Project description:Plants have evolved mechanisms to perceive and face mechanical stress (MS) caused by physical forces, including compacted soils, winds, rain, pathogens, and interactions with animals and plants. Previous research indicated that applying mechanical treatment (MT) to Arabidopsis increases both xylem area and seed yield. To explore sustainable tomato production, we applied MT - combining stem bending, weighting, and particular touching - to 10-day-old seedlings, using a specific weight on the upper stem for 48 h. Two days after the treatment, we observed stem enlargement and increased the number of xylem vessels and area in MT plants. Additionally, we noticed earlier flowering, leading to increased tomato production. The transcriptome of MT-treated plants revealed significant changes in the expression of several essential genes involved in central metabolism, growth responses, and crucial phytohormone signalling. By studying different tomato mutants in the ethylene and auxin signalling pathways, we demonstrated that both hormones play essential roles in the plant responses to combined MT. Our findings suggest that combined MT generates a beneficial MS in tomato plants that induces plant morphoanatomical changes that promote early flowering and increased yield, providing a promising strategy for sustainable agriculture.

Project description:Jasmonates are well known signaling components required for diverse processes ranging from wound and pathogen responses to regulation of developmental processes. A prominent feature of jasmonate biosynthesis or signaling mutants is the loss of fertility. In contrast to the male sterile phenotype of Arabidopsis mutants, the loss of the co-receptor protein COI1 in the tomato mutant jai1-1 results in female sterility with additional severe effects on stamen and pollen development. While a general model of jasmonate effects on male gametophyte development has been proposed to involve the regulation of water transport, the molecular details of this response are scarce. Here we show an extensive temporal profiling of the development, hormone content, transcriptome and metabolome of tomato stamen in six distinct stages of flower development in wild type and jai1-1. We found that the premature stamen desiccation of jai1-1 stamens and the preponed pollen maturation coincide with an accumulation of desiccation-related metabolites. The wild type shows a transient increase of jasmonates up to mid-flowering that is absent in jai1-1. This finding coincides with an early increase of the ethylene precursor ACC in jai1-1 that is similarly reflected in the increased expression of ethylene biosynthesis and response genes. Our data suggests an essential role of jasmonates in the temporal inhibition of ethylene production to prevent premature desiccation of stamens and to ensure proper timing in flower development.

Project description:Tomato fruit ripening is under the control of ethylene as well as a group of ethylene-independent transcription factors, including NON-RIPENING (NOR) and RIPENING INHIBITOR (RIN). During ripening, the linear carotene lycopene accumulates at the expense of cyclic carotenoids. Fruit-specific overexpression of LYCOPENE β-CYCLASE (LCYb) under the control of the PHYTOENE DESATURASE (PDS) promoter resulted in increased levels of β-carotene and ABA and in decreased ethylene levels. Genes regulated by ABA, or involved in its synthesis and signaling, were overexpressed, while those associated with ethylene and cell wall remodeling were repressed. In agreement with the transcriptional data, LCYb-overexpressing fruits exhibited increased density of cell wall material containing linear, under-methylated pectins and displayed an array of additional ripening phenotypes, including delayed softening, increased turgor, enhanced shelf life and a thicker cuticle with a higher content of cutin monomers and triterpenoids. The levels of several primary metabolites and phenylpropanoids also changed in the transgenics, which could be attributed to delayed fruit ripening and to ABA respectively. Network correlation analysis suggests that ABA, acting through NOR and RIN, is responsible for many of the above phenotypes. These data reinforce suggestions that ABA plays an important role in tomato fruit ripening and provide clues that fruit b-carotene, acting as a precursor for ABA, actively participates in controlling the ripening process rather than merely being an output thereof. Overexpression of a LCYb gene from Arabidopsis under the control of the ripening-associated PDS promoter leads to ripe tomato fruits accumulating high β-carotene levels. Using several independent transgenic lines, we conducted a system-wide study of the effect of increased β-carotene levels on tomato fruit ripening and shelf life. Our data suggest that β-carotene, acting through ABA, is involved in a regulatory loop within the network controlling tomato fruit ripening.

Project description:Prior exposure to microbial-associated molecular patterns or specific chemical compounds can promote plants into a primed state with stronger defence responses. b-aminobutyric acid (BABA) is an endogenous stress metabolite that induces resistance protecting various plants towards diverse stresses. In this study, by integrating BABA-induced changes in selected metabolites with transcriptome and proteome data, we generated a global map of the molecular processes operating in BABA-induced resistance (BABA-IR) in tomato. BABA significantly restricts the growth of the pathogens Oidium neolycopersici and Phytophthora parasitica but not Botrytis cinerea. A cluster analysis of the upregulated processes showed that BABA acts mainly as a stress factor in tomato. The main factor distinguishing BABA-IR from other stress conditions was the extensive induction of signalling and perception machinery playing a key role in defence priming. Interestingly, the signalling processes and immune response activated during BABA-IR in tomato differed from those in Arabidopsis with substantial enrichment of genes associated with jasmonic acid and ethylene signalling and no change in Asp levels. Our results revealed key differences between the effect of BABA on tomato and other model plants studied until now. Surprisingly, salicylic acid is not involved in BABA downstream signalization whereas ethylene and jasmonic acid play a crucial role.

Project description:The protein hormone florigen is a universal systemic inducer of flowering and a generic growth terminator across meristems. To understand the developmental rational for its pleiotropic functions and to uncover the deep cellular systems mobilized by florigen beyond flowering we explored the radial expansion of tomato stems. RNAseq, genetic analysis and histological validations show that endogenous, mobile, or induced florigen accelerate secondary cell wall biogenesis (SCWB), and hence vascular maturation, in coordination with but independently of flowering. Conversely, a systemic FT-like florigen antagonist from Ginkgo biloba, arrests SCWB. Downstream of florigen, RNAseq identified MADS/FUL and MIF genes, that similarly impact SCWB independent from flowering. Florigen we show is remarkably stable and distributed to all organs regardless of pre-existing endogenous levels. By accelerating SCWB, florigen triggers a global redistribution of resources, signals and mechanical loads, thereby harmonizing vascular maturation with the reproductive development it had originally set into motion.

The apex is transformed from a vegetative to a floral meristem and shows morphological changes in 8 to 10 days, with flowers developing 8 to 10 weeks later. During eight sampling stages ranging from 6 hours to 8 days after treatment, 7,961 genes were found to exhibit differential expression (DEG) after the application of ethylene. In the first 3 days after treatment, there was little change in ethylene synthesis or in the early stages of the ethylene response. Subsequently, three ethylene response transcription factors (ERTF) were up-regulated and the potential gene targets were predicted to be the positive flowering regulator CONSTANS (CO), a WUSCHEL gene, two APETALA1/FRUTFULL (AP1/FUL) genes, an epidermal patterning gene and a jasmonic acid synthesis gene. We confirm that pineapple has lost the flowering repressor FLOWERING LOCUS C. At the initial stages, the SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1 (SOC1) was not significantly involved in this transition. Another WUSCHEL gene and a PHD homeobox transcription factor, though not apparent direct targets of ERTF, were up-regulated within a day of treatment, their predicted targets being the up-regulated CO, auxin response factors, SQUAMOSA, and histone H3 genes with suppression of abscisic acid response genes. The FLOWERING LOCUS T (FT), TERMINAL FLOWER (TFL), AGAMOUS-like APETELAR (AP2) and SEPETALA (SEP) increased rapidly within 2 to 3 days after ethylene treatment. Two FT genes were up-regulated in the apex and not the leaf bases after treatment, suggesting that transport did not occur. These results indicated that the ethylene response in pineapple and possibly most bromeliads acts directly to promote the vegetative to flower transition via APETALA1/FRUITFULL (AP1/FUL) and its interaction with SPL, FT, TFL, SEP and AP2.
