Phactr1/PP1 phosphatase substrates in mouse NIH3T3 fibroblasts

Project description:It was previously shown that expression of an activated Phactr1 mutant (Phactr1XXX) that constitutively forms the Phactr1/PP1 phosphatase holoenzyme, induces F-actin rearrangements in NIH3T3 fibroblasts. Expression of the Phactr1 PP1-binding domain (C-terminal) alone is also sufficient to induce such cytoskeletal changes. In contrast, expression of Phactr1XXXC derivative, which lacks the PP1 binding sequences does not result in alteration of cytoskeletal morphology (Wiezlak et al., 2012). These observations suggest that Phactr1/PP1 dephosphorylates target proteins involved in cytoskeletal dynamics. To identify potential Phactr1/PP1 substrates, we used differential SILAC phosphoproteomics in NIH3T3 cells inducibly expressing Phactr1XXX, Phactr1XXXC constructs or vector alone. Over 3000 phosphorylation sites were quantified, among which we determined Phactr1/PP1 target dephosphorylation sites by comparative analysis.

Effects of Nogo-B receptor overexpression on transcriptomic profiling in the NIH3T3 fibroblast cell line

Project description:Unrestrained receptor tyrosine kinase (RTK) signaling and epigenetic dysregulation are responsible in part for tumorigenesis. Aberrant RTK signaling results in the remodeling of histone modification. As a Ras modulator, NgBR promotes Ras-mediated RTK signaling pathways. Meanwhile, we observed that NgBR overexpression in NIH3T3 cells significantly increased the acetylation of histone H3 and histone H4. To further elucidate the underlying mechanism that NgBR upregulates histone acetylation in tumorigenesis, we carried out RNA-seq analysis to examine the transcriptome alternation in NIH3T3 cells and NgBR-overexpressed NIH3T3 cells (3T3NgBR cells) and revealed that overexpression of NgBR up-regulates ACSS2 expression, but not ACSS1 and ACSS3. Moreover. ACSS2 overexpression abrogated NgBR deficiency-caused decrease of histone acetylation in NIH3T3 cells. Taken together, all the data suggested that NgBR regulates tumorigenesis in NIH3T3 cells via ACSS2-mediated histone acetylation.

Global gene expression profiling of Jmjd6- and Jmjd4-depleted mouse NIH3T3 fibroblasts

Project description:Emerging evidence suggests Jumonji domain-containing proteins are epigenetic regulators in diverse biological processes including cellular differentiation and proliferation. RNA interference-based analyses combined with gene expression profiling can effectively characterize the cellular functions of these enzymes. We found that the depletion of Jumonji domain-containing protein 6 (Jmjd6) and its paralog protein Jumonji domain-containing protein 4 (Jmjd4) individually by small hairpin RNAs (shRNAs) slowed cell proliferation of mouse NIH3T3 fibroblasts. We subsequently performed gene expression profiling on both Jmjd6- and Jmjd4-depleted mouse NIH3T3 fibroblasts using the Affymetrix GeneChip Mouse Exon 1.0 ST Array.
