Project description:Mycobacterium tuberculosis (MTB) is a major global cause of mortality, responsible for over a million deaths each year. Despite this burden, natural immunity prevents disease in more than 90% of exposed individuals. Previous studies have identified interferon-gamma (IFN-γ) as a key regulator of innate immune defense against MTB. Here, we investigate the impact of IFN-γ timing on macrophage-mediated control of MTB infection. We demonstrate that IFN-γ exposure before infection enhances macrophage antibacterial activity, whereas post-infection exposure does not. Further analysis of this phenotype revealed a strong association between c-Myc signaling and macrophage function in MTB control, as identified through unbiased in vitro systems approaches. Given the difficulty of perturbing c-Myc in primary cells, we developed a lentiviral system for c-Myc inhibition and overexpression. We profiled both datasets (IFN-γ timing and c-Myc inhibition) to characterize the resulting transcriptional shifts.

Project description:Autophagy is a widespread physiological process in the body, which also protects the host by degrading invading pathogens and harmful substances during pathological conditions. However, Mycobacterium tuberculosis (MTB) can affect the process of autophagy by regulating the expres-sion of microRNAs (miRNAs), allowing for immune evasion. In this study, we constructed a model of a strong virulent strain (H37Rv) infection in human macrophage cell line. Following H37Rv infection, we screened 14 differentially expressed miRNAs by RNA-seq and bioinformatics. We predicted and demonstrated that miR-30c-1-3p inhibits autophagy and promotes macrophage survival by targeting ATG4B and ATG9B during the infection process.Additionally, the intervention of miR-30c-1-3p mimics resulted in an increased bacterial load in macrophages, suggesting that MTB achieves immune evasion by upregulating miR-30c-1-3p during infection. In conclusion, our study provides a valuable target for the development of host-directed anti-tuberculosis therapy as well as a new diagnostic marker.

Project description:We report here a modified protocol for generating homogenous populations of macrophage-like cells from human embryonic stem cells. The induced macrophages, referred to as iMAC, presented similar transcriptomic profiles and characteristic immunological features of classical macrophages and were permissive to viral and bacterial infection, in particular Mycobacterium tuberculosis (Mtb). Their production was amenable to scale up and the resulting cells suitable for high-throughput screening.

Project description:Transcriptional changes during early infection of macrophage-like THP-1 cell line with pathogenic bacterium Mycobacterium tuberculosis. RNAseq samples were taken at 0h (THP-1 cells growing in the RPMI medium), and after 4h, 24h and 48h post infection. Bacterial enrichment was performed to increase the amount of bacterial mRNA in the samples. Non-enriched samples were used to map THP-1 cells transcripts; enriched samples were used to map M. tuberculosis transcripts the corresponding genomes.

HnRNPA2B1 tunes antimycobacterial immune responses in macrophages through alternative splicing of Irgm1

Project description:Alternative splicing (AS) has been implicated in host responses to diverse pathogens, but our understanding of its role in Mycobacterium tuberculosis infection remains incomplete. Using RNA-sequencing and splicing-aware computational pipelines, we quantified AS in Mtb-infected bone marrow-derived macrophages (BMDMs). We found that ~5% of expressed macrophage genes exhibit one or more splicing changes at 8h post-Mtb infection, highlighting AS as a key layer of regulation in the macrophage response to intracellular pathogens. We next sought to identify RNA binding proteins that play an outsized role in shaping the macrophage transcriptome during Mtb infection. We discovered that the splicing factor hnRNPA2B1 promotes the early induction of inflammatory genes while dampening several type I interferon-stimulated genes. HnRNPA2B1 also controls alternative splicing of transcripts, notably Irgm1, a critical immunity-related GTPase. We report that the abundance of Irgm1-long, the canonical isoform, and Irgm1-short, an isoform lacking a lysosomal targeting domain is differentially regulated in response to diverse inflammatory cues. Macrophages overexpressing Irgm1-short are defective in restricting Mtb replication and accumulate LC3+ bacilli. These data highlight a key role for AS in shaping the macrophage transcriptome and pinpoint hnRNPA2B1 as a novel restriction factor in the cell-intrinsic response to Mtb.
