. Our findings highlight PRICKLE1's critical role in the PCP pathway within the uterus, revealing its importance in the molecular and cellular responses essential for successful pregnancy establishment.
