Project description:Sugarcane streak mosaic virus (SCSMV) is the most important cause of sugarcane mosaic disease in the main production region. The SCSMV-encoded protein 1 (P1SCSMV) plays a vital role in disease development. However, the molecular mechanisms by which P1SCSMV participates in plant immunity remains unknown. Here, we found that the relative expression levels of P1SCSMV were negatively correlated with the xyloglucan endotransglucosylase/hydrolase gene (XTH) in a linear relationship. P1SCSMV directly interacts with the transcription factor, NbBES1, in vivo and in vitro. P1SCSMV-NbBES1 interaction interferes the binding of NbBES1 to the E-box element on the promoter region of NbXTH1 gene and inhibits the normal transcription of NbXTH1 gene. Furthermore, we found that down-regulation of the NbXTH1 directly resulted in enhanced infection of different plant pathogenic fungi and viruses, disrupted compositions of xyloglucan and xylan in the cell wall, and thinner middle lamellae in cell walls. We found that xyloglucan is a direct trigger of damage-associated molecular pattern- (DAMP) induced plant innate immunity. Moreover, the decrease in xyloglucan content in the cell wall caused by down-regulation of NbXTH1 leads to a reduction in DAMP-triggered immunity. Our study revealed the molecular mechanism of how a plant virus manipulates plant cell wall-mediated plant innate immunity, and identified NbXTH1, a wide-spectrum disease resistance protein for both plant pathogenic fungi and viruses. Our research also provides the basis for the application of the small molecule xyloglucan and the wide-spectrum disease resistance gene, XTH, in wide-spectrum resistance improvement in the future crop molecular breeding.

Part of BES1 activity as a transcription factor is brought about upon dimerization with light-responsive PIF4 transcription factor. To analyse BES1-dependent transcriptional cascade, and to which extent BES1 requires PIF4/PIF5 for target gene regulation, we performed microarray analysis of the following phenotypes: Col-0, bes1-D, pif4pif5 and bes1-D;pif4pif5 in dark grown seedlings. Col-0, bes1-D, pif4pif5 and bes1-D;pif4pif5 5-days seedlings grown in the dark were used for RNA extraction. 3 independent biological samples were collected for each genoptye.

In order to further understand the mechanism by which BES1/BZR1 regulates downstream genes, we performed chromatin immunoprecipitation coupled with tiling arrays (ChIP-chip) to identify BES1 binding sites in the genome. By combining ChIP-chip data with gene expression microarray data, we are able to discover genes that are directly regulated by BES1 (i.e. BES1 target genes). Chromatin from bes1-D Arabidopsis seedlings are immunoprecipitated by anti-BES1 antibody with anti-GFP antibody as control.

Project description:Sugarcane mosaic and leaf fleck diseases are significant viral diseases affecting sugarcane crops in India. The use of resistant sugarcane varieties is considered the most economical and effective approach to manage viral diseases, especially in vegetatively propagated crops such as sugarcane. Sugarcane mosaic virus (SCMV) and Sugarcane streak mosaic virus (SCSMV) are the primary pathogens responsible for mosaic disease in sugarcane-growing regions of India. Sugarcane bacilliform virus (SCBV), causing leaf fleck disease, is also often found in mixed infections with mosaic symptoms. The study aimed to identify new sources of resistance by screening sugarcane germplasm for resistance to SCMV, SCSMV, and SCBV. The screening was carried out under high inoculum using the infector row method in both plant and ratoon crops. Out of 129 genotypes tested, only 8 were found to be free of mosaic viruses, indicating a rare occurrence of resistant sources. The study revealed that mosaic disease is widespread, with nearly 95% of tested varieties/genotypes being infected with mosaic viruses. SCMV, SCSMV, and SCBV were detected in 121 out of 129 genotypes using the RT-PCR and PCR assays. Based on their response to the viruses, the tested genotypes were categorized into different resistance grades: highly resistant (grade 1), resistant (grade 2), moderately resistant (grade 3), susceptible (grade 4), and highly susceptible (grade 5). The results of the study provide valuable information about elite resistance resources that can be used for the prevention and control of mosaic disease. These resistant genotypes could also serve as potential donors for mosaic and leaf fleck disease resistance in breeding programs.

Project description:Sugarcane mosaic disease, mainly caused by Sugarcane streak mosaic virus (SCSMV), has serious adverse effects on the yield and quality of sugarcane. Eukaryotic translation initiation factor 4E (eIF4E) is a natural resistance gene in plants. The eIF4E-mediated natural recessive resistance results from non-synonymous mutations of the eIF4E protein. In this study, two sugarcane varieties, CP94-1100 and ROC22, were selected for analysis of their differences in resistance to SCSMV. Four-base missense mutations in the ORF region of eIF4E resulted in different conserved domains. Therefore, the differences in resistance to SCSMV are due to the inherent differences in eIF4E of the sugarcane varieties. The coding regions of eIF4E included 28 SNP loci and no InDel loci, which were affected by negative selection and were relatively conserved. A total of 11 haploids encoded 11 protein sequences. Prediction of the protein spatial structure revealed three non-synonymous mutation sites for amino acids located in the cap pocket of eIF4E; one of these sites existed only in a resistant material (Yuetang 55), whereas the other site existed only in a susceptible material (ROC22), suggesting that these two sites might be related to the resistance to SCSMV. The results provide a strong basis for further analysis of the functional role of eIF4E in regulating mosaic resistance in sugarcane.
