Muscle Hypertrophy Induced by Myostatin Inhibition Accelerates Degeneration in Dysferlinopathy

Project description:Because myostatin normally limits skeletal muscle growth, there is an extensive effort to develop myostatin inhibitors for clinical use. One potential concern is that in patients with muscle degenerative diseases, inducing hypertrophy may increase stress on dystrophic fibers. Here, we show that blocking the myostatin pathway in dysferlin mutant mice results in early improvement in histopathology but ultimately accelerates muscle degeneration. Hence, benefits of this approach should be weighed against these potential detrimental effects. Affymetrix Mouse Exon 1.0 ST arrays were hybridized in three biologically independent experiments with RNA from quadriceps muscles of wt, Dysf-/-, F66, F66;Dysf-/- mice, and ACVR2B/Fc-injected wt and Dysf-/- mice at the age of 10 weeks (3 duplicates in 6 different groups, 18 samples).

Transcription profiling of Drosophila after exposure to microgravity in the International Space Station and in a microgravity simulator

Project description:Larvae-Pupae transition flies (Drosophila) were recovered and transport for 3 days at 12-14ºC to arrest development until the launch site, then exposed to RT (18-20ºC) for some hours including the launch and trip to the International Space Station, then pupae were exposed to microgravity in the ISS for 4 days and a half at 22ºC. Finally pupae were fixed on acetone and frozen until recovery on Earth.<br><br><br><br>Four groups of samples: 1 ISS (+ground control) as described, 2 RPM (microgravity simulator on Earth) as described, 3 RPM without constrains (No MAMBA container and only 5 days exposure without cold transport) and 4 centrifuge 10g without constrains control..

The purpose was to study the efficacy of microgravity to model impaired muscle regeneration in an engineered muscle platform and then to demonstrate the feasibility of performing drug screening in this model. Engineered human muscle was launched to the International Space Station National Laboratories, where the effect of microgravity exposure for 7 days was examined by transcriptomics.

Single muscle fiber proteomics reveals remodeling of focal adhesions in response to muscle unloading and microgravity

Project description:Spaceflight causes loss of muscle mass and strength, metabolic remodeling and insulin resistance, contributing to severe challenges for astronaut health. We used highly sensitive proteomics to detail single fiber type-specific molecular remodeling caused by muscle unloading in subjects undergoing prolonged bed rest. We then measured the muscle proteome of astronauts before and after a mission on the ISS. Our combined datasets show downregulation of complexes involved in fiber-matrix interaction, force transmission and insulin receptor stabilization on the sarcolemma. Unloading upregulated anti-oxidant responses in slow but not in fast fibers, markers of neuromuscular damage and the hypusination pathway that uniquely modifies translation initiation factor 5A. These muscle intrinsic changes in adhesion, protein turnover and redox balance may contribute to the whole-body detrimental effects caused by prolonged unloading. We conclude that single muscle fiber analysis by proteomics can support the development of advanced countermeasures targeting the mechano-metabolic molecular axis.

Space microgravity increases expression of genes associated with proliferation and differentiation in human cardiac spheres

Project description:Previously, we reported upregulation of cardiac proliferation in cardiac progenitors derived from human induced pluripotent stem cells (hiPSC-CPCs) that were exposed to space microgravity for 3 days. The overall objective of this investigation was to understand how long-term exposure to microgravity affects the expansion and differentiation of hiPSC-CPCs. Cryopreserved cardiomyocytes derived from human-induced pluripotent stem cells (hiPSC-CMs) were exposed to space microgravity for 3 weeks in the International Space Station (ISS) and retrieved for transcriptomic profiling. RNA-seq analysis and gene ontology analysis revealed upregulation of cardiac tissue function and morphological terms while downregulating inflammation and ECM regulation terms. Upregulation of genes associated with cell cycle regulation and proliferation were notable as well. Combined with previous work, these results suggest that space microgravity improved cardiac differentiation and structure formation and potentially, proliferation.

miRNAs expression of soleus response to simulated microgravity

Project description:Identification of gravisensitive miRNAs expression in rat soleus muscle exposed to 7 and 14 days of Hindlimb suspension (HS) simulated microgravity. Microgravity causes muscle atrophy possibly due to muscle wasting overtake regeneration. Results provide insight into the molecular mechanisms regulating muscle atrophy. The expression of 23 out of 174 miRNAs was found to change at least 2-fold of 7 and/or 14 days of TS. By using real-time PCR assays, we verified the microarray data using some of the expected genes.
