Project description:Immunotherapy has emerged as a promising treatment for head and neck squamous cell carcinoma (HNSCC), but clinical responses remain limited. Elevated expression of interferon-stimulated gene 15 (ISG15), associated with oral squamous cell carcinoma (OSCC), may contribute to this limited efficacy. While chronic interferon signaling impairs CD8 T cell function, the specific role of secreted ISG15 in T cell exhaustion remains unclear. Analyzing human OSCC datasets, we observed significant enrichment of the ISG core score, including ISG15, in tumors compared to adjacent non-tumor tissues. Using a novel in vitro model of human T cell dysfunction, we found that acute ISG15 exposure enhances CD8 T cell effector functions, whereas prolonged ISG15 exposure induces severe dysfunction via an endocytic mechanism independent of LFA-1 signaling. In an immunocompetent orthotopic OSCC model, ISG15-expressing tumors exhibited accelerated growth and recruited more tumor-reactive CD8 T cells though these cells were dysfunctional. Importantly, PD-1 blockade treatment significantly slowed tumor progression and restored T cell functionality in ISG15-expressing tumors. Our findings indicate that chronic ISG15 exposure promotes CD8 T cell dysfunction; however, these cells remain responsive to PD-1 blockade. This study reveals ISG15 as a potential biomarker for identifying patients likely to benefit from PD-1 blockade therapy.

However, emerging data suggests more diverse roles for PCs as regulators of immune and inflammatory responses via secretion of factors other than immunoglobulins. The extent to which such responses are pre-programmed in B-lineage cells or can be induced in PCs by the microenvironment is unknown. Here we dissect the impact of IFNs on the regulatory networks of human plasma cells. We show that core PC programs are unaffected, while PCs respond to IFNs with distinctive transcriptional responses. The ISG15-system emerges as a major transcriptional output induced in a sustained fashion by IFN-α in PCs and linked both to intracellular conjugation and ISG15 secretion. This leads to the identification of ISG15-secreting plasmablasts/PCs in patients with active SLE. Although ISG15 was the founding member of the ubl protein family, very little is known about its function. We have found that ISG15 expression in non-phagocytic cells is dramatically induced upon Listeria infection and that surprisingly this induction can be Type I Interferon independent. Listeria-mediated ISG15 induction depends on the cytosolic surveillance pathway, which senses bacterial DNA and signals through STING, TBK1, IRF3 and IRF7. Most importantly, we observed that ISG15 expression restricts Listeria infection both in vitro and in vivo. We then made use of Stable Isotope Labeling in tissue culture (SILAC) to identify the ISGylated proteins that could be responsible for the ISG15-mediated protective effect. Our SILAC analysis revealed that overexpression of ISG15 leads to a striking ISGylation of integral membrane proteins of the endoplasmic reticulum and Golgi apparatus, which correlates with increased canonical secretion of cytokines. Taken together, our data reveal a previously uncharacterized signaling pathway that restricts Listeria infection and acts via ISGylation, reinforcing the view that ISG15 is a key component of the innate immune arsenal of the mammalian host.

Project description:Cytokine storm during respiratory viral infection is an indicator of disease severity and poor prognosis. Type 1 interferon (IFN-I) production and signaling have been reported to be causal in cytokine storm associated pathology in several respiratory viral infections, however, the mechanisms by which IFN-I promotes disease pathogenesis remain poorly understood. We report using Usp18-deficient, USP18 enzymatic inactive and Isg15-deficient mouse models that lack of deISGylation during persistent viral infection leads to severe immune pathology characterized by hematological disruptions, cytokine amplification, lung vascular leakage and death. Transcriptional studies of immune cells from mice bearing the enzymatic inactive version of USP18 (Usp18C61A) revealed a neutrophil influx in the lung of these animals during infection, associated with increased immature neutrophil content. These neutrophil differentiation changes were dependent on ISG15 as deletion of Isg15 from C61A animals reversed the accumulation of immature neutrophils. Importantly, neutrophil depletion reversed morbidity and mortality in Usp18C61A mice. In summary, we reveal Usp18 enzymatic function is crucial for regulating extracellular release of ISG15 which are accompanied by altered neutrophil differentiation, cytokine amplification and mortality following persistent viral infection.

Project description:Proper control of inflammatory responses is essential for embryonic development, but the underlying mechanism is poorly understood. Here, we show that under physiological conditions, inactivation of ISG15, an inflammation amplifier, is associated with the interaction of Beclin 1 (Becn1), via its ECD domain, with STAT3 in the major fetal hematopoietic organ of mice. Conditional loss of Becn1 caused sequential dysfunction and exhaustion of fetal liver hematopoietic stem cells, leading to lethal inflammatory cell-biased hematopoiesis in the fetus. Molecularly, the absence of Becn1 resulted in the release of STAT3 from Becn1 tethering and subsequent phosphorylation and translocation to the nucleus, which in turn directly activated the transcription of ISG15 in fetal liver hematopoietic cells, coupled with increased ISGylation and production of inflammatory cytokines, whereas inactivating STAT3 reduced ISG15 transcription and inflammation but improved hematopoiesis potential, and further silencing ISG15 mitigated the above collapse in the Becn1 null hematopoietic lineage. The Becn1-STAT3-ISG15 axis remains functional in Atg5/7-disrupted fetal hematopoietic organs. These results suggest that Becn1, in an autophagy-independent manner, secures hematopoiesis and survival of the fetus by directly inhibiting STAT3-ISG15 activation to prevent cytokine storms. Our findings highlight a previously undocumented role of Becn1 in governing ISG15 to safeguard the fetus.
