Project description:The phytohormone abscisic acid (ABA) promotes seed dormancy and inhibits seed germination and seedling development. It remains unclear how chromatin remodeling influences ABA responses. In the present study, an artificial microRNA targeting two Sucrose Non-Fermenting 2 (SNF2)-family chromatin remodeling ATPases, SPLAYED (SYD) and PHOTOPERIOD-INDEPENDENT EARLY FLOWERING1 (PIE1), was identified through unbiased forward genetic screenings of artificial microRNA (amiRNA)-expressing pooled seed libraries exhibiting ABA-insensitive seed germination. T-DNA insertional mutants of syd and pie1, along with amiRNA-expressing knockdown lines amiR-SYD and amiR-PIE1, confirmed that SYD, but not PIE1, is essential for promoting ABA-inhibition of seed germination. Transcriptome analyses revealed that the ABA catabolism gene CYP707A3 and the gibberellin (GA) biosynthesis genes GA20OX2, GA20OX3, GA3OX2, GA3OX3, and GA3OX4 were upregulated in amiR-SYD mutant seeds compared to control seeds under ABA treatment during germination. The ABA hyposensitivity observed in amiR-SYD mutants was reversed by treatment with uniconazole, an inhibitor of ABA catabolism and GA biosynthesis, suggesting that the reduced ABA sensitivity in amiR-SYD mutants results from increased ABA degradation and GA production. These findings demonstrate that the SYD-associated chromatin remodeling complex positively regulates ABA responses during seed germination, unlike its close homolog BRAHMA (BRM), which acts as a negative regulator of the ABA response.

Project description:In this study, Three sRNA libraries were generated and sequenced from embryos treated with GA, ABA and ddH2O (CK). A total of 247 miRNAs (69 known and 178 novel ones) were identified and found that 28 known and 70 novel miRNAs were differentially expressed in embryos with different treatments. Using bioinformatics tool, we predict target genes of the differentially expressed miRNAs. GO enrichment and KEGG pathway analysis for these targets showed that miRNAs differentially expressed in the samples are involved in peroxisome, ribosome and plant hormonal signalling. This study not only improves our knowledge about the roles of miRNAs in maize seed germination but also reveals the potential role of miRNAs in the hormonal responses of different pytohormones.

Project description:Seed germination is a major step of plant growth and development. It is critical for species competition and spreading capacity in ecosystems. In agrosystems, it eventually impacts crop growth and yield. To prevent unappropriated germination under environmental conditions that do not guarantee the establishment of a robust plantlet, seeds from temperate species are generally dormant at maturity. Dormancy is a physiological mechanism that blocks seed germination even under favorable conditions and dormancy release is therefore required prior to germination [1]. A range of environmental (e.g. temperature, light, oxygen availability) and endogenous (e.g. hormonal) signals regulate these processes and germination completion, i.e. the early emergence of embryo radicle from seed envelope, can be achieved only when promoting mechanisms overcome inhibiting processes [2]. In that sense, the balance between the two antagonistic hormones abscisic acid (ABA) and gibberellins (GA), that inhibit and stimulate seed germination, respectively, promotes either dormancy (high ABA and low GA contents) or germination (low ABA and high GA contents) [3]

Project description:Although abscisic acid (ABA) and gibberellins (GAs) play pivotal roles in many physiological processes in plants, their interaction in the control of leaf growth remains elusive. In this study, genetic analyses of ABA and GA interplay in leaf growth were performed in Arabidopsis thaliana. The results indicate that for ABA and GA interaction, leaf growth of both the aba2/ga20ox1 and aba2/GA20OX1-OE plants exhibits partially additive effects but is similar to the aba2 mutant. Consistent with this result, transcriptome analysis suggests that a substantial proportion (45-65%) of the gene expression profile of aba2/ga20ox1 and aba2/GA20OX1-OE plants overlaps and shares a similar pattern to the aba2 mutant. Thus, these data support that ABA deficiency dominates leaf growth regardless of GA levels. Moreover, gene ontology (GO) analysis indicates gene enrichment in the categories of hormone response, developmental and metabolic processes, and cell wall organization in these three genotypes. Leaf developmental genes are also involved in ABA-GA interaction. Collectively, these data support that the genetic relationship of ABA and GA interaction involves multiple coordinated pathways rather than a simple linear pathway in the regulation of leaf growth. To better understand the molecular basis of ABA and GA interaction, transcriptome analysis was performed among the genotypes used in this study.

We used the elite Chinese bread wheat cultivar Zhenmai 9023 as material and performed the first comparative transcriptome microarray analysis between embryo and endosperm response to ABA and H2O2 treatments during seed germination using the GeneChip® Wheat Genome Array Wheat seed germination includes a great amount of regulated genes which belong to many functional groups. ABA/H2O2 can repress/promote seed germination through coordinated regulating related genes expression. Project description:The callus used for total RNA extraction was derived from the scutellum of the japonica rice variety Nipponbare and cultivated in Murashige and Skoog medium* containing 10 microM 2,4 dichlorophenoxyacetic acid. Such callus maintains the ability to develop roots and leaves. After the calli had been cultured in the medium for 30 d, they were transferred to a medium containing either ABA or GA (Gibberellin) plant hormones and cultured for 3 d. The concentration of the plant hormone was adjusted to 50 microM. After culturing, we used an RNeasy Plant Mini Kit (QIAGEN, Tokyo, Japan) to extract total RNA from the hormone-treated calli and from the controls. Messenger RNA (mRNA) was isolated with an Oligotex-dt30 (Super) mRNA purification kit (TaKaRa, Shiga, Japan). Purified mRNA was amplified, labeled, and hybridized to the NIAS RICE 22K oligonucleotide array ver1 according to the manufacturer. Keywords: other

Project description:The control of seed germination and seed dormancy are critical for the successful propagation of plant species, and are important agricultural traits. Seed germination is tightly controlled by the balance of gibberellin (GA) and abscisic acid (ABA), and is influenced by environmental factors. The COP9 Signalosome (CSN) is a conserved multi-subunit protein complex that is best known as a regulator of the Cullin-RING family of ubiquitin E3 ligases (CRLs). Multiple viable mutants of the CSN showed poor germination, except for csn5b-1. Detailed analyses showed that csn1-10 has a stronger seed dormancy, while csn5a-1 mutants exhibit retarded seed germination in addition to hyperdormancy. Both csn5a-1 and csn1-10 plants show defects in the timely removal of the germination inhibitors: RGL2, a repressor of GA signaling, and ABI5, an effector of ABA responses. We provide genetic evidence to demonstrate that the germination phenotype of csn1-10 is caused by over-accumulation of RGL2, a substrate of the SCF (CRL1) ubiquitin E3 ligase, while the csn5a-1 phenotype is caused by over-accumulation of RGL2 as well as ABI5. The genetic data are consistent with the hypothesis that CSN5A regulates ABI5 by a mechanism that may not involve CSN1. Transcriptome analyses suggest that CSN1 has a more prominent role than CSN5A during seed maturation, but CSN5A plays a more important role than CSN1 during seed germination, further supporting the functional distinction of these two CSN genes. Our study delineates the molecular targets of the CSN complex in seed germination, and reveals that CSN5 has additional functions in regulating ABI5, thus the ABA signaling pathway.
