Project description:Long non-coding RNA (LncRNA) is a significant epigenetic regulatory factor, and its allele-specific expression (ASE) may play an important role in mental disorders. Therefore, clarifying the relationship between allele-specific expression (ASE) of lncRNA and mental disorders is of great significance for an in-depth understanding of the molecular mechanism of mental disorders. We here report that ASE-driven allelic shift in LINC02449 in Bipolar disorder (BD) and schizophrenia (SZ) contributes to behavioral and synaptic abnormalities in mice. We found that when the alternative allele of LINC02449 was overexpressed in cells and mouse brain regions, the expression of differentially expressed genes would be significantly enriched in mental disorders, mediating enhanced synaptic excitability transmission. Taken together, our data supported that LINC02449 overexpression, driven by the rs149707223 alternative allele, affects the disease susceptibility of BD/SZ.

A genetically defined mPFC-thalamic circuit underlies pain chronicity

Project description:Chronic pain is a major health care problem with great social and economic consequences. Although the medial prefrontal cortex (mPFC) and the thalamus have been implicated in regulating pain, whether and how these regions may involve in pain chronification process have remained largely unknown. Here, we show that Foxp2+ neurons in mPFC, which are corticothalamic (CT) neurons representing the major mPFC output to thalamus, are deactivated in chronic inflammatory pain. Interestingly, prolonged, but not short-term, inactivation of the Foxp2+ neurons in mPFC increases the sensitivity to noxious stimulations. Conversely, chemogenetic activation of this neuronal cluster induces robust antinociceptive effects in both naïve mice and mice with chronic inflammatory pain. Furthermore, we found that the mPFC Foxp2+ neurons project to several thalamic relay nuclei and that activation of the mPFC to the parataenial nucleus of thalamus (PTN) circuit relieves pain. Finally, RNA-seq of the mPFC Foxp2+ neurons revealed that many genes related to neuronal activity are dysregulated in mice with chronic inflammatory pain. Collectively, our studies revealed that Foxp2+ neurons in mPFC that project to thalamus play a critical role in somatosensory processing and pain chronification.

Effect of the Idd2 locus of B10 origin on the transcriptome of B cell of NOD mice

Project description:Type 1 diabetes (T1D) is an autoimmune disease characterized by pancreatic beta-cell destruction. It is a complex genetic trait driven by over 30 genetic loci with parallels between humans and mice. The NOD mouse spontaneously develops autoimmune diabetes and is widely used to identify insulin-dependent diabetes (Idd) genetic loci linked to diabetes susceptibility. While many Idd loci have been extensively studied, the impact of the Idd2 locus on autoimmune diabetes susceptibility remains to be defined. To address this, we generated a NOD congenic mouse bearing B10 resistance alleles on chromosome 9 in a locus coinciding with part of the Idd2 locus and find that NOD.B10-Idd2 congenic mice are highly resistant to diabetes. Bone marrow chimera and adoptive transfer experiments show that the B10 protective alleles provide resistance in an immune cell-intrinsic manner. While no T cell-intrinsic differences between NOD and NOD.B10-Idd2 mice were observed, we find that the Idd2 resistance alleles limit the formation of spontaneous and induced germinal centers. Comparison of the B cell transcriptome profile from NOD and NOD.B10-Idd2 mice reveal that resistance alleles at the Idd2 locus affect the expression of specific MHC molecules, a result confirmed by flow cytometry. Altogether, these data demonstrate that resistance alleles at the Idd2 locus impair germinal center formation and influence MHC expression, both of which likely contribute to reduce diabetes incidence.

4C analysis to capture chromatin conformations of the Tfap2c-Bmp7 locus upon different genomic rearrangements in mice

Project description:A genomic locus tends to form a particular conformational structure in the nucleus, but it is not known how a specific structure is dictated. To understand the mechanism, we made serial genomic rearrangements (deletion and inversion) in the Tfap2c-Bmp7 locus in mice, and analyzed the chromatin conformation in these alleles by 4C. For the wild type allele, we took not only the whole embryo but also different tissues such as the heart, forebrain (lateral and medial part separately), limb buds, and we used only the whole embryos for the mutant alleles.
