Project description:Post-stress hepatic metabolic dysfunction is a critical factor in the development of negative emotional states after liver transplantation (NEALT), severely impairing postoperative patient recovery. However, its pathological mechanisms remain unclear. Microglia (Mg), as central immune cells, express numerous sensory receptors on their membranes and serve as a key link between peripheral organ stress and brain nucleus regulation. By constructing microglia-specific gene knockout mice, liver transplantation, and restraint stress models, combined with behavioral tests and multi-omics analysis, we found that transplanting a stressed donor liver into a non-stressed recipient mouse could mimic clinical NEALT symptoms. The anterior cingulate cortex (ACC) is a key brain region regulating emotional responses and sensory perception. Mechanistic studies revealed that stress-induced liver detoxification dysfunction leads to elevated bilirubin levels, which acts as a ligand to activate the TRPM2 receptor in ACC microglia, enhancing their impact on glutamatergic synapses and ultimately leading to decreased neuronal activity and NEALT development. We propose a hypothesis: stressed livers drive microglia to excessively engulf glutamatergic synapses via the liver-brain-Mg TRPM2 signaling axis. Evaluating interventions targeting this axis holds significant potential for NEALT prevention and treatment, providing significant insights into its mechanisms and therapeutic strategies.

Project description:BackgroundGrowth differentiation factor-15 (GDF15) is a member of the growth differentiation factor subfamily in the transforming growth factor beta superfamily. GDF15 has multiple functions and can regulate biological processes. High levels of GDF15 in the circulation can affect metabolic processes. Studies have shown that GDF15 is associated with changes in body weight.SummaryThis review reviews the current knowledge on the relationship between GDF15 and body weight change, focusing on the role and mechanism of GDF15 in body weight regulation. GDF15 plays an important role in reducing food intake, improving insulin resistance, and breaking down fat, suggesting that GDF15 has an important regulatory effect on body weight. The mechanism by which GDF15 causes reduced food intake may be related to changes in food preference, delayed gastric emptying, and conditioned taste aversion. GDF15 can combat insulin resistance induced by inflammation or protect β cell from apoptosis. GDF15 probably promotes lipolysis through a brain-somatic tissue circuit. Several factors and related signaling pathways are also mentioned that can contribute to the effects of GDF15 on reducing weight.Key messageGDF15 plays an important role in weight regulation and provides a new direction for the treatment of obesity. Its effects on resisting obesity are of great significance to inhibiting the progression of metabolic diseases. It is expected to become a new target for regulating body weight, improving obesity, and treating metabolic diseases such as diabetes.

Genomewide microRNA down-regulation as a negative feedback mechanism in the early phases of liver regeneration.

Project description:The liver is one of the few organs that have the capacity to regenerate in response to injury. We carried out genomewide microRNA (miRNA) microarray studies during liver regeneration in rats after 70% partial hepatectomy (PH) at early and mid time points to more thoroughly understand their role. At 3, 12, and 18 hours post-PH ?40% of the miRNAs tested were up-regulated. Conversely, at 24 hours post-PH, ?70% of miRNAs were down-regulated. Furthermore, we established that the genomewide down-regulation of miRNA expression at 24 hours was also correlated with decreased expression of genes, such as Rnasen, Dgcr8, Dicer, Tarbp2, and Prkra, associated with miRNA biogenesis. To determine whether a potential negative feedback loop between miRNAs and their regulatory genes exists, 11 candidate miRNAs predicted to target the above-mentioned genes were examined and found to be up-regulated at 3 hours post-PH. Using reporter and functional assays, we determined that expression of these miRNA-processing genes could be regulated by a subset of miRNAs and that some miRNAs could target multiple miRNA biogenesis genes simultaneously. We also demonstrated that overexpression of these miRNAs inhibited cell proliferation and modulated cell cycle in both Huh-7 human hepatoma cells and primary rat hepatocytes. From these observations, we postulated that selective up-regulation of miRNAs in the early phase after PH was involved in the priming and commitment to liver regeneration, whereas the subsequent genomewide down-regulation of miRNAs was required for efficient recovery of liver cell mass.Our data suggest that miRNA changes are regulated by negative feedback loops between miRNAs and their regulatory genes that may play an important role in the steady-state regulation of liver regeneration.
